Normal development of dental innervation and nerve/tissue interactions in the colony-stimulating factor-1 deficient osteopetrotic mouse.

Dental innervation occurs concurrently with tooth development, eruption, and root formation and is suggested to interact with developing tissues. The purpose of the present study was to investigate dental innervation in osteopetrotic (op/op) mice, which carry a mutation of colony-stimulating factor-1 (CSF-1) and demonstrate sparse macrophages and osteoclasts, failure of bone resorption, lack of tooth eruption, and poor root formation. Jaw tissues from 21 mice in different age groups (7 days, 18 days, 26 days, 5 weeks, and 3 months) were prepared for immunocytochemistry and light microscopy. Im

Antisense attenuation of Wnt-1 and Wnt-3a expression in whole embryo culture reveals roles for these genes in craniofacial, spinal cord, and cardiac morphogenesis.

Wnt-1 and Wnt-3a proto-oncogenes have been implicated in the development of midbrain and hindbrain structures. Evidence for such a role has been derived from in situ hybridization studies showing Wnt-1 and -3a expression in developing cranial and spinal cord regions and from studies of mutant mice whose Wnt-1 genes have undergone targeted disruption by homologous recombination. Wnt-1 null mutants exhibit cranial defects but no spinal cord abnormalities, despite expression of the gene in these regions. The absence of spinal cord abnormalities is thought to be due to a functional compensation of

Initial stages of retinofugal axon development in the hamster: evidence for two distinct modes of growth.

In order to characterize differences in growth patterns of axons as they elongate toward their targets and during the initial stages of terminal arbor formation within the targets, we examined the primary visual system of fetal and newborn hamsters using three morphological methods: the Cajal-deCastro reduced silver method, the rapid Golgi technique, and anterograde transport of HRP. Axons emerge from the retina between the 10th and 11th embryonic days (E10-E11). The front of retinal axons crosses the chiasm, extends over the primitive dorsal nucleus of the lateral geniculate body (LGBd) by E1

CasNano: An ultracompact CRISPR-Cas12f with a GC-rich PAM for epigenome editing with therapeutic potential.

Clinical use of CRISPR-Cas proteins for genome engineering is constrained by their large size, limiting in vivo delivery. To address this, we engineered an ultracompact and versatile platform from a minimally active, 400-amino acid Cas12f protein from Blautia lenta. A multi-stage strategy of structure-guided protein mutagenesis and systematic single guide RNA (sgRNA) optimization transformed BlCas12f into a "CasNano" as a potent genome targeting tool for mammalian cells. When a catalytically dead CasNano (dCasNano) is fused to a VPR domain, it activates endogenous genes with an efficiency and

Lamin B1 affects nuclear shape and integrity through chromatin stiffness and not lamin stiffness.

The mechanical properties of the nucleus are critical for maintaining nuclear integrity and function. We previously showed that chromatin dominates short-extension mechanics whereas lamins provide long-extension strain stiffening. To distinguish the roles of lamin isoforms, micromanipulation nucleus force measurements were performed on isolated nuclei from lamin A/C (Lmna-/-) and lamin B1 (Lmnb1-/-) knockout mouse embryonic fibroblast cells. Lamin A/C knockout does not alter short-extension nuclear stiffness but is essential for strain stiffening at longer extensions. Oppositely, lamin B1 loss

Methodological Evaluation of a P2C-Based ReMOT CRISPR/Cas9 System in Aedes aegypti.

Mosquito-borne infectious diseases remain a major challenge to public health, highlighting the need for efficient and accessible gene editing approaches. Receptor-mediated ovary transduction of cargo (ReMOT) offers an alternative to embryonic microinjection, in which P2C, an ovary-targeting peptide, enables ovarian delivery of the editing components. However, key design parameters and operational boundaries of the P2C-based ReMOT system have not been clearly defined. Here, we performed a methodological evaluation of the P2C-mediated ReMOT CRISPR/Cas9 system in Aedes aegypti. Cas9-P2C fusion pr

Bacteria-herpesvirus interaction reveals a proviral role of ISGylation in promoting herpesvirus capsid assembly.

Herpesviruses are common pathogens of the oral cavity, yet how they interact with other oral microbes are poorly understood. Using murine gamma-herpesvirus 68 (MHV68) as a model for human gamma-herpesviruses, we discover that ISGylation is hijacked to facilitate viral capsid assembly and lytic replication. Co-infection with the oral Aggregatibacter actinomycetemcomitans ( A.a .) and MHV68 synergistically induced interferon-stimulated gene 15 (ISG15) expression and global ISGylation. Proteomic profiling revealed viral structural proteins as the dominant targets of ISGylation in MHV68-infected c

Self-Assembling Amphiphilic Cationic Peptide-Based Nanoarchitectures for Soft Biomaterials with Potential Antimicrobial Activity against Clinically Isolated Multidrug-Resistant Bacteria.

We herein explore the efficacy of synthesized lysine- and histidine-containing two self-assembled amphiphilic cationic peptides (SAACPs) as nanomedicine against multidrug-resistant clinically isolated bacterial strains. These SAACPs are self-assembled in water (pH 6.9) to form nanospheres with potent antibacterial activities. FEG-TEM and atomic force microscopy studies revealed the nanospherical and nanofibrillar structure. Detailed mechanistic studies probing the antibacterial activities of these self-assembled peptides substantiated that they have high efficacy to kill both Gram-positive and

Situational status and moral incoherence in UK bio-law.

UK bio-law is praised for its pragmatic governance of ethically contested practices, especially embryo research, where the post-Warnock settlement regulates conduct through licensing, oversight, and the 14-day or primitive streak rule without resolving moral-status disputes. This article argues that this pragmatism exposes a structural incoherence, partly narrowed but not resolved by abortion-law reform. Embryo research law treats the embryo as a procedurally governed object, while abortion law has historically operated through a criminal-exception model under the Offences Against the Person A

Early Detection of a Testicular Mixed Germ Cell Tumor by Alpha-Fetoprotein Surveillance After Pediatric Liver Transplantation: Case Report and Literature Review.

Liver transplant (LT) recipients, particularly pediatric survivors, face an elevated lifetime risk of de novo malignancies due to prolonged immunosuppressive therapy. Alpha-fetoprotein (AFP) is widely used as a biomarker for hepatocellular carcinoma (HCC) in post-LT surveillance, but elevations may also reflect extrahepatic tumors. Here, we report a rare case of a testicular tumor incidentally detected through AFP monitoring in a pediatric LT survivor. A 20-year-old male with situs inversus totalis underwent living donor LT for biliary atresia at 1 year of age. Nineteen years post-transplant,

Giant cervical lipoblastoma in infant: a rare case report.

INTRODUCTION AND IMPORTANCE: Lipoblastomas are benign mesenchymal tumors derived from embryonal white fat, primarily affecting children under 3 years of age. While typically located in the extremities and trunk, cervical lipoblastomas are exceedingly rare and may pose diagnostic and surgical challenges due to the proximity of vital structures. CASE PRESENTATION: We report a case of a 13-month-old male with an 8-month history of a progressively enlarging, painless mass in the right anterolateral neck. There were no signs of mass effect. Neck CT scan revealed a large fat-density lesion with mul

Long Noncoding RNA PCALRx Interacts with Pyruvate Carboxylase to Drive Multi-Organ Developmental Toxicity in Zebrafish Embryos Exposed to Amoxicillin.

Amoxicillin is widely used in human and veterinary medicine and is increasingly detected in aquatic systems, raising concerns regarding its potential adverse effects on sensitive developmental stages. Here, a zebrafish embryonic amoxicillin exposure (EAE) model was used to quantify dose- and time-dependent developmental outcomes and to elucidate underlying metabolic-epigenetic mechanisms. EAE induced dose- and time-dependent increases in mortality and malformation rates, and significantly impaired differentiation of liver, interrenal gland, heart and brain, accompanied by mitochondrial structu

The Thymus Regeneration Paradox: The Search for Stemness in an Involuting Organ.

The thymus is emerging as a model for studying organ regeneration and stem cell biology. While research has long focused on how antigen-presenting cells shape the T cell repertoire, recent discoveries unveil a far richer cellular landscape that challenges long-held views of thymus structure and function. This review traces the history of early thymic reconstitution assays, the paradigm of clonal stem cells and serial transplantation, assessing evidence for "stemness" within the thymus. A key focus is the paradox that an involuting thymus retains cells able to expand in culture and reconstitute

Proteomic Cysteine Profiling Links DMI and DMF to Antiviral Activity.

Dimethyl fumarate (DMF) and dimethyl itaconate (DMI) are electrophilic metabolite derivatives with potent immunomodulatory properties. Both have been reported to covalently modify cysteine residues in proteins, thereby altering their structure and function. Thus, we hypothesized that DMI and DMF share common cysteine targets that account for their overlapping immunosuppressive effects. Using tandem mass tag activity-based protein profiling (TMT-ABPP) in mouse embryonic fibroblasts (MEFs), we systematically mapped cysteine sites whose activity toward an iodoacetamide-desthiobiotin probe was alt

Spatiotemporal remodelling of the cloacal region determines the position of the anal opening in mouse embryos.

Conventional embryological models propose that the urorectal septum (URS) fuses with the cloacal membrane (CM) to form separate exits for the hindgut and urogenital sinus (UGS). However, previous studies have been re-evaluating this assumption, and the precise morphogenetic mechanisms determining the position of the anal opening remain unclear. To elucidate the spatial and temporal dynamics of cloacal development, we analysed mouse embryos from embryonic day (E)11.5 to E13.5 using serial midsagittal histological sections combined with three-dimensional reconstruction. We focused on the positio

HMGA2 links opposing morphogen signaling to long-term low-frequency auditory function in the cochlea.

The cochlea resolves sound frequencies through molecular, structural, and functional specializations established along its tonotopic axis. Although transient morphogen gradients establish positional information during embryogenesis, the mechanisms that maintain and refine this information as cochlear function matures remain poorly understood. Here, we identify the chromatin modifier HMGA2 as a candidate molecular link between opposing hedgehog (HH) and retinoic acid (RA) signals and long-term auditory function. In vivo manipulation of RA and HH signaling demonstrated that RA suppresses, wherea

Somatic Mosaicism of the Brain: The Functional Implications of Post-Zygotic Mutations to Neural Circuits.

The human brain is a genomic mosaic shaped by post-zygotic mutations that accumulate from the earliest embryonic divisions through aging. This review synthesizes current evidence on the origins, detection, functional consequences and disease relevance of neural somatic mosaicism. We find that somatic variants-including single-nucleotide variants, copy-number variations, mobile element insertions, structural rearrangements and aneuploidies-arise through distinct molecular mechanisms such as replication errors, oxidative stress, defective DNA repair and transposable element activity, each leavin

SOX Genes in Spinal Cord Injury: Redefining Neural Stem Cell Regeneration Strategies.

The study design is literature review. The sex-determining region Y gene (SRY)-related high mobility group box (HMG)-box (SOX) gene family has primarily been associated with neural development and sex determination and is a key component of human embryonic development. Recent studies on zebrafish models have demonstrated that the unique ability of the latter for central nervous tissue (CNS) repair following injury is largely mediated by SOX genes. Given that efforts aimed at the structural regeneration and functional restoration of neural tissue still represent a major therapeutic challenge in

Renin Cells Drive Kidney Neurovascular Development and Arterial Remodeling when Renin Activity is Deficient.

Renin cells synthesize and release the hormone-enzyme renin to regulate blood pressure and fluid-electrolyte homeostasis. Their function and identity depend on communication with surrounding cells and nerve fibers within complex kidney structure. Because renin cells are rare -0.01 % of kidney cells- conventional histological approaches cannot capture their interaction with nerve fibers and surrounding cells within the nephron and its vasculature. Using a novel ultrabright renin cell-specific tdTomato reporter mouse, high-resolution 3D imaging, and single-cell RNA-Seq, we mapped the interaction

Retinoic acid signaling and metabolism in heart failure.

Nearly 70 years after studies first showed that the offspring of vitamin A (retinol, ROL)-deficient rats exhibit structural cardiac defects and over 20 years since the role of vitamin A's potent bioactive metabolite hormone, all-trans retinoic acid (ATRA), was elucidated in embryonic cardiac development, the role of the vitamin A metabolites, or retinoids, in adult heart physiology and heart and vascular disease, remains poorly understood. Studies have shown that low serum levels of retinoic acid correlate with higher all-cause and cardiovascular mortality, though the relationship between circ
