Zinc deficiency exacerbates and supplementation attenuates early-stage obesity-related podocyte injury via Nrf2/HO-1-mediated pyroptosis.

Zinc deficiency is prevalent in obesity and chronic kidney disease; however, its therapeutic potential and underlying mechanisms in obesity-related glomerulopathy (ORG) remain to be fully elucidated. This study investigated the protective effects of zinc against obesity-induced podocyte injury and pyroptosis. Analysis of the NHANES (2011-2016) cohort, comprising 4837 adults, revealed an inverse association between BMI and serum zinc levels. Furthermore, zinc deficiency was significantly associated with reduced estimated glomerular filtration rate (eGFR) and increased urinary albumin-to-creatin

Exercise Improves Sarcopenic Obesity Through Inhibition of Ferroptosis and Activation of the AMPK/ACC Pathway.

Sarcopenic obesity, characterized by skeletal muscle loss concurrent with adipose tissue accumulation, has emerged as a global health threat. Exercise is established as an effective intervention; however, the molecular mechanisms underlying its protective effects remain incompletely defined. This study investigated whether exercise mitigates high-fat diet (HFD)-induced sarcopenic obesity, and whether the mechanism was related to the activation of the adenosine monophosphate-activated protein kinase (AMPK)/Acetyl-CoA carboxylase pathway (ACC) pathway and the inhibition of ferroptosis. Stabilization of β-catenin by 7,8-Dihydroxyflavone protects against osteoblast ferroptosis and bone loss in type 2 diabetic osteoporosis.

Type 2 diabetic osteoporosis (T2DOP) is characterized by a long-term imbalance in bone metabolism, where the diabetic environment heightens the risk of fragile fractures and reduces bone density. Emerging evidence highlights ferroptosis as a key mechanism underlying glucolipotoxicity-induced osteoblast death. 7,8-Dihydroxyflavone (7,8-DHF), a plant-derived flavonoid with potent antioxidant and anti-ferroptotic properties, has not been thoroughly investigated in the context of T2DOP. First acknowledged in 1984 as a key type of secondary osteoporosis, T2DOP is distinguished by notable cortical p

The deubiquitinase YOD1 in renal tubular epithelial cells promotes diabetic kidney disease by stabilizing KEAP1.

Renal tubular epithelial cell (RTEC) injury is an early event in diabetic kidney disease (DKD) and is a key driver of its development and progression. Deubiquitinating enzymes (DUBs) play crucial roles in the progression of diseases by regulating the stability of substrate proteins. This study aims to elucidate the regulatory role and underlying mechanisms of the deubiquitinating enzyme ovarian tumor domain-containing 2 (YOD1) in RTEC injury and DKD. Through transcriptomics and single-cell mRNA sequencing, we confirmed that YOD1 expression is upregulated in diabetic kidneys and that YOD1 is pr

Cardiomyocyte BMAL1 deficiency worsens obesity-related cardiomyopathy with heightened PINK1/Parkin-linked mitophagy and mitochondrial dysfunction.

Obesity-related cardiomyopathy (OCM) is characterized by pathological cardiac remodeling and progressive functional decline, often accompanied by mitochondrial dysfunction, particularly aberrant mitophagy. The role of the core circadian gene brain and muscle ARNT-like protein 1 ( Bmal1) in OCM remains unclear. In this study, we employed a high-fat diet (HFD)-induced OCM mouse model, a cardiomyocyte-specific Bmal1 knockout ( Bmal1 CMKO) model, and a palmitic acid (PA)-induced H9c2 cardiomyocyte injury model to investigate the function of Bmal1. In vivo, BMAL1 expression was reduced in hearts of

Schisandrin B inhibits hepatic inflammation and steatosis via activation of pregnane X receptor signaling in methionine and choline-deficient high fat diet-induced steatohepatitis mouse model.

Schisandrin B (Sch B), a key bioactive lignan extracted from Schisandra chinensis (Turcz.) Baill, exhibits lipid lowering and anti-inflammatory properties in vivo and in vitro. However, few studies investigate the anti-metabolic dysfunction associated steatohepatitis (MASH) effect of Sch B. In this study, we employed a methionine and choline-deficient high fat diet (MCDHF)-induced MASH mouse model and palmitate acid (PA)-induced MASH cell models to evaluate the anti-inflammatory and anti-steatosis effects of Sch B. We also explore the molecular targets of Sch B for its anti-MASH effect. The re

Linarin alleviates high-fat diet-induced hepatic steatosis by inhibiting PDE4D and activating the cAMP/PKA/CREB pathway.

Metabolic-associated fatty liver disease (MAFLD), a major consequence of obesity and metabolic dysfunction, lacks effective treatments. The cAMP/PKA signaling pathway regulates lipid metabolism, and inhibition of its upstream target PDE4D alleviates MAFLD progression. Linarin, a natural flavonoid glycoside with hepatoprotective properties, remains underexplored for MAFLD mechanisms. To investigate linarin's therapeutic effects on high-fat diet (HFD)-induced MAFLD and elucidate its mechanisms, focusing on PDE4D inhibition and cAMP/PKA/CREB pathway activation. Sirtuin 6 activator UBCS039 ameliorates hepatic lipogenesis through liver X receptor deacetylation.

Histone deacetylase sirtuin 6 (SIRT6) regulates hepatic lipogenesis by modulating key transcription factors, such as liver X receptor (LXR) and sterol regulatory element binding transcription factor 1 (SREBF1). Despite its potential role in the regulation of hepatic lipogenesis, SIRT6 activators that inhibit hepatic lipogenesis and ameliorate steatotic liver diseases have not been developed yet. We investigated the effects of UBCS039, a selective SIRT6 activator, on lipogenic gene expression and inflammation in hepatocytes. Analysis of publicly available RNA sequencing datasets revealed signif
