Evolution of Gastrointestinal Inflammatory Diseases and Neoplasm Burden in Super-Elderly Populations: Integrated GBD 2023, CHARLS, and CLHLS Analyses of China and G20 Countries.

BACKGROUND: Rapid population aging has profoundly altered the epidemiological profile of gastrointestinal diseases in individuals aged ≥75 years. Although enteric infections (EI) have been effectively controlled through public health interventions, the burden of gastrointestinal inflammatory diseases (GIID) and neoplasms (GIN) continues to evolve. Systematic comparisons of temporal trends, sex differences, and age-subgroup patterns between China and G20 nations remain limited. METHODS: Using GBD 2023 data, we analyzed incident cases, age-standardized rates (ASR), and Disability-Adjusted Life

Machine learning-driven identification of critical gene programs and key transcription factors in migraine.

BACKGROUND: Migraine is a complex neurological disorder characterized by recurrent episodes of severe headaches. Although genetic factors have been implicated, the precise molecular mechanisms, particularly gene expression patterns in migraine-associated brain regions, remain unclear. This study applies machine learning techniques to explore region-specific gene expression profiles and identify critical gene programs and transcription factors linked to migraine pathogenesis. METHODS: We utilized single-nucleus RNA sequencing (snRNA-seq) data from 43 brain regions, along with genome-wide assoc

Co-occurrence patterns and modifiable risk factors of intracerebral hemorrhage and hypertensive heart disease in adults aged 35 and older: a cross-sectional spatial analysis.

BACKGROUND: Intracerebral hemorrhage (ICH) and hypertensive heart disease (HHD) are major contributors to global mortality and disability. While both conditions share common risk profiles, most existing studies have examined them in isolation, limiting understanding of their co-occurrence patterns and shared determinants. This study aimed to systematically assess the global co-burden of ICH and HHD in adults aged ≥35 years, identify their shared and distinct risk factors, and develop an integrated risk index to inform comprehensive prevention strategies. METHODS: Data were obtained from the G

An integrative computational approach for identification of NLRP3 inhibitors through machine learning, docking, dynamics and DFT analysis.

Neuroinflammation, mediated by NLR family pyrin domain containing 3 (NLRP3) inflammasome, plays a crucial role in the development of many central nervous system (CNS) diseases such as Alzheimer's disease, Parkinson's disease, multiple sclerosis, and stroke. Despite extensive efforts, there are no clinically approved NLRP3 inhibitors due to issues like poor selectivity, undesirable drug-like properties, and safety concerns. In this study, a machine learning-based virtual screening strategy was used to identify phytochemicals that inhibit the NLRP3 NACHT domain, a key region involved in ATP-driv

Machine Learning Approaches to Investigate the Structure-Activity Relationship of Angiotensin-Converting Enzyme Inhibitors.

Angiotensin-converting enzyme inhibitors (ACEIs) play a crucial role in treating conditions such as hypertension, heart failure, and kidney diseases. Nevertheless, the ACEIs currently available on the market are linked to a variety of adverse effects including renal insufficiency, which restricts their usage. There is thus an urgent need to optimize the currently available ACEIs. This study represents a structure-activity relationship investigation of ACEIs, employing machine learning to analyze data sets sourced from the ChEMBL database. Exploratory data analysis was performed to visualize th

Automatic discrimination of actinic keratoses from clinical photographs.

BACKGROUND AND OBJECTIVE: Actinic keratoses (AK) are common premalignant skin lesions that can progress to invasive skin squamous cell carcinoma (sSCC). The subtle accumulation of multiple AK in aging individuals increases the risk of sSCC development, and this underscores the need for efficient treatment and patient follow-up. Our objectives were to develop a method based on color texture analysis of standard clinical photographs for the discrimination of AK from healthy skin and subsequently to test the developed approach in the quantification of field-directed treatment interventions. Graft arteriosclerosis and glomerulosclerosis correlate with flow and resistance to machine perfusion in kidney transplantation.

INTRODUCTION: Machine perfusion to preserve kidneys for transplantation has grown over the past decade with demonstrated diagnostic and therapeutic benefits. Flow and resistance patterns are used to predict delayed graft function (DGF) and posttransplant graft survival. Preimplantation biopsies obtained serve a similar role in evaluating kidneys especially if they meet expanded criteria. The reliability of available data is greater if there is a correlation among various forms of assessment. In this study we attempted to study serial pump parameters that might correlate with abnormal findings

Concurrent Associations of Gaming Time and Problematic Gaming with Depressive Symptoms in Children and Adolescents: A Three-Wave Cohort Study with Complementary Machine Learning Classification.

Background/Objectives: Digital media use is integral to the daily lives of children and adolescents, intensifying concerns regarding potential associations with mental health. However, much of the literature linking gaming and depression is based on cross-sectional designs and does not adequately distinguish within-person change from between-person differences. This study examined concurrent associations between gaming use patterns, problematic gaming behavior, and depressive symptoms across three repeated assessment waves and, as a complementary analysis, evaluated the concurrent classificati

Evaluating Laryngeal Stability in Drug-Induced Parkinsonism Using Hilbert-Huang Transform and Chaos Dynamics: A Multi-Vowel Nonlinear Acoustic Framework for Differential Diagnosis.

Background/Objectives: Drug-induced parkinsonism (DIP) is a common extrapyramidal adverse effect of dopamine-receptor blocking agents, yet interpretable biomarkers of its acoustics remain limited. This study investigated whether nonlinear dynamics could provide physiologically interpretable markers of DIP and clarify how these abnormalities vary across vowels and extrapyramidal symptoms. Methods: Sustained phonations of five vowels (/a/, /i/, /u/, /e/, /o/) were analyzed in 82 patients with DIP and 30 healthy controls. Hilbert-Huang Transform (HHT) features were integrated along with Sample En

Developing a Comprehensive Model for Explaining Treatment Adherence Behaviour: Evidence From the International Beamer Consortium.

INTRODUCTION: Optimizing treatment adherence behaviour has become a focus in healthcare, as improved adherence enhances clinical outcomes, patient experiences and health system sustainability. For this optimization endeavour scholars have posited diverse variables divided into two clusters. The first variable cluster emphasizes treatment priority, treatment concerns and health consciousness. The second variable cluster highlights acceptance, control and subjective health experience. Although some of these variables have been tested in isolation, few comprehensive models have been established.

Sex-Specific Digital Phenotyping of Lower Urinary Tract Symptoms: A Cross-Sectional Study of 1586 Adults From 35 Centers in China.

BACKGROUND AND PURPOSE: Lower urinary tract symptoms (LUTS) represent a complex clinical syndrome with significant sexual dimorphism. Traditional "one-size-fits-all" management often overlooks intra-sex phenotypic heterogeneity. This study utilized unsupervised machine learning to identify sex-specific digital phenotypes of LUTS within a large-scale clinical cohort to provide a foundation for precision urological care. MATERIALS AND METHODS: We analyzed a multicenter database of 1586 treatment-seeking adults (1098 females, 488 males) across 35 centers. Baseline clinical characteristics and LU

From heterogeneity to mechanism: a systems-level analysis pinpoints the S100A8/A9 axis in colorectal cancer.

BACKGROUND: The functional roles of the S100 protein family in colorectal cancer (CRC) are enigmatic, marked by widespread dysregulation but limited prognostic utility. This heterogeneity obscures their potential as therapeutic targets. METHODS: We employed a multi-omics strategy to deconstruct this complexity, integrating bulk transcriptomics (TCGA), large-scale single-cell RNA-seq atlases, and machine learning-based prioritization. Cell-type-specific expression patterns were contextualized using the spatial annotation of cell origins available within the Pan-GI Cell Atlas. Key findings were

Formulaic and novel language dissociate in Alzheimer's clinical syndrome: evidence for the Dual-Process Model.

INTRODUCTION: Spontaneous speech is commonly disrupted in persons with Alzheimer's disease (AD) and/or Alzheimer's clinical syndrome (ACS). Importantly, different aspects of speech (e.g. formulaic versus more novel or flexible speech) place different demands on distinct cognitive systems. Formulaic language may rely on automatized procedural processes, while more novel or diverse speech requires more flexible lexical-semantic processes associated with the subsystems of declarative memory. Given that AD/ACS are associated with impaired declarative processes and relatively spared procedural proc

Using machine learning to estimate the riverine fluxes of per- and polyfluoroalkyl substances to estuaries in 2000-2022, China.

Per- and polyfluoroalkyl substances are persistent contaminants of global concern. China released an estimated 250 t of perfluorooctanoic acid (PFOA) and its salts during 2004-2012, but riverine transport to coastal waters remains poorly quantified. We compiled 4978 concentration records from 136 publications covering more than 100 rivers nationwide. The analysis based on the database revealed that PFOA dominated the PFAS profiles (median: 12.80 ng/L), followed by perfluorobutanoic acid (PFBA, 4.00 ng/L), while the remaining compounds were substantially lower (0.34-2.27 ng/L). Moreover, substa
