Different mechanisms for human rhinovirus survival in the presence of deleterious amino acid substitutions at virion protein-protein or RNA-protein interfaces.

A combination of structure-function and genetic analyses of viruses is unveiling a wide repertoire of adaptive mechanisms for restoring biological function in the face of negative selection pressures. The fixation of second-site compensatory mutations in highly heterogeneous RNA virus populations constitutes a frequent and versatile response to recover infectivity in the presence of deleterious mutations. In this study, we explored the following: (i) survival mechanisms of human rhinovirus (RV) when infectivity-impairing amino acid substitutions were introduced at either capsid protein-protein

African swine fever virus pC147L inhibits retinoic acid-inducible gene I-like receptors pathway via targeting MAVS-TRAF6 complex.

African swine fever virus (ASFV) suppresses the host's innate immunity by interfering with pattern recognition receptors (PRRs) signaling pathways; however, the molecular mechanisms by which it inhibits PRR signaling remain poorly characterized. This study aimed to identify the ASFV-encoded protein pC147L. The pC147L, a subunit of the viral RNA polymerase, acts as a potent inhibitor of the Retinoic Acid-inducible Gene I (RIG-I)-like receptors (RLRs) pathway, which mediates the detection of viral double-stranded RNA. We used quantitative reverse transcription-polymerase chain reaction (qRT-PCR)

Development of a Ready-To-Use Bioluminescence Immunosensor for the One-Step Sensitive Detection of Antibodies Against African Swine Fever Virus.

Rapid and reliable analytical techniques play important roles in various research fields and are particularly crucial for diagnosing infectious diseases in clinical settings. African swine fever (ASF) is a devastating viral pig disease for which no effective vaccine is available. The ongoing ASF pandemic has highlighted the importance of rapid and accurate diagnosis, which enables the timely implementation of control and eradication measures. In this study, a ready-to-use bioluminescence immunosensor based on a split-nanoluciferase (NanoLuc) reporter system was proposed for the one-step sensit

A novel glutathione peroxidase (EnGPX) from Eimeria necatrix contributes to oocyst wall biogenesis and confers protective immunity in chickens.

Coccidiosis, caused by Eimeria spp., represents a major threat to poultry health and global food security. The oocyst wall, a crucial structure for parasite survival and transmission, is assembled through redox-dependent protein cross-linking. While glutathione peroxidases (GPXs)-mediated redox reactions play a role in oocyst wall formation, studies on GPX in Eimeria necatrix are limited. In this study, we identified and characterized a novel glutathione peroxidase (EnGPX) from E. necatrix. Bioinformatic analysis showed that EnGPX belongs to the Cys-dependent GPX (Cys-GPX) and contains a nonca

[Biosynthesis of Salmonella enteritidis O antigen-based glycoproteins].

Salmonella enteritidis (SE) has been recognized as an important zoonotic pathogen, and the prevention and control of salmonellosis has long been a conundrum. However, glycoconjugate vaccines seem to be a promising solution. Glycoproteins are conventionally synthesized by chemical cross-linking which features complex procedure and cost-intensiveness. Therefore, a stable biosynthesis method at lower cost is in urgent need. For the biosynthesis of SE O-antigen-based glycoproteins, we used CRISPR/Cas9 to develop the waaL-deleted SE strain ∆waaL. The synthesis of lipopolysaccharide (LPS) was detect

Immunogenicity and protective efficacy of a novel foot-and-mouth disease virus empty-capsid-like particle with improved acid stability.

Foot-and-mouth disease virus (FMDV) is the etiological agent of a highly contagious disease that affects cloven-hoofed animal species. The FMDV capsid is highly acid labile and viral particles lose their immunogenicity when they disassemble at mildly acidic pHs. The viral capsid of FMDV serotype O is more sensitive than those of other serotypes, making it more difficult to acquire enough empty-capsid-like particles in the acidic insect cell environment for research. In this study, novel FMDV mutants with increased acid resistance were isolated using BHK-21 cell cultured under low-pH conditions

Phenotypic and functional characterization of human memory T cell responses to Burkholderia pseudomallei.

BACKGROUND: Infection with the Gram-negative bacterium Burkholderia pseudomallei is an important cause of community-acquired lethal sepsis in endemic regions in southeast Asia and northern Australia and is increasingly reported in other tropical areas. In animal models, production of interferon-gamma (IFN-gamma) is critical for resistance, but in humans the characteristics of IFN-gamma production and the bacterial antigens that are recognized by the cell-mediated immune response have not been defined. METHODS: Peripheral blood from 133 healthy individuals who lived in the endemic area and had

Identification of a defined linear epitope in the OspA protein of the Lyme disease spirochetes that elicits bactericidal antibody responses: Implications for vaccine development.

The lipoprotein OspA is produced by the Lyme disease spirochetes primarily in unfed ticks. OspA production is down-regulated by the blood meal and it is not produced in mammals except for possible transient production during late stage infection in patients with Lyme arthritis. Vaccination with OspA elicits antibody (Ab) that can target spirochetes in the tick midgut during feeding and inhibit transmission to mammals. OspA was the primary component of the human LYMErix™ vaccine. LYMErix™ was available from 1998 to 2002 but then pulled from the market due to declining sales as a result of unsub

Coupling of the Hla-IsdB fusion antigen to mi3 nanoparticles enhances dendritic cell activation and protects against Staphylococcus aureus skin infection.

Staphylococcus aureus remains a leading cause of skin and soft-tissue infections, and antibiotic resistance undermines treatment. Vaccination is a key strategy that has the potential to address antibiotic resistance. However, subunit vaccines require multiple doses and have a slower response. In this study, we engineered a nanoparticle vaccine that multivalently displays a fusion antigen (HI) composed of the non-hemolytic HlaH35L variant and the N2 domain of IsdB on the self-assembling mi3 scaffold. We systematically characterized the physicochemical properties of HI-mi3 and evaluated its immu

Selection of immunodominant fragments from envelope gene for vaccine against Japanese encephalitis virus in DNA priming-protein boosting protocols.

Fragmentation of E gene of JEV into smaller fragments, none of the fragments either in plasmids form or in recombinant protein form can induce optimal protection against the virus infection. It is only when DNA priming-protein boosting strategies are used then the N-terminal E(A) and the C-terminal E(B) showed full protection against JEV as those induced by commercial vaccine, provided both fragments are preceded in the N-terminal by a signal peptide M(15) derived from C-terminal of prM gene in JEV genome. When the subfragments of E(A): E(A1) and E(A2) and E(B): E(B1) and E(B2) are tested, onl

Exploring the out of sight antigens of SARS-CoV-2 to design a candidate multi-epitope vaccine by utilizing immunoinformatics approaches.

SARS-CoV-2 causes a severe respiratory disease called COVID-19. Currently, global health is facing its devastating outbreak. However, there is no vaccine available against this virus up to now. In this study, a novel multi-epitope vaccine against SARS-CoV-2 was designed to provoke both innate and adaptive immune responses. The immunodominant regions of six non-structural proteins (nsp7, nsp8, nsp9, nsp10, nsp12 and nsp14) of SARS-CoV-2 were selected by multiple immunoinformatic tools to provoke T cell immune response. Also, immunodominant fragment of the functional region of SARS-CoV-2 spike (

Comparison of heat shock response in Brucella abortus and Brucella melitensis.

Heat shock protein (hsp) is highly conserved, that serves a wide range of function in protein folding and transport. It protect from various type of stress including heat shocks. However, it is well known that the virulence of B. melitensis is more than B. abortus, but there is not any strong evidence to verify it. For this purpose, in refer to potent antigenicity of hsps in various infectious as well as some hsp molecules act as potent activator of macrophage (danger signal), we hypothesized that difference in virulence between B. abortus and B. melitensis may be originated from difference in

Synthetic peptides identify promiscuous human Th1 cell epitopes of the secreted mycobacterial antigen MPB70.

MPB70 is a secreted protein of Mycobacterium bovis and Mycobacterium tuberculosis which stimulates both cellular and humoral immune responses during infection with bovine and human tubercle bacilli. In addition, vaccination with MPB70 has been shown to induce Th1 cell responses and protection in animal models of tuberculosis. The present study was carried out to map the dominant human Th1 cell epitopes of MPB70 in relation to major histocompatibility complex (MHC) class II restriction in healthy subjects showing strong T-cell responses to complex mycobacterial antigens. Presentation of the protective epitope in a Chlamydia muridarum MOMP vaccine is dependent on γ-interferon-inducible lysosomal thiol reductase (GILT).

The Chlamydia outer membrane complex has approximately 20 cysteine-rich proteins, including the major outer membrane protein (MOMP) with 8 cysteines. The enzyme γ-interferon-inducible lysosomal thiol reductase (GILT) is expressed constitutively in lysosomes of antigen presenting cells and reproductive tract epithelial cells. GILT catalyzes disulfide bond reduction during protein degradation that generates T cell epitopes. To evaluate the role that GILT plays in chlamydial infections, C57BL/6 WT mice and mice deficient in Ifi30 (GILT-KO) were challenged vaginally with Chlamydia muridarum (Cm).

ESAT-6-derived lipopeptide-based mucosal vaccine elicits distinct humoral immune responses concomitant with a reduction in mycobacterium infection.

Respiratory mycobacterial infections remain a major global health threat, yet current vaccine strategies, which focus primarily on cell-mediated immunity, have met with limited success. We previously demonstrated that a vaccine composed of five lipopeptides (LPs) encoding immunodominant ESAT-6 epitopes elicits protective cellular immune responses against Mycobacterium tuberculosis infection. In this study, we enhanced the ESAT-LPs vaccine with a novel mucosal immune modulator, heat-killed Caulobacter crescentus (HKCC), and examined both cellular and humoral immunity in a mouse model (BALB/c) o

Intramuscular administration of influenza hemagglutinin in one hind leg elicits similar protective immune responses as administration in two hind legs.

The objective of this study was to determine if distributing vaccine antigen across two intramuscular vaccination sites, rather than a single site could elicit more efficient protective immune responses. Most commercial injectable vaccines are administered unilaterally in a single location. The hypothesis of this study was vaccination in two distinct intramuscular sites would produce a more robust immune response compared to a single injection site. Mice vaccinated with a bilateral vaccination strategy may elicit broader antigen dissemination leading to increased germinal center activation and

Computational identification of antigens and development of a multi-epitope vaccine for bacterial vaginosis caused by Gardnerella vaginalis.

Bacterial vaginosis (BV), a prevalent form of vaginal dysbiosis, is primarily caused by Gardnerella vaginalis. The increasing prevalence of BV has raised global health concerns, prompting the need for a vaccine that can enhance human immunity and mitigate BV transmission. The study developed a multi-epitope vaccine for BV infection using immunoinformatic methodologies. B-cell and T-cell epitopes were identified using the IEDB recommended server and NetCTL. Specifically, HTL epitopes were predicted against HLA-DR alleles (including DRB101:01, DRB103:05, and DRB104:04), while CTL epitopes were p

Peptide fibrils as a vaccine: Proof of concept.

BACKGROUND: Rapid vaccine platforms development is crucial for responding to epidemics and pandemics of emerging infectious diseases, such as Ebola. This study explores the potential of peptide vaccines that self-organize into amyloid-like fibrils, aiming to enhance immunogenicity while considering safety and cross-reactivity. METHODS: We synthesized two peptides, G33 and G31, corresponding to a segment of the Ebola virus GP2 protein, with G33 known to form amyloid-like fibrils. Their toxicity was assessed in vitro using an MTT assay on MDCK and A549 cell cultures. For in vivo studies, balb/c

Rift Valley fever virus subunit vaccines confer complete protection against a lethal virus challenge.

Rift Valley fever virus (RVFV) is an emerging mosquito-borne virus causing significant morbidity and mortality in livestock and humans. Rift Valley fever is endemic in Africa, but also outside this continent outbreaks have been reported. Here we report the evaluation of two vaccine candidates based on the viral Gn and Gc envelope glycoproteins, both produced in a Drosophila insect cell expression system. Virus-like particles (VLPs) were generated by merely expressing the Gn and Gc glycoproteins. In addition, a soluble form of the Gn ectodomain was expressed and affinity-purified from the insec
