Genetic distances among the Ho tribe and other groups of Central Indians.

The Ho, a settled tribal group of Chota Nagpur, India, were tested for five genetic characters. Genetic distance among eleven tribal groups of Bihar, Orissa and Madhya Pradesh are calculated according to Edwards ('71). Affinities of these tribes are discussed taking into consideration the languages spoken by them. These breeding groups may have drifted apart along the paths of their dialect differentiations. Cultural and geographical factors further enhanced their isolation.

Invasion and growth of Plasmodium falciparum in different types of human erythrocyte.

The susceptibility of human red blood cells to invasion by Plasmodium falciparum was investigated in microtissue cultures with different pop … ignificantly retarded. This might confer a selective advantage on individuals with thalassaemia and sickle cell trait, in which HbF levels are raised in early life.

Cerebral hemodynamic changes after haploidentical hematopoietic stem cell transplant in adults with sickle cell disease.

Preliminary evidence from a series of 4 adults with sickle cell disease (SCD) suggests that hematopoietic stem cell transplant (HSCT) improves cerebral hemodynamics. HSCT largely normalizes cerebral hemodynamics in children with SCD. We tested the hypothesis in adults with SCD that cerebral blood flow (CBF), oxygen extraction fraction (OEF), and cerebral metabolic rate of oxygen (CMRO2) measured using magnetic resonance imaging, normalized to healthy values, comparing measurements from ∼1 month before to 12 to 24 months after HSCT (n = 11; age, 33.3 ± 8.9 years; 389 ± 150 days after HSCT) with

[Human globin genes: what can we learn from their polymorphism?].

Molecular analysis of human alpha and beta globin genes reveals extensive polymorphism at these loci. Worldwide distribution of the sickle cell trait has been well known for some time. However, the molecular basis and distribution of thalassemia have been more recently studied. These are the commonest monogenic disorders. For most of them, beta-thalassemia is due to single nucleotide substitutions, small deletions or insertions. They are very heterogeneous and widely dispersed in the Old World. alpha-thalassemia is mainly due to the deletion of one to four alpha genes. On the whole, their dist…

Site-specific base changes in the coding or promoter region of the human beta- and gamma-globin genes by single-stranded oligonucleotides.

SSOs (single-stranded oligonucleotides) can mediate site-specific alteration of base-pairs in episomal and chromosomal target genes in mammalian cells. The TNE (targeted nucleotide exchange) can result in either repair or mutation of a gene sequence and is mediated through endogenous DNA repair pathway(s). Thus the approach provides a technique for the treatment of monogenic disorders associated with specific point mutations such as SCD (sickle cell disease). We studied the potential application of SSOs for SCD by introducing either an A to T substitution at the sixth codon of the human beta-g

Clinical and hematological responses to hydroxyurea in Sicilian patients with Hb S/beta-thalassemia.

Although, several reports have detailed that hydroxyurea can ameliorate the clinical course of adult and pediatric patients with sickle cell anemia (Hb S or beta(S)), few clinical studies have been carried out in patients with beta(S)/beta-thalassemia. In a two-year clinical study, we evaluated the efficacy of hydroxyurea in a group of 22 adult Sicilian patients with beta(S)/beta-thalassemia with severe phenotypes. Among the 20 patients evaluated during 2 years of treatment, we observed a very good clinical response with a 93% reduction of the annual number of crises (median 7 versus 0.5 crise

Circulating erythropoietin concentration associates with thromboembolism in sickle cell disease.

Elevated erythropoietin (EPO) concentration associates with thrombotic risk in hypoxic conditions, hereditary erythrocytosis and treatment of anaemia with recombinant EPO. We evaluated sickle cell disease (SCD) patients from the University of Illinois at Chicago (UIC) and the Treatment of Pulmonary Hypertension and SCD with Sildenafil Therapy (Walk-PHaSST) study and found that higher serum EPO concentration associated with increased thromboembolic risk (combined odds ratio [OR] = 1.9, p = 0.0029, N = 557). Percent haemoglobin F and haemoglobin concentration strongly correlated with EPO concent

Gallbladder disease in young Congolese with sickle cell anemia: an ultrasound survey.

Patients with sickle cell anemia have been shown to present with a high prevalence of gallbladder disease. Relatively few data exist on the prevalence of gallbladder disease in Africans with autosomal recessive gene for sickle hemoglobin. To establish this relationship, we examined the 1-year incidence of gallbladder disease among 190 consecutive children and young adults with sickle cell anemia aged 3-24 years using ultrasound. There were 130 males (68.4 per cent) and 60 females (31.6 per cent). Incidence of cholelithiasis, cholecystitis, hydrops of gallbladder, and biliary sludge were 58.4,

The M gamma chain of human fetal hemoglobin is an A gamma chain with an in vitro modification of gamma 141 leucine to hydroxyleucine.

We have reanalyzed the structure of the gamma T-15 peptide from the minor M gamma chain of human hemoglobin (Hb) F. Amino acid analysis confirmed that the Leu 141 residue was missing from position 9 of this peptide, and liquid secondary ion mass spectrometry indicated that it was replaced, not by methionine (residue mass 131) as previously believed, but by an amino acid of mass 129. By analogy with the recently reported oxidation of the corresponding leucine at position gamma 141 of the unstable Hb Atlanta, it appears that the M gamma chain also results from the oxidation of gamma 141 to hydro

Genetic disorders and congenital anomalies in Nigeria: a scoping review.

Nigeria, with its rich cultural and genetic diversity, faces a largely undocumented burden of genetic disorders and congenital anomalies. Un … ported at least one genetic disorder. Hemoglobinopathies–particularly sickle cell disease (SCD) and sickle cell traits–were the most frequently reported conditions, with prevalence estimates as high as 61%, underscoring Nigeria’s substantial disease burden. Glucose-6-phosphate dehydrogenase (G6PD) deficiency was also commonly reported, with prevalence reaching up t …

Prophylactic vs therapeutic blood transfusions impact on pregnant sickle cell patients.

OBJECTIVE: To observe the fetomaternal outcome of therapeutic versus prophylactic blood transfusions in patients with sickle cell disease (SCD) during pregnancy. METHOD: This single-center retrospective observational study was conducted on consecutive pregnant women with SCD between January 2018 and December 2020. All the pregnant women with SCD were included in this study. Sickler women carrying sickle cell traits (HbAS, HbAC), multiple pregnancy, or other medical diseases were excluded from the study. We used non-random, convenience sampling and collected their demographic details, medical …

A cost-effectiveness evaluation of newborn hemoglobinopathy screening from the perspective of state health care systems.

OBJECTIVE: To determine the most cost-effective strategy for newborn hemoglobinopathy screening from the perspective of state health care systems. STUDY DESIGN: Using Alaska as an example, we used decision analysis to compare a policy of no screening to universal or targeted screening with selective follow-up only of infants who are homozygous or compound heterozygous for an abnormal hemoglobin variant and to universal or targeted screening with complete follow-up, including follow-up of infants with clinically insignificant traits. Probabilities and costs were varied over values that might b

Newborn screening results for sickle cell disease from the ASH Consortium on Newborn Screening in Africa (CONSA).

The Consortium on Newborn Screening in Africa (CONSA), launched by the American Society of Hematology in 2020, is designed to initiate and expand sustainable newborn screening (NBS) programs for sickle cell disease (SCD) across sub-Saharan Africa. This multiyear pilot program includes 11 clinical sites in 7 countries, namely Ghana, Kenya, Liberia, Nigeria, Tanzania, Uganda, and Zambia. After extensive training of laboratory and clinical personnel, dried blood spots were collected from newborns and tested by isoelectric focusing at central laboratories within each country. Positive samples were

Beta thalassaemia in Nigeria: myth or fact?

BACKGROUND: The high prevalence of sickle cell disorders and the mild deletional alpha-thalassaemia among Nigerians is well known, but beta-thalassaemia is believed to be almost nonexistent. Beta thalassaemia trait (BTT) was screened for in patients with unexplained recurrent haemolytic anaemia and healthy individuals. METHODS: beta-thalassaemia trait (BTT) was screened for using MCH and HbA2 of 27 pg and 3.5% respectively as cut off in 151 Nigerians which included 29 patients and 122 apparently healthy individuals. The subjects were categorized into four, Group I (high HbA2, low MCH), Group
