Unusual late responses in a patient with an Arnold-Chiari malformation.

We studied a young man with spastic right hemiparesis, in whom supramaximal stimulation of the left posterior tibial nerve produced toe movements of the both feet and associated late responses in the flexor hallucis brevis muscle bilaterally. These findings indicate that, in this patient, there are central connections between peripheral afferents and contralateral alpha-motor neurons. It may be that such connections are normally present but that they are too weak in normal subjects to produce firing of the alpha-motor neurons by themselves. If so, the loss of cortical inhibition in our patient

Presynaptic inhibition of primary olfactory afferents mediated by different mechanisms in lobster and turtle.

Presynaptic regulation of transmission at the first olfactory synapse was investigated by selectively imaging axon terminals of receptor neurons in the lobster olfactory lobe and turtle olfactory bulb. In both species, action potential propagation into axon terminals after olfactory nerve stimulation was measured using voltage-sensitive dyes. In addition, in the turtle, calcium influx into terminals was measured by selectively labeling receptor neurons with dextran-conjugated calcium indicator dyes. In the lobster, application of the inhibitory transmitters GABA or histamine suppressed action

Neuromodulatory inputs maintain expression of a lobster motor pattern-generating network in a modulation-dependent state: evidence from long-term decentralization in vitro.

Neuromodulatory inputs play a critical role in governing the expression of rhythmic motor output by the pyloric network in the crustacean stomatogastric ganglion (STG). When these inputs are removed by cutting the primarily afferent stomatogastric nerve (stn) to the STG, pyloric neurons rapidly lose their ability to burst spontaneously, and the network falls silent. By using extracellular motor nerve recordings from long-term organotypic preparations of the stomatogastric nervous system of the lobster Jasus lalandii, we are investigating whether modulatory inputs exert long-term regulatory inf

Activation of P2X receptors for adenosine triphosphate evokes cardiorespiratory reflexes in anaesthetized rats.

1. We tested the hypothesis that activation of P2X receptors associated with vagal afferent nerves can evoke a Bezold-Jarisch (B-J) depressor reflex in anaesthetized rats. 2. Injection of alphabeta-methylene ATP (alphabeta-MeATP; 0.6-600 nmol i.v.) evoked a dose-dependent B-J reflex comprising bradycardia, hypotension and apnoea in rats anaesthetized with pentobarbitone. Apnoea was commonly preceded by hyperventilation. Bilateral vagotomy significantly reduced the bradycardia and most of the apnoeic response without affecting hyperventilation, and unmasked a vasopressor response. Hypotension a

Ice-water test in the urodynamic evaluation of spinal cord injured patients.

The purpose of this study was to determine the clinical utility of the ice-water test (IWT) during urodynamic evaluation in spinal cord injured (SCI) patients. Thirty-two suprasacral SCI patients with documented detrusor hyperreflexia (DH) underwent urodynamic study and IWT. Fifteen patients had repeated testing 2 weeks later. The IWT was performed with the patient in the supine position; 100 mL of sterile saline water at 4-8 degrees C was injected manually. If cystometric capacity was <200 mL, a volume of about 50% of the individual cystometric bladder capacity was used. The cold fluid was le

Acetazolamide reduces peripheral afferent transmission in humans.

Carbonic anhydrase has been localized in skeletal muscle and nerve, thus, inhibition with acetazolamide (ACZ) may alter nerve and/or muscle function in healthy humans. ACZ (3 oral doses 14, 8, and 2 h prior to testing) reduced isometric force (37%) and peak to peak electromyographic (EMG) amplitude (1.38 mV to 0.83 mV), while increasing EMG latency associated with a unilateral Achilles tendon-tap. Reflex recovery profiles, following a contralateral conditioning tap, were similar in both placebo and ACZ experiments. ACZ led to significant changes in Hmax/Mmax ratio (52.19/14.42 to 45.73/15.65)

Mastication-related neurons in the orofacial first somatosensory cortex of awake cats.

In the orofacial area of the first somatosensory cortex (SI), we recorded single unit activity from 699 neurons in 11 awake cats. Fifty-two percent (362/699) were mastication-related neurons (MRNs) showing activity related to some aspects of masticatory movements. MRNs were divided into three types by their activity patterns: (1) the rhythmical type, showing rhythmical bursts in pace with the masticatory rhythm; (2) the sustained type, showing a sustained firing during the period of taking food and (3) the transient (biting) type, showing intense discharges in coincidence with biting hard food

Selective phonotaxis to advertisement calls in the grey treefrog Hyla versicolor: behavioral experiments and neurophysiological correlates.

1. The significance of particular acoustic properties of advertisement calls for selective phonotaxis by the gray treefrog, Hyla versicolor (= HV), was studied behaviorally and neurophysiologically. Most stimuli were played back at 85 dB SPL, a level typically measured at 1-2 m from a calling male. 2. Females preferred stimuli with conspecific pulse shapes at 20 degrees and 24 degrees C, but not at 16 degrees C. Tests with normal and time-reversed pulses indicated the preferences were not influenced by the minor differences in the long-term spectra of pulses of different shape. 3. Pulse shape

Role of ventrolateral medulla in reflex cardiovascular responses to activation of skin and muscle nerves.

Central neuronal circuits mediating reflex cardiovascular responses to skin and muscle nerve stimulation were studied in rats under urethan anesthesia. Responses of right rostral ventrolateral medulla (RVLM) and caudal ventrolateral medulla (CVLM) cardiovascular neurons to stimulation of contralateral skin and muscle afferent fibers were investigated. Stimulation of the tibial (muscle) nerve excited 19 (86%) of 22 CVLM neurons and inhibited 18 (82%) of 22 RVLM neurons. Stimulation of the sural (skin) nerve excited 20 (91%) of the 22 RVLM neurons but did not affect the firing rate of any of the

Monoamines and nitric oxide are employed by afferents engaged in midline thalamic regulation.

The neurochemical identities of afferents to the midline thalamus were investigated in chloral hydrate-anesthetized adult Sprague-Dawley rats. The retrograde tracers, FluoroGold or cholera toxin B subunit, were centered on the paraventricular thalamic nucleus (n.Pvt), a periventricular member of the diffuse thalamocortical projection system that is reciprocally linked with visceral areas of cerebral cortex and implicated in food intake and addictive behavior. Tissues were processed with antisera raised against 5-HT, the catecholamine-synthesizing enzymes, tyrosine hydroxylase or phenylethanola

Effect of nitric oxide inhibition on capsaicin-elicited vasodilation in the rat oral circulation.

The effects of local application of capsaicin on the vascular conductance of the oral structures (upper gingiva, lower gingiva, tongue, right and left submandibular glands) were studied with and without pretreatment with NG-nitro-L-arginine methyl ester (L-NAME), a potent inhibitor of nitric oxide formation in rats. Alterations in tissue blood flow were measured by laser-Doppler-flowmetry; systemic blood pressure was monitored continuously during the experiments. Local application of increasing concentrations of capsaicin (1.0; 3.33; 10.0; 33.3 mM; in a volume of 5 microliters) resulted in a d

Overexpression of nerve growth factor in transgenic mice induces novel sympathetic projections to primary sensory neurons.

Peripheral nerve crush induces novel projections from noradrenergic sympathetic neurons to sensory ganglia, and it has been suggested that these projections provide an anatomical substrate for chronic pain syndromes that occur after nerve injury. The present study demonstrates that novel sympathetic projections to sensory neurons are also induced in transgenic mice that overexpress nerve growth factor (NGF) in the skin. Specifically, a large proportion of trigeminal neurons in NGF transgenic mice were innervated by tyrosine hydroxylase (TH)-positive pericellular arborizations that were seen on

Abdominal vagal afferents excite A1 area neurons antidromically activated from the region of the supraoptic nucleus in the rabbit.

We made extracellular recordings from 113 spontaneously active neurons in the A1 area, after identifying the cells by antidromically activating them from the region of the supraoptic nucleus in urethane-anesthetized rabbits. We tested the response of these neurons to inputs from abdominal vagal, renal and somatic nerves. Electrical stimulation of the abdominal vagus nerve activated 64/85 neurons tested (75%), and had no effect on the remaining 25%. Latency was 195 +/- 25 ms, (conduction velocity 0.7 m/s). Stimulation of renal afferents had no effect on the discharge rate of 4 neurons tested. S

Vagal afferent stimulation-produced effects on nociception in capsaicin-treated rats.

1. The effects of electrical stimulation of cervical vagal afferent fibers on the nociceptive tail-flick (TF) reflex and responses of spinal dorsal horn neurons to noxious cutaneous stimulation were studied in adult rats treated as neonates with either capsaicin or vehicle. 2. Vagal afferent stimulation (VAS) produced biphasic, intensity-dependent effects on the TF reflex in vehicle-treated and untreated control rats. The TF reflex was facilitated in both groups of rats at lesser intensities of VAS (2.5-50 microA) and fully inhibited at greater intensities of VAS (50-100 microA). In contrast,

Physiology and morphology of neurons in the dorsal motor nucleus of the vagus and the nucleus of the solitary tract that are sensitive to distension of the small intestine.

We have recently shown that distension-sensitive vagal afferents are part of a neural circuit affecting absorption of water in the rat small intestine. Our results indicated that vagal afferent activity directly or indirectly influences the activity of neurons in the dorsal motor nucleus of the vagus (DMNV). In the present study we pursued this interaction by examining the structure and function of neurons in the DMNV and nucleus of the solitary tract (NST) that responded to moderate distension of the small intestine. Distension-sensitive cells were filled by intracellular iontophoretic inject

Visceral targets specify calcitonin gene-related peptide and substance P enrichment in trigeminal afferent projections.

Rat trigeminal ganglion projections to a visceral target (intracranial blood vessels) are enriched in calcitonin gene-related peptide (CGRP) and substance P (Sub P) compared to trigeminal ganglion projections to a cutaneous target (the forehead skin). We asked if transplants of a novel visceral target (fetal stomach antrum tissue) into the path of the neonatal rat trigeminal frontal nerve projection to forehead skin would induce neuronal CGRP and Sub P enrichment. By postnatal day (P) 25, the percentage of nerves containing CGRP increased from 14-15% in the control trigeminal projection to for

Somatosensory evoked response evaluation of cervical spondylytic myelopathy.

There were 13 patients with cervical spondylytic myelopathy (CSM) evaluated. All had extradural defects with distortion of the cervical cord and partial or complete obstruction of myelographic dye. Posterior tibial scalp (SSEPs) were absent or delayed in all 13, whereas median SSEPs were abnormal in 9. Median SSEP abnormalties were limited to those with cord lesions at C5-C6 or above. There were eight patients with associated radiculopathies confirmed by electromyography. No meaningful differences in SSEPs were noted between those with or without root injury. Surgery confirmed the level of cor

Evaluation of the dermatomal somatosensory evoked potential in the diagnosis of lumbo-sacral root compression.

The dermatomal somatosensory evoked potential from the lumbo-sacral dermatomes was recorded from 21 patients with radiographically and surgically (20) proven lumbo-sacral root compression due to prolapsed intervertebral disc or canal stenosis. The potential was abnormal in 19 of the 20 surgically proven cases. The dermatomal somatosensory evoked potential is as accurate as myelography for diagnosis but has the advantage of being non-invasive and repeatable. It provides useful additional diagnostic and pathophysiological information about lumbo-sacral root compression.

Further evidence for, and nature of, the facilitatory GABAergic influence on central noradrenergic transmission.

In order to explore the nature of the facilitatory GABAergic control of cerebral noradrenergic neurons, we have studied the effect of a variety of GABA mimetics (given systemically or injected locally into brain areas containing noradrenergic cell bodies or terminals) on several indices of noradrenaline turnover in the rat brain. Systemic administration of both direct and indirect acting GABA mimetics enhanced; 1) the pargyline induced accumulation of normetanephrine in the hypothalamus; 2) total DOPEG levels in a number of brain regions innervated by noradrenergic neurons; 3) both DOPAC and M
