Korean red ginseng extract inhibits microplastic translocation via the gut-liver axis by ameliorating alcohol-induced intestinal disruption.

BACKGROUND: Impaired intestinal mucosal barrier in alcoholic liver disease leads to the entry of toxic substances into the liver parenchyma. Ingestion of microplastics (MPs; plastic particles sized <5 mm) can induce inflammation, metabolic disorders, oxidative stress, and cancer. Red ginseng extract (RGE) is a widely used herbal medicine globally. The effects of RGE on the accumulation of MPs and the underlying mechanisms remain unclear. METHODS: Nine-week-old male wild-type C57BL/6 mice were fed a control liquid diet or ethanol diet with or without MP and RGE. MPs (fluorescent-tagged 2.16-μm

Design, synthesis, and evaluation of anti-solid-tumor activity of niclosamide-based STAT3/HDAC dual-target inhibitors.

Inspired by reports that HDAC blockade can trigger compensatory activation of the LIFR-JAK1-STAT3 axis in solid tumors, we designed and synthesized a series of niclosamide-based STAT3/HDAC dual-target inhibitor candidates by incorporating a SAHA-derived hydroxamate zinc-binding group into the pleiotropic, STAT3-modulating niclosamide scaffold. Biological evaluation identified NS06 as the best-balanced analogue, with IC50 values of 1.49 and 1.41 μM against MDA-MB-231 and HCT116 cells, respectively. Mechanistic studies showed that NS06 bound STAT3 in vitro (SPR, KD = 5.82 μM), suppressed STAT3 p

Cholic acid derivatives as novel PTP1B inhibitors: Their synthesis, inhibition mechanism, and hypoglycemic effects.

Protein tyrosine phosphatase 1B (PTP1B) is a validated therapeutic target for type 2 diabetes mellitus (T2DM). In this study, 37 cholic acid derivatives were designed and synthesized via a molecular hybridization strategy. Biological evaluation identified the 4-fluorophenylalanine derivative 6h as the most potent PTP1B inhibitor (IC50 = 32.70 μM) with a selectivity index of 7.0 over TCPTP. Structure-activity relationship (SAR) studies revealed that the 3-hydroxy-7-oxo steroid core, a free carboxylic acid group, and electron-withdrawing aromatic substituents are essential for maintaining activi

A Spermidine Derivative Ameliorates Dextran Sulfate Sodium-Induced Colitis in Mice by Inhibiting the MAPK4/AKT Signaling Pathway.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by recurrent episodes and an inability to achieve a complete c … ting colitis (p < 0.05). In addition, SPDD treatment also ameliorated TNF-α-induced inflammation in Caco-2 cells. Overall, our study demonstrated that SPDD reversed colonic inflammation in colitis mice through the MAPK4/AKT pathway and might be a promising candidate for UC intervention.

Novel Liposome Eencapsulated Guanosine Di Phosphate based Therapeutic Target against Anemia of Inflammation.

Hepcidin, master regulator of iron homeostasis, causes anemia under infectious and inflammatory conditions by reducing intestinal absorption of iron with decreased release of iron from macrophages and liver despite adequate iron stores leading to Anemia of Inflammation (AI). Many therapeutic trials have been carried out but none have been effective due to its adverse effects. In present study, we discover that Guanosine 5'-diphosphate (GDP) encapsulated in lipid vesicle (NH+) was found to inhibit NF-ҝB activation by limiting phosphorylation and degradation of IҝBα, thus, attenuating IL-6 secre

Chickpea aquafaba-stabilised pickering-type emulsions for chilli oleoresin encapsulation: capsaicin release, bioaccessibility and cellular bioactivity following in vitro digestion.

Aquafaba (AF) from cooked British Kabuli chickpeas, containing naturally occurring nanoparticles and denatured proteins generated during cooking, was evaluated as a Pickering-type stabiliser for chilli oleoresin encapsulation and capsaicin delivery. An optimised emulsion containing 15% AF, 6.64% oil phase (1,1 oleoresin, MCT) and 78.36% water was selected due to its high physical stability. Encapsulation efficiencies exceeded 97% for both oleoresins. Particle size and ζ-potential measurements indicated stability during digestion. Low capsaicin release during oral and gastric phases suggested e

Lactose-Gated Mesoporous Silica Particles for Intestinal Controlled Delivery of Essential Oil Components: An In Vitro and In Vivo Study.

Mesoporous silica microparticles functionalized with lactose for the specific release of essential oil components (EOCs) in the small intestine are presented. In vitro and in vivo intestinal models were applied to validate the microparticles (M41-EOC-L), in which the presence of lactase acts as the triggering stimulus for the controlled release of EOCs. Among the different microdevices prepared (containing thymol, eugenol and cinnamaldehyde), the one loaded with cinnamaldehyde showed the most significant Caco-2 cell viability reduction. On the other hand, interaction of the particles with ente

The Intestinal Efflux Transporter Inhibition Activity of Xanthones from Mangosteen Pericarp: An In Silico, In Vitro and Ex Vivo Approach.

The capacity of α-mangostin (α-MG) and β-mangostin (β-MG) from mangosteen pericarp on P-glycoprotein (Pgp) in silico, in vitro, and ex vivo was investigated in this study. Screening with the ADMET Predictor™ program predicted the two compounds to be both a Pgp inhibitor and Pgp substrate. The compounds tended to interact with Pgp and inhibit Pgp ATPase activity. Additionally, bidirectional transport on Caco-2 cell monolayers demonstrated a significantly lower efflux ratio than that of the control (α-(44.68) and β-(46.08) MG versus the control (66.26); p < 0.05) indicating an inhibitory effect

Modified Dietary Fiber from Cassava Pulp and Assessment of Mercury Bioaccessibility and Intestinal Uptake Using an In Vitro Digestion/Caco-2 Model System.

The ability of modified dietary fiber (MDF) generated from cassava pulp to modulate the bioaccessibility and intestinal absorption of heavy metals may be helpful to mitigate health risk associated with select foods including select fish high in methyl mercury. Using a coupled in vitro digestion/Caco-2 human intestinal cell model, the reduction of fish mercury bioaccessibility and intestinal uptake by MDF was investiaged. MDF was prepared from cassava pulp, a byproduct of tapioca production. The highest yield (79.68%) of MDF was obtained by enzymatic digestion with 0.1% α-amylase (w/v), 0.1% am

Complete amino acid sequence of human cartilage link protein (CRTL1) deduced from cDNA clones and chromosomal assignment of the gene.

Little is known about the primary amino acid structure of human cartilage link protein (CRTL1). We screened a human genomic library with a cDNA encoding the 3' untranslated region and the adjoining B1 domain of chicken link protein. One clone was isolated and characterized. A 3.5-kb EcoRI-KpnI fragment from this genomic clone that contains the human B1 exon was used to map the gene to chromosome 5q13----q14.1. The same fragment was used to screen a cDNA library prepared from mRNA of Caco-2, a human colon tumor cell line. Two overlapping clones were isolated and shown to encode all of CRTL1. Th

Polydextrose reduces the infection of Klebsiella pneumoniae in mice by downregulating the expression of TamA.

Polydextrose (PDX), as a prebiotic, is an extensively branched glucose polymer that can promote the growth of beneficial bacteria in the gut. Recent research indicates that PDX regulates intestinal function and supports immune balance, which helps to protect the gut from pathogenic bacteria. However, scarce research has been found that PDX prevents the host infection through the direct effects on the pathogen. In this study, we developed a mouse model infected with Klebsiella pneumoniae by pretreating with PDX, assessed the effect of PDX on K. pneumoniae acute infection in mice, and explored i

hsa_circ_0015388 Reduces Macrophage Derived Reactive Oxygen Species in Crohn's Disease.

BACKGROUND: Crohn's disease (CD) is a refractory inflammatory bowel disease with an unclear etiology. CircularRNA (circRNA) has been highlighted as a novel class of functional noncoding RNAs associated with the pathogenesis of various diseases. However, the functions of circRNA in CD remain unclear. METHODS: Biopsies were obtained from noninflammatory sites in the terminal ileum of the CD group (n = 4) and non-CD group (n = 4) and analyzed for circRNA expression using RNA sequencing. The significantly altered circRNAs were validated in the CD group (n = 45) and non-CD group (n = 15) using qua

Biological Insights into Intestinal Adaptation from Preclinical Models of Short Bowel Syndrome.

Short bowel syndrome is a rare and clinically heterogeneous condition resulting from extensive intestinal resections or functional impairment, leading to malabsorption, fluid and electrolyte losses, and potential progression to intestinal failure requiring long-term parenteral nutrition. The long-term outcome of SBS is largely determined by intestinal adaptation, a progressive physiological response involving epithelial remodeling, intestinal stem cell expansion, lineage-specific proliferation, vascular remodeling, and the activity of trophic mediators-most notably glucagon-like peptide-2 (GLP

Chloroquine synergizes sunitinib cytotoxicity via modulating autophagic, apoptotic and angiogenic machineries.

Tyrosine kinases play a pivotal role in oncogenesis. Although tyrosine kinase inhibitors as sunitinib malate are used in cancer therapy, emerging studies report compromised cytotoxicity when used as monotherapy and thus combinations with other anti-cancer agents is recommended. Chloroquine is a clinically available anti-malarial agent which has been shown to exhibit anti-cancer activity. In the current study, we questioned whether chloroquine can modulate sunitinib cytotoxicity. We found that chloroquine synergistically augmented sunitinib cytotoxicity on human breast (MCF-7 and T-47D), cervic

West Nile virus infection causes endocytosis of a specific subset of tight junction membrane proteins.

West Nile virus (WNV) is a blood-borne pathogen that causes systemic infections and serious neurological disease in human and animals. The most common route of infection is mosquito bites and therefore, the virus must cross a number of polarized cell layers to gain access to organ tissue and the central nervous system. Resistance to trans-cellular movement of macromolecules between epithelial and endothelial cells is mediated by tight junction complexes. While a number of recent studies have documented that WNV infection negatively impacts the barrier function of tight junctions, the intracell

A Novel Epigenetic Mechanism of Intestinal NHE3 Gene Regulation by Histone Acetylation.

NHE3 is an essential Na+ transporter in the intestine, contributing to diarrhea associated with inflammatory bowel disease (IBD) or intestinal infections. Nevertheless, the influence of histone acetylation on intestinal NHE3 gene expression has not yet been investigated. Various methods were employed, including human intestinal epithelial cells (Caco2 cells), ex vivo mouse and human ileal enteroids, and in vivo mouse models, along with techniques such as real-time qPCR, Western blot, immunofluorescence staining, ChIP, and H3K27Ac HiChIP sequencing analyses. Our findings indicated that valproic

Chlorogenic acid regulates the expression of NPC1L1 and HMGCR through PXR and SREBP2 signaling pathways and their interactions with HSP90 to maintain cholesterol homeostasis.

BACKGROUND: Hypercholesterolemia is widely implicated in the etiology of coronary heart disease, stroke, and dementia. Evidence suggests that chlorogenic acid (CA) reduces the risk of cardiovascular disease. PURPOSE: The current study aims to explore the underlying molecular mechanism of CA in lowering cholesterol based on pregnane X receptor (PXR) and sterol regulatory element-binding protein 2 (SREBP2) regulatory pathways and their interactions with heat shock protein 90 (HSP90). METHODS: A hypercholesterolemic mouse model, HepG2 and Caco2 cell models, metabolomics analysis, and co-immunop

Link of a ubiquitous human coronavirus to dromedary camels.

The four human coronaviruses (HCoVs) are globally endemic respiratory pathogens. The Middle East respiratory syndrome (MERS) coronavirus (CoV) is an emerging CoV with a known zoonotic source in dromedary camels. Little is known about the origins of endemic HCoVs. Studying these viruses' evolutionary history could provide important insight into CoV emergence. In tests of MERS-CoV-infected dromedaries, we found viruses related to an HCoV, known as HCoV-229E, in 5.6% of 1,033 animals. Human- and dromedary-derived viruses are each monophyletic, suggesting ecological isolation. One gene of dromedar
