Epidemiological studies have shown an inverse correlation between the incidence of lymphatic filariasis (LF) and the incidence of allergies and autoimmunity. However, the interrelationship between LF and type-2 diabetes is not known and hence, a cross sectional study to assess the baseline prevalence and the correlates of sero-positivity of LF among diabetic subjects was carried out (n = 1416) as part of the CURES study. There was a significant decrease in the prevalence of LF among diabetic subjects (both newly diagnosed [5.7%] and those under treatment [4.3%]) compared to pre-diabetic subjec

Lymphatic filariasis is targeted to be eliminated globally, at least as a public-health problem, by 2020. The comprehensive strategy for achieving this goal includes the interruption of the transmission of the causative parasites, by drastically reducing the prevalences of microfilaraemia in at-risk communities. In a descriptive, comparative, cross-sectional and community-based study, the impact of the 2004 mass drug administration (MDA) on filarial infection, in selected areas of the Western province of Sri Lanka, has now been assessed 1-2 and 11 months after the administration of the diethyl

In a study undertaken among rural and urban communities in a district of Orissa, India, the personal-protection measures used against mosquitoes, and the household costs of these measures, were investigated. Most people living in the study communities perceived mosquitoes as a problem, both as a biting nuisance and as vectors of human disease. Almost all (99%) of the urban households investigated and most (84%) of the rural each reported the use of at least one measure against mosquitoes. Most of the study households (92% of the urban and 64% of the rural) used a 'modern' chemical method (coil

Nematode polyprotein allergens (NPA) are lipid binding/transport molecules that elicit elevated levels of IgE response in the infected host, leading to Th2 type of immune response. They also transport arachidonic acid and its metabolites that are known to be involved in the action of antifilarial drug, Diethylcarbamazine and hence are of great significance for the control of lymphatic filariasis. We investigated the polymorphism of gp15/400 polyprotein of 35 isolates of lymphatic filarial parasite Wuchereria bancrofti collected from different geographic locations of India. The repeat sub-unit

BACKGROUND: Lymphatic filariasis (LF) is a mosquito-borne nematode infection that causes permanent lymphatic dysfunction in virtually all infected individuals and clinical disease in a subset of these. One major sequel of infection is lymphoedema of the limbs. Lymphoedema of the leg affects an estimated 15 million persons in LF-endemic areas worldwide. Acute dermatolymphangioadenitis (ADLA) in people with filarial lymphoedema causes acute morbidity and increasingly severe lymphoedema. Episodes of ADLA are believed to be caused by bacteria, and it has been shown that entry lesions in the skin p

This study describes a novel phage display method based on an iterative subtraction strategy to identify candidate vaccine antigens of Brugia malayi. A cDNA library of the infective larval stage of B. malayi expressed on the surface of T7 phage was sequentially screened with sera samples from human subjects showing different manifestations of the disease. Antigens that selectively and specifically bind to immune sera were then enriched using a multi-step panning procedure. This strategy identified five antigens, four of which were previously reported (ALT-2, TPX-2, VAH and COX-2) and the other

The geographical distribution of human infection with Wuchereria bancrofti was investigated in four West African countries (Benin, Burkina Faso, Ghana and Togo), using a commercial immunochromatographic test for filarial antigen. Efforts were made to cover each health-system implementation unit and to ensure no sampling point was >50 km from another, but otherwise the 401 study communities were selected at random. The aim was to enable spatial analysis of the data, to provide a prediction of the overall spatial relationships of the infection. The results, which were subjected to an independent

The elimination of lymphatic filariasis as a public-health problem is currently dependent on the delivery of annual drug treatments to at least 80% of the eligible members of endemic populations for at least 5 years. However, for various reasons, this goal may not be achievable by the health systems of most endemic countries in sub-Saharan Africa, particularly if treatment is not community-directed. In Ghana, community-directed ivermectin treatment involving the regular public-health services at the implementation level (ComDT/HS) has recently been compared with mass-treatment in which only th

BACKGROUND: In lymphatic filariasis due to Wuchereria bancrofti infections, the relationship between the natural course of infection and development of clinical disease remains controversial. The two hypotheses that are widely considered are (1) microfilaraemia represents an early stage of infection which progresses to amicrofilaraemic clinical disease and (2) microfilaraemia and clinical disease are two sequentially unrelated independent entities of the filarial infection and disease. Aim To determine whether microfilaraemic individuals develop lymphatic disease.
METHODS: The study was condu

A filariasis survey was conducted in July 1998 in Pathankot town of Punjab covering a population of 28, 041. A total of 2136 blood smears were collected from migratory and local inhabitants. Microfilaria (Mf) rate and mean Mf density was 1.19 and 15.05 respectively. Disease rate was nil. All the 20 microfilaria carriers detected were known to hall from filaria endemic areas of different states. Mf rate was highest in 20-49 years age-group, whereas Mf density was high in younger age group. Wuchereria bancrofti was the only infection encountered. 10 Men Hour Density of Culex quinquefasciatus ran

Brugia malayi-infected patients, endemic normals with high levels of specific antibodies and European controls were investigated for kidney disorders by noninvasive techniques. Groups of patients with filarial infections included asymptomatic, microfilaraemic cases (group 1), patients with filarial fever (group 2) and with obstructive filariasis (group 3). Several patients underwent a treatment course with diethylcarbamazine (DEC) when blood and urine samples were collected. Urine samples were investigated for proteinuria and analysed by SDS-PAGE to discriminate between proteinurias caused by

This year-long study investigated the epidemiology of acute filarial episodes due to Wuchereria bancrofti in 2 rural villages in south India. The annual incidence of 96.5 episodes/1000 population was significantly higher in males (108.5) than females (84.1) an strongly age dependent. First occurrence of acute disease was observed in 0.86% of the population, and the average duration of each episode was 3.6 +/- 2.0 d. Although more than half (63.5%) of the affected individuals suffered only 1 episode, a few experienced as many as 8 over the one-year period. Individuals with chronic disease were

Patients with chronic helminth infections, despite having abundant basophils and mast cells specifically sensitized with antiparasite IgE and often exposed repeatedly to parasite Ag, rarely manifest allergic symptoms. This control of clinical allergic reactivity likely results from Ag-specific IgG "blocking antibodies" shown previously to be abundant in the sera of such patients. In the present study we used two approaches to determine in which of the four IgG subclasses this blocking activity was localized. First, specific antifilarial antibodies of each of the four IgG subclasses were quanti

Rates of acquisition and loss of Brugia malayi microfilaraemia were estimated using the parasitological data of a cohort of population in Shertallai, South India. The rate of acquisition of microfilaraemia was found to be dependent on age but not gender. The decline in the rate of acquisition of microfilaraemia in adults above 35 years could be due to the development of acquired immunity. The mean reproductive lifespan for the periodic Brugia malayi adult female worm was estimated to be 3.4 years and it was independent of host age and gender. The age-specific estimated proportion of population

OBJECTIVES: Circulating filarial antigen (Ag) is used by elimination programs to monitor lymphatic filariasis (LF) transmission; however, antifilarial antibodies (Ab) may be more sensitive than Ag for detecting LF. Our objectives were to describe Ab seroprevalence, identify risk factors for Ab seropositivity, investigate age-specific associations between Ag and Ab, and evaluate geographic clustering of seropositivity.
METHODS: Community-based serosurveys of participants aged ≥5 years were conducted in 35 primary sampling units (PSUs). Ag-positivity was detected using Alere™ Filariasis Test St

BACKGROUND: Onchocerciasis and lymphatic filariasis (LF) are endemic in Equatorial Guinea with notable variations in disease incidence between island and mainland regions. Historically, efforts to control and map these diseases were concentrated in Bioko Island, where loiasis is absent, allowing for targeted onchocerciasis interruption strategies. With the cessation of onchocerciasis transmission on Bioko and no reported cases on Annobon island, assessing the transmission status in the previously unaddressed mainland region has become imperative. Mapping efforts in mainland Equatorial Guinea h
