A mouse model of a patient derived P544L mutation in the Slc6a8 gene shows hypoactivity and cognitive deficits.

Creatine (CR) is essential for normal brain function. A lack of brain CR results in intellectual disability, epilepsy, and language delay in humans. The most common cause of CR deficiency in humans results from mutations in the CR transporter (SLC6A8). Several large deletion models of Slc6a8 have been characterized and are excellent models for global creatine loss. However, other SLC6A8 variants are reported in humans with creatine transporter deficiency (CTD), including missense mutations, deletions, and point mutations resulting in phenotypes ranging from mild to severe in humans. The purpos

In-context adaptation of VLMs for few-shot cell detection in optical microscopy.

Foundation vision-language models (VLMs) excel on natural images, but their utility for biomedical microscopy remains underexplored. In this paper, we investigate how in-context learning enables state-of-the-art VLMs to perform few-shot object detection when large annotated datasets are unavailable, as is often the case with microscopic images. We introduce the Micro-OD benchmark, a curated collection of 252 images specifically curated for in-context learning, with bounding-box annotations spanning 11 cell types across four sources, including two in-lab expert-annotated sets. We systematically

Hybrid rule-based and on-premises LLM pipeline for extracting CMR and CPET metrics from free-text reports in repaired tetralogy of Fallot.

BACKGROUND: Patients with repaired tetralogy of Fallot (rTOF) require lifelong surveillance with cardiovascular magnetic resonance (CMR) and cardiopulmonary exercise testing (CPET). However, results are frequently stored as unstructured free-text reports, hindering large-scale analysis and research. OBJECTIVES: This study aimed to develop and evaluate a privacy-preserving hybrid natural language processing pipeline combining regular expressions (regex) and an on-premises large language model (LLM) to accurately extract key CMR and CPET metrics from legacy free-text reports in patients with rT

Granger-TSllm: Granger causality enhanced LLMs with residual-quantized tokenizer for multivariate time series forecasting.

Multivariate time series (MTS) forecasting is essential for process monitoring and decision optimization in numerous real-world domains. Existing deep models need to be trained from scratch on abundant data, resulting in limited generalization. Inspired by the success of large language models (LLMs) on various cross-modality tasks, recent research introduces LLMs for MTS forecasting. However, due to the inherent modality gap between MTS and text, there remain two challenges in generating MTS representations suitable for LLMs and handling the intricate inter-variable dependencies. To tackle the…

On the Role of Academic Buoyancy and Self-Efficacy in Predicting Teachers' Work Engagement: A Case of Chinese English as a Foreign Language Teachers.

A plethora of scientific evidence has pinpointed that teaching English as a foreign or second language (EFL) is difficult, challenging, and emotionally burdensome. Nevertheless, most English language teachers remain committed to the teaching profession and actively engage in the instructional environment. This has inspired several scholars worldwide to explore what personal, emotional, and psychological factors motivate English language teachers to engage enthusiastically in their workplace. While a large body of studies have to date examined the personal, emotional, and psychological predicto

Brain-inspired cognitive state modeling of complex English discourse reading: an LLM-parameterized hierarchical semi-Markov approach.

INTRODUCTION: Complex English discourse reading requires readers to integrate lexical access, syntactic parsing, local meaning construction, cross-sentence inference, and discourse monitoring over time. METHODS: We developed a brain-inspired, large-language-model-parameterized hierarchical hidden semi-Markov model that encodes lexical-, sentence-, and discourse-level representations and maps contextual embeddings to state-emission, transition, and duration parameters. Discourse-structure and cognitive-load constraints were incorporated into global decoding. RESULTS: Experiments on 1,200 Engl

Right hemisphere dominance for language in a woman with schizophrenia and a porencephalic cyst of the left hemisphere.

A large left hemisphere porencephalic cyst was incidentally found in a 48-year-old woman (MS) with a Diagnostic and Statistical Manual (DSM)-5 diagnosis of schizophrenia. The encephaloclastic characteristics of the cyst indicated that it was acquired between the 22nd and 24th gestational weeks, after the major waves of neuronal migration had tapered off. The cyst destroyed the left temporal and occipital lobes, and the inferior parietal lobule. Surprisingly, MS had no evidence of aphasia, alexia, agraphia, or ideational apraxia; in contrast, cognitive functions dependent on the integrity of th

FHIR-AgentEval: A Modular Sandbox for Benchmarking Clinical LLM Agents with an Evaluation of Memory-Augmented Configurations.

Healthcare data exchange increasingly relies on HL7 FHIR, but FHIR's implementation complexity creates barriers for clinical workflows. Large language model (LLM) agents could bridge this gap by translating natural language requests into structured FHIR operations, yet their reliability remains unproven. We present FHIR-AgentEval, an extensible evaluation sandbox comprising 43 modular tasks for benchmarking LLM agents on realistic appointment management and genetic testing workflows. Each task executes against a resettable FHIR server with custom deterministic validation of both agent response

Alzheimer's disease (AD) is a progressive neurological condition characterized by cognitive decline, memory loss, and aberrant behaviour. It affects millions of people globally and is one of the main causes of dementia. The neurodegenerative condition known as AD has intricate, multifaceted mechanisms that make it difficult to comprehend and identify in its early stages. Conventional diagnostic techniques frequently fail to detect the disease in its early stages. By combining Natural Language Processing (NLP) and Large Language Models (LLMs), this research suggests a novel approach for identif…
