Chromatographic separation of reaction products from the choline acetyltransferase and carnitine acetyltransferase assay: differential ChAT and CrAT activity in brain extracts from Alzheimer's disease versus controls.

Choline acetyltransferase (ChAT) catalyzes the reaction between choline and acetylcoenzyme A (AcCoA) to form acetylcholine (ACh) in nerve terminals. ACh metabolism has implications in numerous aspects of physiology and varied disease states, such as Alzheimer's disease. Therefore a specific, sensitive, and reliable method for detecting ChAT enzyme activity is of great utility in a number of situations. Using an existing radionuclide-based enzyme activity assay, we have observed detectable ChAT signals from non-cholinergic cells, suggesting a contaminant in the assay producing an artifactual si

Cognitive impairment in Parkinson disease correlates with loss of cholinergic axon projections to cortex in an autopsy study.

INTRODUCTION: People with Parkinson's disease (PD) frequently develop dementia. We investigated the relationship between cognitive impairment and the loss of cholinergic axon projections from the nucleus basalis of Meynert to neocortical regions in PD. METHODS: Levels of choline acetyltransferase (ChAT), the enzyme required for acetylcholine synthesis, were measured with a sandwich enzyme-linked immunosorbent assay (ELISA) in eight brain regions from 37 PD autopsy cases. Analyses focused on relationships between measures of global cognitive impairment and average ChAT levels in four neocortic

Cholinergic autonomic dysfunction in veterans with Gulf War illness: confirmation in a population-based sample.

BACKGROUND: The authors of prior small studies raised the hypothesis that symptoms in veterans of the 1991 Gulf War, such as chronic diarrhea, dizziness, fatigue, and sexual dysfunction, are due to cholinergic autonomic dysfunction. OBJECTIVE: To perform a confirmatory test of this prestated hypothesis in a larger, representative sample of Gulf War veterans. DESIGN: Nested case-control study. SETTING: Clinical and Translational Research Center, University of Texas Southwestern Medical Center, Dallas. PARTICIPANTS: Representative samples of Gulf War veterans meeting a validated case definit

Regulation of synaptic plasticity in a schizophrenia model.

The pathology of schizophrenia is characterized by increased hippocampal activity at baseline and during auditory hallucinations. Animal-model studies in which the flow of activity to the hippocampus is increased through decreased amygdalar GABAergic inhibition have shown alterations of hippocampal circuitry similar to schizophrenia, but the functional importance of this phenomenon remains unclear. We provide evidence of decreased hippocampal feed-forward and tonic GABA-mediated inhibition in this animal model, complementing increased hippocampal activity seen in neuroimaging and postmortem st

Compensatory increase of responses to nerve stimulation of the partially denervated rat urinary bladder.

The rat urinary bladder was deprived of half of its innervation by removing the pelvic ganglion on one side. The motor response of such a partially denervated bladder to stimulation of the pelvic nerve on the other side were examined 1 week, 1 month and 2 months postoperatively. On all three occasions the increase in pressure of the operated bladder was larger than that of the control bladder; the enlargement was most marked 2 months after operation. The responses were further enlarged by eserine, and markedly reduced by atropine. The present results combined with those of previous investigati

Effects and mechanisms of transcutaneous auricular vagus nerve stimulation on post-stroke cognitive impairment in animal studies: a systematic review and meta-analysis.

BACKGROUND: Post-stroke cognitive impairment (PSCI) substantially undermines rehabilitation adherence and poses a major obstacle to functional recovery. Transcutaneous auricular vagus nerve stimulation (taVNS), a noninvasive neuromodulation technique, has the potential to improve cognitive function, as demonstrated in various clinical and preclinical studies. Against this backdrop, we performed a meta-analysis of taVNS in experimental stroke models to quantify its cognitive benefits and probe the biological mechanisms involved in its effects. METHODS: We systematically searched databases, inc

Neurophysiological signals as predictive translational biomarkers for Alzheimer's disease treatment: effects of donepezil on neuronal network oscillations in TgF344-AD rats.

BACKGROUND: Translational research in Alzheimer's disease (AD) pathology provides evidence that accumulation of amyloid-β and hyperphosphorylated tau, neuropathological hallmarks of AD, is associated with complex disturbances in synaptic and neuronal function leading to oscillatory abnormalities in the neuronal networks that support memory and cognition. Accordingly, our recent study on transgenic TgF344-AD rats modeling AD showed an age-dependent reduction of stimulation-induced oscillations in the hippocampus, and disrupted long-range connectivity together with enhanced neuronal excitability

Selective antagonism of amino acid-induced and synaptic excitation in the cat spinal cord.

1. The effects of D-alpha-aminoadipate (DalphaAA), D-alpha-aminosuberate (DalphaAS) and other excitatory amino acid antagonists have been compared on the excitatory responses of neurones of the cat spinal cord to acetylcholine, a range of glutamate-related amino acids and stimulation of appropriate excitatory synaptic pathways. The ionophoretic technique was used for administration of excitants and antagonists. 2. DalphaAA and DalphaAS had little or no effect on acetylcholine-induced excitation of Renshaw cells. Responses of either Renshaw cells or dorsal horn neurones in the spinal cord to ex

Traumatic brain injury exacerbates alcohol consumption and neuroinflammation with decline in cognition and cholinergic activity.

Traumatic brain injury (TBI) is a global health challenge that substantially contributes to disability and is responsible for 30% of injury-related deaths. Annually, over 50 million TBIs occur worldwide, with many adult TBI patients in emergency departments presenting with alcohol in their system. TBI is also a known risk factor for alcohol abuse, yet its interaction with alcohol consumption remains poorly understood. In this study, we demonstrate that the fluid percussion injury (FPI) model of TBI in male C57BL/6 mice significantly increases alcohol consumption and impairs cognitive function.

Systemic oxybutynin decreases afferent activity of the pelvic nerve of the rat: new insights into the working mechanism of antimuscarinics.

AIMS: In a rat model, intravesical oxybutynin was recently shown to suppress pelvic afferent nerves. This study evaluates if a similar effect exists after systemic administration of oxybutynin. METHODS: Twenty-four single afferent bladder nerves were identified in 15 rats. Based on their conduction velocities they were grouped as C or Adelta fibers. Bladder filling parameters and afferent nerve spike rate were simultaneously recorded 30 min before administration of saline (nine fibers) or oxybutynin (15 fibers, 1 mg/kg), and again 30, 60, 90, 120, and 150 min after systemic saline or drug adm

Role of dentate gyrus cells in retention of a radial arm maze task and sensitivity of rats to cholinergic drugs.

Male adult Fischer-344 rats that received bilateral injections of colchicine into two rostrocaudal sites showed relatively long-lasting alterations in the performance of a previously acquired radial arm maze task and specific destruction of dentate granule cells. Results of subsequent experiments with cholinergic drugs indicated that physostigmine or nicotine had no effect on the number of errors made in the maze, although other signs of cholinergic or pharmacological activity were present. RS-86, an analog of the muscarinic agonist arecoline, decreased errors in colchicine-treated rats, but t

Cognitive enhancing effect of Canagliflozin in aluminum-induced rat model of Alzheimer's-like disease: Cross-talk between Amyloid-Β and BDNF/GSK-3β signaling.

The strong relationship between Alzheimer's Disease (AD) and diabetes mellitus (DM) is described by the term "type 3 diabetes". Canagliflozin (CAN), a sodium-glucose co-transporter 2 inhibitor (SGLT2i), is an antidiabetic agent under investigation as a potential new treatment for AD due to its acetylcholinesterase (AChE) inhibitory properties. We aimed to examine the effect of CAN on the efficacy of the anti-acetylcholinesterase, rivastigmine (RIV), against aluminum chloride (AlCl3)-induced AD rat model. The efficacy of CAN, RIV, and CAN plus RIV against abnormal behavioral, biochemical, and h

Maternal anti-collapsin response mediator protein 1 antibody inducing autism-like behaviors in offspring.

Maternal autoantibodies targeting neural proteins represent a major environmental contributor to autism spectrum disorder (ASD). Although anti-collapsin response mediator protein 1 (CRMP1) antibodies (Abs) have been detected in mothers of ASD children, the pathogenic role of anti-CRMP1 Ab in ASD remains unclear. Here, we evaluated the ability of maternal anti-CRMP1 Ab to induce pathology in a mouse model. Using a cell-based assay, we found that maternal anti-CRMP1 Ab persisted throughout gestation and was detected in offspring serum on postnatal day 16. Exposed offspring exhibited delayed deve

Neuroactive Phytoconstituents of Glycyrrhiza glabra for the Treatment of Alzheimer's Disease.

Alzheimer's Disease (AD), a prevalent neurodegenerative disorder, poses a significant global health challenge with complicated pathogenesis. Pathological characteristics of AD include increasing loss of cholinergic neurons, oxidative stress, mitochondrial dysfunction, and amyloid beta accumulation. Due to the limited availability of effective therapeutic options with only symptomatic relief and their severe adverse effects, there is a significant need to search and explore new agents for the management of AD. Recently, natural products and/or phytoconstituents of plants have gained notable att…

Mechanistic Insights into the Neuroprotective Potential of Aegle marmelos (L.) Correa Fruits against Aβ-Induced Cell Toxicity in Human Neuroblastoma SH-SY5Y Cells.

Background/Objectives: Amyloid-β (Aβ) plaque accumulation, oxidative stress, and cholinergic dysfunction are hallmarks of Alzheimer's disease (AD), a neurodegenerative disability that progresses over time, ultimately resulting in the loss of neurons. The side effects and limitations of current synthetic drugs have shifted attention toward natural alternatives. This study investigates the ethanolic extract of Aegle marmelos (L.) Corrêa fruits for their antioxidant, AChE-inhibitory, and anti-amyloidogenic properties, as well as their neuroprotective effects against amyloid beta-peptide (Aβ1-42).

Targeting GPR3 as a novel approach for nicotine cessation therapeutic development.

Tobacco use remains the leading cause of preventable death worldwide. Unfortunately, currently available cessation aids have limited long-term efficacy. GPR3 is a Gαs coupled receptor expressed in discrete brain regions, with notably high expression in cholinergic neurons of the medial habenula. Here, we investigated whether modulation of GPR3 could be a viable target for therapeutic development to promote nicotine cessation. We first examined whether our recently developed GPR3 receptor agonist, RTI-19318-32, could induce effects on intravenous nicotine self-administration at low, moderate or…
