Trichuris trichiura egg extract proteome reveals potential diagnostic targets and immunomodulators.

Embryonated eggs are the infectious developmental stage of Trichuris trichiura and are the primary stimulus for the immune system of the definitive host. The intestinal-dwelling T. trichiura affects an estimated 465 million people worldwide with an estimated global burden of disease of 640 000 DALYs (Disability Adjusted Life Years). In Latin America and the Caribbean, trichuriasis is the most prevalent soil transmitted helminthiasis in the region (12.3%; 95% CI). The adverse health consequences impair childhood school performance and reduce school attendance resulting in lower future wage-earn…

Demographic Factors Driving Schistosomiasis and Soil-Transmitted Helminthiases in Milola Ward, Lindi District, Tanzania: A Useful Guide for Launching Intervention Programmes.

BACKGROUND: Current information on the distribution of and risk factors for schistosomiasis and soil-transmitted helminthiases is scarce for … .95; prevalence, P=.88). DISCUSSION: These data confirm that in this setting, the mean age of peak helminthiasis prevalence decreases as transmission pressure increases, with non-school children below 18 years old being most at risk of acquiring parasitic infections. This was the first baseline survey of parasitic infections in Milola Ward, so the results will …

Imaging features of central nervous system paragonimiasis in children based on a large-scale case series study.

OBJECTIVES: Central nervous system (CNS) paragonimiasis is relatively uncommon and its imaging findings have not been fully described in the limited literature. Our aim was to investigate the comprehensive imaging features of CNS paragonimiasis based on a large-scale case series study, with an emphasis on the unreported imaging findings. METHODS: The clinical and imaging data of 206 pediatric patients with CNS paragonimiasis were continuously collected from 1181 cases of paragonimiasis in the past 12 years at the Children's Hospital of Chongqing Medical University (China). All patients underw

Maternal helminths rewire the microbiota to promote offspring antiviral immunity.

Maternal environmental exposures can alter microbiome composition and lead to changes in offspring immunity. Industrialization has led to significant shifts in the microbiome, but whether these have transgenerational impacts remains unclear. Here, we discovered that maternal helminths, an evolutionarily conserved mammalian partner lost in industrialized societies, confer broad and lasting protection against respiratory viruses in offspring. This heterologous antiviral immunity is mediated by helminth-induced changes in the maternal microbiota. The tryptophan metabolite indole-3-propionic acid

ILC2s govern imprinting of alveolar macrophage-mediated immune responses upon secondary helminth infection in the lung.

Hookworms migrate through the lung as part of their lifecycle, causing significant tissue damage that activates tissue-resident populations such as ILC2s. These drive type 2 immune responses critical for wound healing and the development of protective immunity. Monocyte-derived macrophages that seed the lung after infection are central effectors of these responses, but the cellular and molecular factors guiding their development and function remain poorly understood. We find that ILC2s and IL-4/IL-13-producing CD4+ T cells regulate alveolar macrophage populations during helminth infection. In

Cohort profile: the Entebbe Mother and Baby Study (EMaBS).

PURPOSE: The Entebbe Mother and Baby Study (EMaBS) was established in 2001 to test the hypothesis that treating helminth infections during pregnancy and early childhood could improve children's responses to Bacillus Calmette-Guérin (BCG) and other vaccines given in infancy and influence immune responses to other infectious pathogens. Follow-up was subsequently extended to address further research questions and continue to the present day. PARTICIPANTS: Two thousand five hundred and seven pregnant women were recruited when attending antenatal services at Entebbe General Hospital, Uganda; 2345

Type I IFN-induced IFN-γ instructs monocyte-mediated systemic bacterial protection during type 2 intestinal helminth infection.

Alterations to monocyte output and function occur during infections driving T helper 1 (TH1)-type inflammation. The degree to which monocytes respond to infections initiating alternative types of responses is poorly understood. Here, we describe a distinct state of the monocyte compartment associated with type 2-polarizing intestinal helminths. Unexpectedly, the adapted monocyte state in a type 2 setting was associated with acquisition of an interferon (IFN) signature. This IFN-induced state provided helminth-infected animals with systemic protection against secondary bacterial infection and a

Prevalence, common helminthes, and factors associated with helminthes among pregnant women attending antenatal clinic at a tertiary hospital in Uganda.

BACKGROUND: Helminthes in pregnant women is among the neglected tropical diseases. The Uganda ministry of health adopted the WHO recommendation of routine biannual deworming for girls and women of reproductive age and twice in pregnancy during the second and third trimesters. Despite the measures put in place, the prevalence of Helminthes among pregnant women in Uganda is still high which has implications for both the mother and to the developing fetus. METHODS: This was a hospital-based cross-sectional study carried out from January to April, 2024. Using Consecutive sampling method, 334 preg

Transitioning from microscopy to PCR for protozoa in Norway - Impact on detection of protozoa and helminths: A register study.

The aim of this study was to describe how the detection of protozoan and helminth parasites has been affected by the introduction of polymerase chain reaction (PCR) and changes in test algorithms. We extracted data about faecal samples tested for parasites (n = 114839) at five Norwegian clinical microbiology laboratories. Samples were classified into prePCR or postPCR depending on whether they were submitted before or after the introduction of PCR, and into diagnostic episodes (n = 99320). The number of diagnostic episodes increased 3.7-fold from prePCR to postPCR. Giardia positive episodes do

High burden of soil-transmitted helminth infections among preschool and school aged-children in North Kodi, Southwest Sumba, Indonesia: A crosssectional study.

5 Soil-transmitted helminth (STH) infections remain a significant global health concern due to their high prevalence in many areas and ease of transmission via the fecal-oral route. To control and enhance the declining helminth infection rate in children, Indonesia initiated a nationwide mass drug administration (MDA) in 2017, encompassing rural areas such as Southwest Sumba (SWS). Nevertheless, the prevalence of STH in SWS remained at 54% by 2021. Children are particularly susceptible owing to frequent exposure to helminth-egg-contaminated environments, which may lead to nutrient deficiencies

Prevalence and risk factors of gastrointestinal helminths infection in Brazilian horses: A retrospective study of a 12-year (2008-2019) diagnostic data.

Understanding gastrointestinal parasite distribution is crucial for effective control programs in horses. This study reports the prevalence of helminth infections in horses and selected risk factors (i.e., breed, age, climate, season) by analyzing 19,276 fecal samples from the Laboratory of Veterinary Clinical Parasitology, in Curitiba, Southern Brazil. The analyses were carried out from 2008 to 2019, coming from 153 stud farms located in 60 municipalities of nine Brazilian states. The parasite prevalence was 73.3%, with 72.1% present in the adult population and 80.6% in young horses. Strongyl

Interplay Between Helminth Infections, Malnutrition, and Gut Microbiota in Children and Mothers from Pemba, Tanzania: Potential of Microbiota-Directed Interventions.

BACKGROUND/OBJECTIVES: Despite efforts within the framework of the Sustainable Development Goal to end malnutrition by 2030, malnutrition and soil-transmitted helminth infections persist in sub-Saharan Africa. A significant barrier to success is the inadequate understanding of effective intervention methods. Most research on the gut microbiota's role in health has been conducted in developed countries, leaving a critical gap in knowledge regarding low-income populations. This study addresses this gap by expanding research on the gut microbiota of underprivileged populations to help tackle thes

Prevalence and intensity of soil-transmitted helminth infections among school-aged children in five districts in Uganda.

BACKGROUND: Soil-transmitted helminth (STH) infections, commonly caused by roundworms (Ascaris lumbricoides), whipworms (Trichuris trichiura), and hookworms (Necator americanus and Ancylostoma duodenale), were widespread among Ugandan schoolchildren in the late 1990s and early 2000s. Since 2003, the Ugandan Ministry of Health has administered biannual preventive chemotherapy to children aged 1-14 years to control these infections. Twenty years after the program's inception, there is scant data to show the long-term impact of these national deworming efforts. METHODS: To estimate the prevalenc

Context-dependent roles of B cells during intestinal helminth infection.

The current approaches to reduce the burden of chronic helminth infections in endemic areas are adequate sanitation and periodic administration of deworming drugs. Yet, resistance against some deworming drugs and reinfection can still rapidly occur even after treatment. A vaccine against helminths would be an effective solution at preventing reinfection. However, vaccines against helminth parasites have yet to be successfully developed. While T helper cells and innate lymphoid cells have been established as important components of the protective type 2 response, the roles of B cells and antibo

Fifteen Years of Sm-p80-Based Vaccine Trials in Nonhuman Primates: Antibodies From Vaccinated Baboons Confer Protection in vivo and in vitro From Schistosoma mansoni and Identification of Putative Correlative Markers of Protection.

Recent advances in systems biology have shifted vaccine development from a largely trial-and-error approach to an approach that promote rational design through the search for immune signatures and predictive correlates of protection. These advances will doubtlessly accelerate the development of a vaccine for schistosomiasis, a neglected tropical disease that currently affects over 250 million people. For over 15 years and with contributions of over 120 people, we have endeavored to test and optimize Sm-p80-based vaccines in the non-human primate model of schistosomiasis. Using RNA-sequencing o
