Teratogenic effects of external egg applications of methyl mercury in the mallard, Anas platyrhynchos.

The embryotoxic potential of external applications of methyl mercury on mallard eggs was investigated to assess the possible impact of mercury transferred from the plumage of effluent-contaminated aquatic birds to their eggs. Eggs were treated on day 3 of development with microliter applications of methyl mercury that was dissolved with ethyl acetate into an aliphatic hydrocarbon vehicle. Mercury analysis by atomic absorption indicated that almost half of the mercury applied entered the eggs past the shell membranes within several days of treatment. Most mortality occurred within this period a

[On the method of injecting into the yolk-sac in teratological studies in rabbits (author's transl)].

A method is described by which the embryotoxic and teratogenic effects of substances can be examined through their application into the yolk-sac. On the ninth day of gestation rabbits (white New Zealand) were laparatomised and the uterus bicornis together with the blastoderms were treated in 3 different experiments. The results show that after application of physiological saline into the yolk-sac the high rate of malformations and resorptions are caused mainly by an avoidable venous obstruction and by postoperative inflammations and infections.

Drug-induced nail disorders: incidence, management and prognosis.

A large number of drugs of different classes, ranging from antibacterials to chemotherapeutic agents to psoralens, can be responsible for the development of nail changes. Drug-induced nail changes usually involve several or all 20 nails and appear in temporal correlation with drug intake. Some nail changes are asymptomatic and only cause cosmetic problems, while others cause pain and discomfort and impair manual activities or deambulation. Drug-induced nail abnormalities are usually transitory and disappear with drug withdrawal, but sometimes persist in time. The pathogenesis of the nail chang

Safety of Valproic Acid Use in Pregnant Women: A Systematic Review and Meta-Analysis.

OBJECTIVE: To evaluate valproic acid (VPA) safety in pregnancy versus other antiepileptic drugs (AEDs), focusing on fetal malformation risks. METHODS: A literature search was conducted across PUBMED, MEDLINE, EMBASE, and Web of Science databases. Two researchers independently screened studies, extracted data, assessed quality, and analyzed VPA safety. RESULTS: This meta-analysis of 25 safety studies showed that compared with other AEDs, VPA's teratogenic risks were as follows: combined major congenital malformations (RR = 2.36, 95% CI: 2.17-2.56), neural tube defects (RR = 6.54, 95% CI: 4.51

Thalidomide-induced limb malformations: an update and reevaluation.

Historically, thalidomide-induced congenital malformations have served as an important example of the enhanced susceptibility of developing embryos to chemical perturbation. The compound produced a wide variety of congenital malformations in humans, which were initially detected by an association with a relatively rare limb defect labeled phocomelia. Although true phocomelia in the most severe form is a transverse defect with intercalary absence of limb regions, it is proposed that thalidomide produces a longitudinal limb phenotype in humans under usual circumstances that can become transverse

Risk of malformation after ondansetron in pregnancy: An updated systematic review and meta-analysis.

Ondansetron is increasingly used off label to treat nausea and vomiting during pregnancy. The main objective of this study was to evaluate the risk of major congenital malformations (MCM), cardiac defects and orofacial clefts associated with first trimester exposure to ondansetron using a meta-analytic approach. MEDLINE, ClinicalTrials.gov and Scopus were searched until November 2019. All comparative cohort and case-control studies on MCM, cardiac or orofacial defects and use of ondansetron during pregnancy were included. A team of paired reviewers independently extracted data using a propriet

The good and the bad of poisonous plants: an introduction to the USDA-ARS Poisonous Plant Research Laboratory.

This article provides an overview of the Poisonous Plant Research Laboratory (PPRL), about the unique services and activities of the PPRL and the potential assistance that they can provide to plant poisoning incidences. The PPRL is a federal research laboratory. It is part of the Agricultural Research Service, the in-house research arm of the U.S. Department of Agriculture. The mission of the PPRL is to identify toxic plants and their toxic compounds, determine how the plants poison animals, and develop diagnostic and prognostic procedures for poisoned animals. Furthermore, the PPRL's mission

A population-based case-control teratologic study of promethazine use during pregnancy.

Objective of the study was to investigate the teratogenic effect of oral promethazine, a phenothiazine derivate in the population-based data set of the Hungarian Case-Control Surveillance of Congenital Abnormalities during the years 1980-1996. Of 22,843 cases with 25 congenital abnormalities, 3648 (16.0%) were born to mothers treated orally with promethazine during pregnancy. Of 38,151 matched population controls without congenital abnormalities, 6025 (15.8%) had mothers with promethazine treatment during the study pregnancy. Of 834 malformed controls (Down syndrome), 142 (17.0%) had mothers w

What is the best approach to reducing birth defects associated with isotretinoin?

Isotretinoin is an effective treatment for severe acne, a condition which can be physically, emotionally, and socially disabling. Because the drug is teratogenic, causing severe birth defects, women taking the drug are directed to avoid pregnancy. In the United States, a series of risk reduction programs have been implemented that aim to prevent pregnant women from taking the drug and to prevent women taking it from getting pregnant. The most recent, and most stringent, is an Internet-based, performance-linked system called iPLEDGE, which tries to ensure that the drug is dispensed only when th

Maternal progestin intake and risk of hypospadias.

BACKGROUND: Previous studies have suggested that maternal intake of progestins during early pregnancy may be associated with an increased risk of hypospadias. Progesterone and its derivatives are commonly prescribed during early pregnancy, for example, in cases of luteal phase dysfunction and in conjunction with ovulation stimulation drugs. OBJECTIVE: To examine whether risk of hypospadias was associated with periconceptional progestin intake. DESIGN AND SETTING: The National Birth Defects Prevention Study, a population-based, multistate, case-control study including deliveries that had esti

[Hysteroplasty for enlargement: apropos of the results].

The DiethylstilBbestrol (DES) is a synthetic estrogen which was prescribed from 1941 onwards for the prevention of miscarriage. As well as a possible risk of cancer, another side effect of this treatment was the possible of abnormality of the genitalia in the female issue of the prescribed user. Apart from possibly having a hypoplasic uterus, the patient is also prone, in the case where she has an undersized uterus, to having a much narrower than normal cavity. Consequently there is a tendency for an excess of muscle tissue on the uterus walls. This can be observed on an RMN. The most signific

Microphthalmia, brachygnathia superior, and palatocheiloschisis in a foal associated with griseofulvin administration to the mare during early pregnancy.

An 18 year old Friesian mare was treated with griseofulvin for dermatomycosis in the second month of pregnancy. Pregnancy was uneventful and after 331 days a male foal was born. The foal showed bilateral microphthalmia, severe brachygnathia superior, and palatocheiloschisis. The lesions were incompatible with life and the animal was euthanized. As similar lesions have been described in other species associated with griseofulvin administration during pregnancy, and the development of the eyes and facial bones in the horse occurs in the second month of pregnancy, the lesions most likely can be a

Teratogenic effects of selenium in natural populations of freshwater fish.

The prevalence of abnormalities and associated tissue selenium residues were assessed for the fish population of Belews Lake, North Carolina, and two reference lakes in 1975, 1978, 1982, and 1992. Teratogenic defects identified included lordosis, kyphosis, scoliosis, and head, mouth, and fin deformities. Many fish exhibited multiple malformations and some were grossly deformed and distorted in appearance. Other abnormalities observed were edema, exophthalmus, and cataracts. Whole-body tissue residues of selenium in the fishes of Belews Lake were up to 130 times those in the reference lakes and

Teratological aspects of dental amalgam.

The teratogenic effect is determined by four factors: (1) the agent, (2) the dose, (3) the stage of embryonic development, and (4) the genetic constitution of the embryo. The first two factors are of particular interest and warrant further comment. It should be emphasized that the mercury released from dental amalgam is mainly metallic mercury vapor. The dose of mercury vapor from dental amalgam fillings in the order of 5 micrograms/day is very low compared with the doses in a teratological study and is not likely to exceed the threshold necessary for a teratogenic effect to occur. The concent
