Quantitative evaluation of radiation-induced heart disease in patients with lung cancer: a three-dimensional speckle tracking imaging study.

BACKGROUND: Adverse cardiovascular events due to radiation-induced heart disease (RIHD) have become the leading cause of death in cancer survivors, and early screening for RIHD has become an important clinical issue. Our objective was to determine the utility of three-dimensional speckle tracking echocardiography (3D-STE) for detecting RIHD. METHODS: According to inclusion and exclusion criteria, patients with lung cancer who received radiotherapy in our hospital for the first time were recruited as subjects. All subjects underwent the conventional echocardiography and 3D-STE examination at s

Ultraviolet A-induced cellular membrane damage in the photosensitivity dermatitis/actinic reticuloid syndrome.

Abnormal photosensitivity of skin in the photosensitivity dermatitis/actinic reticuloid syndrome (PD/AR) correlating to an abnormal photosensitivity of dermal fibroblasts from these patients has been established previously. Cultured human skin fibroblasts from normal infant and adult donors, foreskin tissue and patients with PD/AR were assayed for oxidative stress induced membrane damage following ultraviolet A (UVA) radiation (340-450 nm) and hydrogen peroxide treatment by measurement of lactate dehydrogenase release. Skin fibroblasts from PD/AR patients were, in general, 2- and 4-fold more s

[Radiation-protective agent and anti-aging agents: random and predictable matches.].

Radiation-protective and anti-aging properties are often combined. Combination of this properties is linked to the common mechanisms of action such as direct and indirect antioxidant activities, inhibition of free radicals formation, increase resistance to stress impacts at the cellular level, acceleration of DNA reparation, prevention of chronic diseases linked to abnormalities in regeneration processes, activation of immune inflammatory processes and carcinogenesis. Regulation of cell cycle and apoptosis can often be considered as an implementing driver of radiation-protective and anti-aging

Localized Nonablative Neuromodulatory Irradiation of the Nucleus Accumbens Ameliorates Voluntary Excessive Alcohol Consumption in Miniature Pigs.

OBJECTIVES: We previously reported that low-dose (Dmax < 40 Gy, 7.5-mm collimator) focal irradiation could induce durable localized neuromodulation in the brains of miniature pigs without evidence of tissue damage (Yeh et al, 2021). It remained unclear whether this approach could be used for treating maladaptive behaviors. In this study, we evaluate the therapeutic neuromodulation potential of noninvasive, nonablative, bilateral irradiation of the nucleus accumbens (NAc) in a large-animal model of voluntary excessive alcohol intake. We hypothesize that focal irradiation within the NAc at a dos

Estimation of the hereditary risks of exposure to ionizing radiation: history, current status, and emerging perspectives.

This paper provides a brief overview of the advances in the field of the estimation of the genetic risks of exposure of human populations to ionizing radiation from the early 1950's to the present and of the developments that are anticipated in the coming years. The latter are based on the view that the insights gained from human genetics, especially human molecular genetics, will be increasingly applied to address problems in risk estimation. Owing to the paucity of human data on radiation-induced mutations, mouse data on radiation-induced mutations are used to predict the risk of genetic dis

Targeting SMPDL3B to Ameliorate Radiation- and Cisplatin-Induced Renal Toxicity.

Kidney toxicity remains a major dose-limiting complication of radiation therapy and platinum-based chemotherapy, yet the molecular determinants of renal susceptibility and resilience to these genotoxic treatments are incompletely understood. Podocytes are particularly vulnerable to such insults, and emerging evidence implicates lipid dysregulation in podocyte injury. This study investigated the role of sphingomyelin phosphodiesterase acid-like 3B (SMPDL3B), a podocyte-enriched lipid-modulating enzyme, in radiation- and cisplatin-induced nephrotoxicity. Using a doxycycline-inducible, podocyte-s

Malignancy and cancer treatment-related hair and nail changes.

The hair and nails are complex cutaneous adnexal structures influenced by the patient's physiologic state. Systemic diseases may affect either or both. Remote internal malignancies may induce skin changes that are neither genetically related nor part of a tumor syndrome. The treatment of cancer with chemotherapy and radiation therapy may also induce abnormalities of the hair and nails. In this article, various paraneoplastic and cancer treatment-related hair and nail changes are described. The proper evaluation of such abnormalities may allow for the prompt and efficient diagnosis and manageme

Mechanisms of Disease: molecular genetics of childhood thyroid cancers.

Childhood thyroid cancers are uncommon and have a fairly good prognosis. Papillary adenocarcinoma is the most prevalent malignant tumor of the thyroid in children and adults with radiation-induced or sporadic cancer. The incidence of thyroid cancer in children increased dramatically in the territories affected by the Chernobyl nuclear accident; this increase is probably attributable to (131)I and other short-lived isotopes of iodine released into the environment. There was a broad range of latency periods in children who developed thyroid cancer; some periods were less than 5 years. The mutati

Amphora algae with low-level ionizing radiation exposure ameliorate D-galactosamine-induced inflammatory impairment in rat kidney.

d-Galactosamine (d-GalN) is a well-known toxin that causes many metabolic and morphological abnormalities resulting in advanced renal failure and liver damage. Occupational exposure to low-level ionizing radiation (<1 Gy) was shown to enhance cell protection via attenuating an established inflammatory process. The present study was therefore aimed to investigate the protective impact of Amphora coffaeiformis extract and low dose gamma radiation against d-GalN induced renal damage in rats. Forty-eight adult male Swiss albino rats were distributed equally into eight groups. The measurements incl

Hypersensitivity of Ku80-deficient cell lines and mice to DNA damage: the effects of ionizing radiation on growth, survival, and development.

We recently have shown that mice deficient for the 86-kDa component (Ku80) of the DNA-dependent protein kinase exhibit growth retardation and a profound deficiency in V(D)J (variable, diversity, and joining) recombination. These defects may be related to abnormalities in DNA metabolism that arise from the inability of Ku80 mutant cells to process DNA double-strand breaks. To further characterize the role of Ku80 in DNA double-strand break repair, we have generated embryonic stem cells and pre-B cells and examined their response to ionizing radiation. Ku80(-/-) embryonic stem cells are more sen

Defect of the cerebellar vermis induced by prenatal gamma-ray irradiation in radiosensitive BALB/c mice.

The developing fetal brain is one of the most susceptible organs to irradiation insult. Prenatal irradiation-induced abnormalities in the cerebrum have been well examined in mouse fetuses. However, little information on abnormalities in the cerebellum caused by irradiation is available. Moreover, few reports have examined the chronological changes of the brain from the prenatal to the postnatal period. To analyze the chronological changes induced by irradiation, we exposed pregnant mice to gamma-ray irradiation on embryonic day 13.5 (E13.5) and investigated the histopathology of the cerebellum

Radiation-induced lung injury: a hypersensitivity pneumonitis?

Radiation pneumonitis occurs 6 to 12 weeks after thoracic irradiation, and is thought to be due to direct radiation-induced lung injury. Four patients who developed pneumonitis after unilateral thoracic irradiation for carcinoma of the breast were studied with bronchoalveolar lavage, gallium scan of the lung, and respiratory function tests. On the irradiated side of the chest, all four patients showed an increase in total cells recovered from the lavage fluid and a marked increase in the percentage of lymphocytes. When results for the unirradiated lung were compared with results for the irradi

Magnetic resonance imaging in childhood leukemia survivors treated with cranial radiotherapy: a cross sectional, single center study.

BACKGROUND: Children treated with cranial radiotherapy (CRT) for leukemia are at risk of developing central nervous system injuries. Magnetic resonance imaging (MRI) represents the examination method of choice for evaluating radiation-induced brain complications. The purpose of this report is to describe the spectrum of MRI abnormalities detected in a group of survivors of leukemia treated with cranial irradiation. PROCEDURES: In this cross-sectional, single center study, 56 patients (median age at follow-up 19 years) receiving CRT as cranial prophylaxis (CP) included in the leukemia protocol

Effects of different doses of gamma-radiation on the developing brain of mice.

Morphogenetic changes in the developing brain induced by doses of 60Co gamma-irradiation ranging from 0 to 1.5 Gy on day 13 of pregnancy (E13) were studied in 6-week-old mice. Dose-related reductions in brain weight and cortical thickness (field 3 of Caviness) were significant for all irradiated groups, but abnormal cortical architecture was evident only in mice exposed to 1.0 and 1.5 Gy. Ectopic gray matter, enlarged lateral ventricles, and the absence of trunks from the corpus callosum were observed respectively in 23%, 30% and 10% of the mice exposed to 1.0 Gy, but these anomalies rose to 9

Malformations after radiation exposure of preimplantation stages.

Our studies have shown that, contrary to the opinion in most textbooks, it is possible to increase the number of malformed fetuses in one of our mouse strains (originally "Heiligenberger Stamm", meanwhile HLG/Zte) by radiation exposure of zygotes or of subsequent preimplantation stages. The malformation affected most pronouncedly is gastroschisis, a defect occurring at a frequency of 1 to 4% in the controls. The observed increase is strain specific (C57Bl mice or (HLGxC57Bl)F1 hybrids do not react in the same way), it is accompanied by an increased frequency of chromosomal aberrations in skin

Mechanistic insights into microwave radiation induced cognitive impairments: The role of m6A epigenetic modifications and HNRNPA2B1 in TrkB regulation.

Microwave radiation, a prevalent environmental stressor, significantly impacts human health. Based on previous studies, we hypothesize that microwave-induced cognitive impairments and vulnerability in the hippocampal dentate gyrus (DG) region are due to abnormal synaptic plasticity regulated by both newborn and mature neurons derived from neural stem cells (NSCs). Epigenetics links external factors to organisms, offers insights into the health effects of environmental influences. To explore the molecular mechanisms underlying the effects of microwave radiation on neuronal synaptic plasticity f

GM1 ganglioside treatment protects against long-term neurotoxic effects of neonatal X-irradiation on cerebellar cortex cytoarchitecture and motor function.

Exposure of neonatal rats to a 5 Gy dose of X-irradiation induces permanent abnormalities in cerebellar cortex cytoarchitecture (disarrangement of Purkinje cells, reduction of thickness of granular cortex) and neurochemistry (late increase in noradrenaline levels), and motor function (ataxic gait). The neuroprotective effects of gangliosides have been demonstrated using a variety of CNS injuries, including mechanical, electrolytic, neurotoxic, ischemic, and surgical lesions. Here, we evaluated whether systemically administered GM1 ganglioside protects against the long-term CNS abnormalities in

The morphologic changes induced by hormone and radiation therapy on prostate carcinoma.

The morphologic changes induced by hormone and radiation therapy were evaluated in prostate biopsy and prostatectomy specimens from patients with residual prostate carcinoma. The two therapeutic methods induce changes both in the nonmalignant and malignant residual prostatic tissue. Following hormone therapy, the nonmalignant prostatic tissue showed atrophy of prostatic acini associated with fibrosis, basal cell hyperplasia, degenerative changes of the secretory epithelial cells, and a marked decrease of high-grade intraepithelial neoplasia (HGPIN). In the fragments of residual carcinoma, squa
