Changes in the levels of headspace volatiles, including acetaldehyde and formaldehyde, in red and white wine following light irradiation.

The effects of fluorescence light irradiation on the changes in the levels of volatiles, especially acetaldehyde and formaldehyde, were determined in red and white wines. Three different red or white wine brands were mixed and subjected to light irradiation for 5 days. Generally, the levels of total volatiles in white wine were higher than those in red wine were and decreased during light irradiation. The level of 1,1,6-trimethyl-1,2-dihydronaphthalene, an aromatic compound commonly found in aging wine, decreased significantly following light irradiation (p < 0.05), whereas those of acetaldehy…

Evaluation of the effect of ethanol's toxic metabolite acetaldehyde on the gastrointestinal oligopeptide transporter, PEPT1: in vitro and in vivo studies.

BACKGROUND: The effects of alcohol consumption and its subsequent metabolism on drug transport, absorption and pharmacokinetics are poorly understood. This study examines the effects of the ethanol metabolite, acetaldehyde, on the clinically relevant drug transporter, PEPT1. The metabolism of ethanol and the following acetaldehyde formation is thought to modulate the uptake capacity of PEPT1 within the gastrointestinal tract for a variety of clinically important peptidomimetic drug compounds. METHODS: Glycylsarcosine ([(3)H]-GlySar), a nonhydrolysable PEPT1 specific substrate was used in our …

Concentration-time profiles of ethanol and acetaldehyde in human volunteers treated with the alcohol-sensitizing drug, calcium carbimide.

1. The disposition kinetics of ethanol and its toxic metabolite acetaldehyde were investigated in 10 healthy male volunteers who ingested 0.25 g kg-1 ethanol after an overnight fast. This dose of ethanol was given 2 h after they swallowed a tablet of either calcium carbimide CC (50 mg), a potent inhibitor of low Km aldehyde dehydrogenase (ALDH), or placebo according to a single-blind crossover design. 2. The pulmonary blood concentrations of ethanol and acetaldehyde were estimated indirectly by means of a gas chromatographic method modified for analysis of end-expired breath. This non-invasive…

Determination of the proximate teratogen of the mouse fetal alcohol syndrome. 2. Pharmacokinetics of the placental transfer of ethanol and acetaldehyde.

CD-1 mice were treated ip on Day 10 of gestation with 4, 6, or 7 g/kg ethanol. Maternal and embryonic tissues were analyzed for ethanol and acetaldehyde levels by head-space gas chromatography 5 min to 24 hr after treatment. Dose-dependent ethanol concentrations were observed in maternal blood and liver. Ethanol rapidly crossed the placenta and appeared in the embryo 5 min after treatment. Acetaldehyde was detectable in maternal blood following all treatments and in maternal liver and embryos following treatment with 7 g/kg ethanol. Coadministration of 100 mg/kg 4-methylpyrazole, an alcohol de…

Long-term exposure to the ethanol-derived metabolite acetaldehyde elevates structural genomic alterations but not base substitutions.

Acetaldehyde is the primary metabolite of ethanol, and routes of exposure include endogenous sources, food and cigarette smoke. To explore whether the mutagenic effect of acetaldehyde is responsible for the carcinogenicity of ethanol, we use whole genome sequencing on four human cell lines subjected to long-term, physiologically relevant, analytically validated acetaldehyde treatments. Unexpectedly, the treatments do not induce increased base substitution and short insertion/deletion mutagenesis, nor the appearance of the alcohol-associated cancer mutation signature SBS16. In contrast, we obse…

Real-time sensing of acetaldehyde in the headspace of yeast cultures using mid-infrared laser absorption spectroscopy.

Mid-infrared laser absorption spectroscopy measurements of acetaldehyde in the headspace of yeast cultures are reported. An interband cascade laser (ICL) with single-mode tuning between 1770.19 and 1770.35 cm-1 was used to excite specific rotational-vibrational modes of motion in the R-branch of the carbonyl stretch absorption band for acetaldehyde. Real-time acetaldehyde measurements of seven different yeast cell cultures showed the effects of sucrose feeding, malted barley extract feeding, and anaerobic culturing. Acetaldehyde concentrations were as high as 62.8 ppm, observed 3 h after feedi…

An Inverse Electron-Demand Diels-Alder Approach to Selective Activity-Based Sensing of Acetaldehyde in Living Cells.

Acetaldehyde (AA) is a reactive aldehyde primarily produced in cells as a metabolic intermediate during ethanol oxidation. Excess AA, often resulting from impaired AA detoxification, leads to aberrant DNA, protein, and/or lipid damage and increases risk of diseases such as cancer, hepatitis, and cirrhosis. Traditional methods for detecting biological AA often require sample destruction or extensive processing, which compromise spatiotemporal resolution, or do not exhibit sufficient selectivity for this two-carbon metabolite over other competing aldehydes and reactive carbon species in living s…

Associations between periodontitis and serum anti-malondialdehyde-acetaldehyde antibody concentrations in rheumatoid arthritis: A case-control study.

BACKGROUND: Malondialdehyde-acetaldehyde (MAA) adducts lead to generation of anti-MAA autoantibodies and have been independently identified in inflamed periodontal and rheumatoid arthritis (RA) tissues. This study evaluates serum samples from RA cases and osteoarthritis (OA) controls to quantify associations between periodontal clinical measures, alveolar bone loss (ABL), and anti-Porphyromonas gingivalis, anti-Prevotella intermedia, and anti-Fusobacterium nucleatum antibody concentrations with anti-MAA antibody concentrations. METHODS: Participants (n = 284 RA cases, n = 330 OA controls) und…

Effective practical removal of acetaldehyde by a sandwich-type plasma-in-honeycomb reactor under surrounding ambient conditions.

The effective removal of acetaldehyde by humidified air plasma was investigated with a high throughput of contaminated gas in a sandwiched honeycomb catalyst reactor at surrounding ambient temperature. Here, acetaldehyde at the level of a few ppm was successfully oxidized by the honeycomb plasma discharge despite the harsh condition of large water content in the feed gas. The conversion rate of acetaldehyde increased significantly with the presence of catalysts coating on the surface channels. The increased conversion rate was also obtained with a high specific energy input (SEI) and total flo…

Effects of "Essential AD2" Supplement on Blood Acetaldehyde Levels in Individuals Who Have Aldehyde Dehydrogenase (ALDH2) Deficiency.

BACKGROUND: It is estimated that 1 billion people in the world have a point mutation in the gene encoding the aldehyde dehydrogenase 2 (ALDH2) enzyme, the primary enzyme responsible for the metabolism of acetaldehyde. The presence of this mutation is called ALDH2 deficiency. Because of limited ability to metabolize acetaldehyde, individuals with ALDH2 deficiency experience elevated levels of blood acetaldehyde after exposure to various common sources such as recreational alcohol. Because of higher levels of acetaldehyde, individuals with ALDH2 deficiency are at higher risk for numerous disease…

Highly enantioselective cross-aldol reactions of acetaldehyde mediated by a dual catalytic system operating under site isolation.

Polystyrene-supported (PS) diarylprolinol catalysts 1 a (Ar = phenyl) and 1 b (Ar = 3,5-bis(trifluoromethyl)phenyl) have been developed. Operating under site-isolation conditions, PS-1 a/1 b worked compatibly with PS-bound sulfonic acid catalyst 2 to promote deoligomerization of paraldehyde and subsequent cross-aldol reactions of the resulting acetaldehyde in one pot, affording aldol products in high yields with excellent enantioselectivities. The effect of water on the performance of the catalytic system has been studied and its optimal amount (0.5 equiv) has been determined. The dual catalyt…

Early response of alpha2(I) collagen to acetaldehyde in human hepatic stellate cells is TGF-beta independent.

Acetaldehyde is fibrogenic and induces the expression of type I collagen genes in hepatic stellate cells. Some of these acetaldehyde-dependent events are mediated by H(2)O(2) and thus establish a direct connection between oxidative stress and collagen upregulation. We localized to the -378 to -183 region of the alpha2(I) collagen (COL1A2) promoter an acetaldehyde-responsive element (AcRE) functional in human hepatic stellate cells (HHSCs) and investigated molecular mechanisms whereby acetaldehyde stimulates and modulates its transcriptional activity. Because the AcRE co-localized with a previo…

Prevention of HNSCC by chewing gum delivery of enzymes/antimicrobials to reduce carcinogenic acetaldehyde produced by pathogenic microbes.

Acetaldehyde, a WHO Group 1 human carcinogen, produced by oral microbes, plays a critical role at the onset of head and neck squamous cell carcinoma (HNSCC). Candida albicans isolated from different HNSCC patient saliva samples produced similar levels of acetaldehyde; the production level increased in a dose-dependent manner, with the highest level observed at 100 mM glucose or ethanol. Streptococci produced acetaldehyde in ethanol-free media, but the level increased up to 500-1,000 mM alcohol, whereas Fusobacterium nucleatum and Porphyromonas gingivalis showed ethanol-induced acetaldehyde pro…

A novel quantification method for sulfur-containing biomarkers of formaldehyde and acetaldehyde exposure in human urine and plasma samples.

A novel method for the quantification of the sulfur-containing metabolites of formaldehyde (thiazolidine carboxylic acid (TCA) and thiazolidine carbonyl glycine (TCG)) and acetaldehyde (methyl thiazolidine carboxylic acid (MTCA) and methyl thiazolidine carbonyl glycine (MTCG)) was developed and validated for human urine and plasma samples. Targeting the sulfur-containing metabolites of formaldehyde and acetaldehyde in contrast to the commonly used biomarkers formate and acetate overcomes the high intra- and inter-individual variance. Due to their involvement in various endogenous processes, fo…

Effect of Alcohol Sensitivity in Healthy Young Adults on Breath Pharmacokinetics of Acetaldehyde After Mouth Washing with Alcohol.

BACKGROUND: Acetaldehyde is causally related to head and neck cancer. Individuals with aldehyde dehydrogenase 2 deficiency experience alcohol sensitivity and are referred to as "flushers" because of their skin-flushing response to high blood acetaldehyde levels after alcohol consumption. Acetaldehyde is produced in the oral cavity after local alcohol exposure without alcohol ingestion. However, the relationship between the oral acetaldehyde level after local alcohol exposure and alcohol sensitivity is unclear. Herein, sampling the exhaled breath, we evaluated the effect of alcohol sensitivity …

Ethanol-induced conditioned place preference, but not aversion, is blocked by treatment with D -penicillamine, an inactivation agent for acetaldehyde.

RATIONALE: There is evidence to suggest that acetaldehyde is involved in the control of ethanol-seeking behavior and reward. D -penicillamine, a thiol amino acid, is a highly selective agent for the inactivation of acetaldehyde. Previous studies from our laboratory have demonstrated that D -penicillamine prevents both behavioral stimulation induced by ethanol and acetaldehyde-produced locomotor depression in mice. OBJECTIVES: The contribution of ethanol-derived acetaldehyde to the affective effects of ethanol (preference and aversion) was assessed using an unbiased place conditioning design. …

Metabolism of acetaldehyde to methyl and acetyl radicals: in vitro and in vivo electron paramagnetic resonance spin-trapping studies.

Acetaldehyde oxidation by enzymes and cellular fractions has been previously shown to produce radicals that have been characterized as superoxide anion, hydroxyl, and acetyl radicals. Here, we report that acetaldehyde metabolism by xanthine oxidase, submitochondrial particles and whole rats produces both the acetyl and the methyl radical, although only the latter was unambiguously identified in vivo. Electron paramagnetic resonance (EPR) characterization of both radicals was possible by the use of two spin traps, 5,5-dimethyl 1-pyrroline N-oxide (DMPO) and alpha-(4-pyridyl 1-oxide)-N-t-butylni…

Yeast cells with a specific cellular make-up and an environment that removes acetaldehyde are prone to sustained glycolytic oscillations.

Glycolytic oscillations can be induced by adding glucose to starved Saccharomyces cerevisiae cells and, after a steady state has been establ … red activities remained constant. Cyanide was found not only to block respiration, but also to trap acetaldehyde. Both cyanide actions appear necessary for the occurrence of sustained glycolytic oscillations.

Effects of high hepatic acetaldehyde level following simultaneous administration of ethanol and cyanamide on liver function in rats.

Extremely high concentrations of hepatic acetaldehyde were induced in rats by the intragastric administration of ethanol and cyanamide, an aldehyde dehydrogenase inhibitor; and these high levels were maintained for 4 weeks. Liver function tests, including mitochondrial ornithine carbamoyltransferase (OCT) and GOT activities, were within normal limits, and no increase in either hepatic triglyceride or collagen contents was observed. These results suggest that hepatotoxic effects of ethanol are not derived from the high acetaldehyde levels in the liver.
