Ovicidal effects of albendazole in human ascariasis, ancylostomiasis and trichuriasis.

Albendazole, a broad spectrum anthelmintic, was administered as a 400 mg single dose to 20 patients harbouring Ascaris (ten cases), hookworms (four cases each of Ancylostoma duodenale and Necator americanus) and trichuriasis (ten cases). Faeces were obtained before treatment and during the following five days. Coprocultures were made for 90 days for Ascaris and Trichuris eggs, hookworm eggs were cultured by the Harada-Mori technique for at least eight days. Albendazole was ovicidal against all four genera of nematodes.

Efficacy of mebendazole and albendazole against Trichinella spiralis in mice.

Changes in the sensitivity of Trichinella spiralis to anthelmintic treatment during the first 3 days of infection in mice were studied. Oral administration of either mebendazole or albendazole at 6.25 mh/kg 2 hr after exposure to infection eliminated 95-100% of the worms as determined at necropsy on day 7 postinoculation. Beyond the first day of infection the sensitivity of the parasite to benzimidazole therapy was much reduced and an oral dose of 50 mg/kg was only partially but significantly active against the adult worms. Despite decline in drug sensitivity during the enteral phase, gavage a…

Ultrastructural changes in the advanced third larval stage of Gnathostoma binucleatum Almeyda-Artigas, 1991 (Nematoda: Spirurida) following albendazole and albendazole sulfoxide in vitro exposure.

We report here the ultrastructural changes in the third advanced larval stage (AL3) of Gnathostoma binucleatum Almeyda-Artigas, 1991, induced by in vitro treatment with albendazole and the metabolite albendazole sulfoxide. During the whole period of the experiment, the controls remained active and vital throughout the 108-h exposure period. The primary site of action of both drugs appears to be the mitochondria and myofibrils of the polymyarian musculature. Degenerative changes in the secretory portion of the esophagus and somatic musculature were clearly observed. These degenerative changes, …

Poor Response to Albendazole in Hookworm-Infected Abnormal Haemoglobin Carriers.

BACKGROUND: In southern Thailand, the low efficacy of albendazole contributes to the unsuccessful elimination of hookworms. Individuals infected with hookworms exhibiting low albendazole efficacy show decreased mean corpuscular volume and mean corpuscular haemoglobin and are suspected carriers of abnormal haemoglobin. This observational study aimed to compare the treatment outcomes of albendazole for hookworm infection in individuals with normal versus abnormal haemoglobin types. METHODS: 227 stool samples were examined using the Kato-Katz and agar plate culture methods. Blood samples were co…

Reduced efficacy of albendazole against Ascaris lumbricoides in Rwandan schoolchildren.

Control of human soil-transmitted helminths (STHs) relies on preventive chemotherapy of schoolchildren applying the benzimidazoles (BZ) albendazole or mebendazole. Anthelmintic resistance (AR) is a common problem in nematodes of veterinary importance but for human STHs, information on drug efficacy is limited and routine monitoring is rarely implemented. Herein, the efficacy of single dose albendazole (400 mg) was evaluated in 12 schools in the Huye district of Rwanda where Ascaris is the predominant STH. Ascaris eggs were detected by wet mount microscopy and the Mini-FLOTAC method to assess c…

Liver hydatid disease: morphological changes of protoscoleces after albendazole therapy.

BACKGROUND: Postoperative recurrence of the liver hydatid disease befalls approximately 10-30% of patients. Preoperative or postoperative therapy with albendazole in single therapeutic protocol (800 mg/d, within 28 days) indicated the need to evaluate the hydatid cyst liquid protoscoleces viability. Morphological changes of protoscoleces following the treatment with drugs are not well known. AIM: To estimate the viability of protoscoleces after preoperative or postoperative albendazole therapy, and their ability for cystic metamorphosis. METHODS: A prospective, randomized clinical trial incl…

Separation of albendazole sulfoxide enantiomers by chiral supercritical-fluid chromatography.

The enantioseparation of albendazole sulfoxide (ABZSO) by chiral supercritical-fluid chromatography (SFC) on two columns, based on the polysaccharide derivatives Chiralpak AD and Chiralcel OD, was studied. The effect of different modifiers, methanol, ethanol, 2-propanol, and acetonitrile, was examined. The results showed that ABZSO can be separated on both columns, using an alcohol-type modifier. Using the Chiralpak AD column, the best results were obtained with 2-propanol and, in the case of the Chiralcel OD, with methanol.
