Report of natural Mayaro virus infection in Mansonia humeralis (Dyar & Knab, Diptera: Culicidae).

BACKGROUND: Arboviruses are a group of viruses transmitted to vertebrate hosts by certain blood-feeding arthropods. Among urban vectors of arboviruses, mosquitoes of the genus Aedes are the most common. However, other mosquitoes may be susceptible to infection and involved in the transmission, such as Mansonia spp. Therefore, this study aimed to investigate whether Mansonia humeralis can be infected with the Mayaro virus (MAYV). METHODS: These insects were collected from 2018 to 2020 in chicken coops of rural communities in Jaci Paraná in Porto Velho, Rondônia, Brazil, while performing blood-

Antibody cross-reactivity among alphaviruses in clinical practice.

Alphaviruses are mosquito-borne pathogens causing rheumatological disease and encephalomyelitis. Their spread, enhanced by climate change, presents a growing global health threat. To date, no specific treatments and, except for chikungunya virus, no preventive vaccines exist. Alphavirus infection induces cross-reactive antibodies targeting conserved epitopes in glycoprotein E1 and E2. While cross-reactivity can facilitate the development of broad-spectrum vaccines, it complicates serological diagnosis in regions with multiple co-circulating alphaviruses. This review examines how antibody cross…

Imported arbovirus infections in Canada 1974-89.

From 1974 to 1989, sera from symptomatic patients with histories of recent travel outside Canada were tested for antibodies to several arboviruses, principally of the alphavirus and flavivirus families. Diagnostic seroconversions were documented in 84 individuals from six provinces, including one alphavirus (Chikungunya) and 83 flavivirus seroconvertors. Dengue 1 virus was isolated from the blood of one patient. Most flavivirus seroconvertors were likely infected with dengue virus, but infections with tick-borne encephalitis, St Louis encephalitis and Powassan viruses were also recognized. Pat…

The residue 86 of the Getah virus E2 glycoprotein mediates both glycosaminoglycan- and LDLR-dependent infection.

Getah virus (GETV), a mosquito-borne alphavirus, poses an emerging threat to public health with its increasingly broad host spectrum. While glycosaminoglycans (GAGs) serve as critical attachment factors for many alphaviruses and the low-density lipoprotein receptor (LDLR) facilitates the cellular entry of several members, the precise viral determinants governing these interactions and their implications for viral virulence remain poorly defined. Here, we introduced an H86Y substitution, a potential adaptive mutation site, within the E2 glycoprotein of GETV using reverse genetics. The H86Y muta…

Vector competence of temperate and tropical mosquito species for Babanki virus.

Babanki virus (BBKV), an alphavirus belonging to the Western Equine Encephalitis antigenic complex, was first isolated from Mansonia africana mosquitoes in 1969 but remains poorly characterized. Transmitted to humans and animals through the bite of an infectious female mosquito, it is associated with febrile illness, rash, and arthritis, similarly to its close relative Sindbis virus. No specific treatment or vaccine is currently available. Here, we experimentally assessed the vector competence of mosquito species from tropical and temperate regions under ecologically relevant temperature condi…

VLDLR and ApoER2 are receptors for multiple alphaviruses.

Alphaviruses, like many other arthropod-borne viruses, infect vertebrate species and insect vectors separated by hundreds of millions of years of evolutionary history. Entry into evolutionarily divergent host cells can be accomplished by recognition of different cellular receptors in different species, or by binding to receptors that are highly conserved across species. Although multiple alphavirus receptors have been described1-3, most are not shared among vertebrate and invertebrate hosts. Here we identify the very low-density lipoprotein receptor (VLDLR) as a receptor for the prototypic alp…

The Old World and New World alphaviruses use different virus-specific proteins for induction of transcriptional shutoff.

Alphaviruses are widely distributed throughout the world. During the last few thousand years, the New World viruses, including Venezuelan equine encephalitis virus (VEEV) and eastern equine encephalitis virus (EEEV), evolved separately from those of the Old World, i.e., Sindbis virus (SINV) and Semliki Forest virus (SFV). Nevertheless, the results of our study indicate that both groups have developed the same characteristic: their replication efficiently interferes with cellular transcription and the cell response to virus replication. Transcriptional shutoff caused by at least two of the Old …

Western equine encephalitis virus is a recombinant virus.

The alphaviruses are a group of 26 mosquito-borne viruses that cause a variety of human diseases. Many of the New World alphaviruses cause encephalitis, whereas the Old World viruses more typically cause fever, rash, and arthralgia. The genome is a single-stranded nonsegmented RNA molecule of + polarity; it is about 11,700 nucleotides in length. Several alphavirus genomes have been sequenced in whole or in part, and these sequences demonstrate that alpha-viruses have descended from a common ancestor by divergent evolution. We have now obtained the sequence of the 3'-terminal 4288 nucleotides o…

Contrasting Ecological Patterns of Venezuelan Equine Encephalitis and Madariaga Viruses in Small Mammal and Mosquito Populations from Two Enzootic Regions of Panama.

Background: Understanding enzootic cycles is key to arbovirus control. Rodents are primary reservoirs of Venezuelan equine encephalitis virus (VEEV), whereas the vertebrate host of Madariaga virus (MADV) remains unknown. Culex (Melanoconion) mosquitoes are considered primary vectors. We studied small mammals and mosquito ecology in the Darién province and Panamá Oeste province, two enzootic regions of Panama. Additional alphavirus seroprevalence was assessed in small mammals. Methods: Small mammals were trapped using Sherman and Tomahawk traps, and blood samples were tested for MADV and VEEV n…

Mayaro virus infection elicits a robust pro-inflammatory and antiviral response in human macrophages.

Mayaro virus (MAYV), the etiological agent of Mayaro fever (MAYF), is an emergent arbovirus pathogen belonging to Togaviridae family. MAYF is characterized by high inflammatory component that can cause long-lasting arthralgia that persists for months. Macrophages are viral targets and reservoirs, key components of innate immunity and host response. Given the importance of this pathogen, our aim was to determine the inflammatory and antiviral response of human monocyte-derived macrophages (MDMs) infected with MAYV. First, we established the replication kinetics of the virus. Thereafter, we dete

Prolonged polyarthralgia in a German traveller with Mayaro virus infection without inflammatory correlates.

BACKGROUND: Mayaro virus is endemic in South America and sporadic outbreaks have been described. It causes a dengue-like febrile illness accompanied by severe and long-lasting polyarthralgias. Outside endemic regions, however, the disease is not well known and can be misdiagnosed as dengue. International travellers are at risk to acquire Mayaro virus and due to increased worldwide travel infectious disease specialists need to be aware of such rare clinical entities. CASE PRESENTATION: We report the first Mayaro virus infection imported into Germany. A 20-year-old woman developed fever, myalgi

Characterization of an early-stage fusion intermediate of Sindbis virus using cryoelectron microscopy.

The sequential steps in the alphavirus membrane fusion pathway have been postulated based on the prefusion and postfusion crystal structures of the viral fusion protein E1 in conjunction with biochemical studies. However, the molecular structures of the hypothesized fusion intermediates have remained obscure due to difficulties inherent in the dynamic nature of the process. We developed an experimental system that uses liposomes as the target membrane to capture Sindbis virus, a prototypical alphavirus, in its membrane-binding form at pH 6.4. Cryoelectron micrograph analyses and 3D reconstruct…

Structural proteins of Western equine encephalitis virus: amino acid compositions and N-terminal sequences.

The structural proteins of Western equine encephalitis virus, a member of the alphavirus group, have been characterized by the determination of their amino acid compositions and by N-terminal sequence analysis. More than 60 residues of the N-terminal sequences of each of the envelope glycoproteins have been determined. A comparison of these sequences with the previously determined sequences of two related alphaviruses. Sindbis virus and Semliki Forest virus, strongly supports the view that all three viruses have evolved from a common ancestor and provides information on the pattern of this evo…

Insulin reduces the transmission potential of chikungunya virus and activates the toll pathway in Aedes aegypti mosquitoes.

Chikungunya virus (CHIKV) is an alphavirus that has re-emerged globally over the last two decades and has the potential to become endemic in the United States due to the presence of competent mosquito vectors, Aedes aegypti and Aedes albopictus. CHIK disease is characterised by fever, rash, and joint pain, and causes chronic debilitating joint pain and swelling in >50% of infected individuals. Given the disease severity caused by CHIKV and the global presence of vectors to facilitate its spread, strategies to reduce viral transmission are desperately needed; however, the human biological facto…

Attenuated Getah virus confers protection against multiple arthritogenic alphaviruses.

Alphaviruses are important arthropod-transmitted pathogens of humans and livestock. Getah virus (GETV) is an arthritogenic alphavirus that causes disease in horses and piglets; it also poses a potential threat to humans. A live attenuated vaccine candidate named GETV-3ΔS2-CM1, harbouring a deletion in nonstructural protein 3 and substitutions in the capsid protein, is genetically stable and exhibits robust immunogenicity. It was shown to confer passive protection to piglets born to immunized sows. In mice, a single dose of GETV-3ΔS2-CM1 protected against infection with different strains of GET…

Weather extremes associated with increased Ross River virus and Barmah Forest virus notifications in NSW: learnings for public health response.

OBJECTIVE: To examine the sequence of environmental and entomological events prior to a substantial increase in Ross River virus (RRV) and Barmah Forest virus (BFV) notifications with a view to informing future public health response. METHODS: Rainfall, tidal, mosquito and human arboviral notification data were analysed to determine the temporality of events. RESULTS: Following two extremely dry years, there was a substantial increase in the abundance of mosquitoes along coastal New South Wales (NSW) two weeks after a significant rainfall event and high tides in February 2020. Subsequently,

Venezuelan equine encephalitis virus disrupts STAT1 signaling by distinct mechanisms independent of host shutoff.

Venezuelan equine encephalitis virus (VEEV) is an important human and veterinary pathogen causing sporadic epizootic outbreaks of potentially fatal encephalitis. The type I interferon (IFN) system plays a central role in controlling VEEV and other alphavirus infections, and IFN evasion is likely an important determinant of whether these viruses disseminate and cause disease within their hosts. Alphaviruses are thought to limit the induction of type I IFNs and IFN-stimulated genes by shutting off host cell macromolecular synthesis, which in the case of VEEV is partially mediated by the viral ca…

Exploring Options for Syndromic Surveillance in Aquaculture: Outbreak Detection of Salmon Pancreas Disease Using Production Data from Norwegian Farms.

Syndromic surveillance (SyS) is an important tool for early warning and monitoring of health in human and animal populations, but its use in aquaculture has been limited. Our study objective was to design a SyS system for Atlantic salmon aquaculture and to evaluate its performance in detecting pancreas disease (PD) outbreaks caused by salmonid alphaviruses on farms. We defined SyS outbreak alarms as cases where monthly farm mortality exceeded predefined cutoffs or deviated significantly from expected values based on predictive generalized linear models. These models were trained for each salmo…

Infectivity of three Mayaro Virus geographic isolates in human cell lines.

Mayaro virus (MAYV) is an emergent arthropod-borne virus that causes an acute febrile illness accompanied by arthralgia, similar to chikungunya virus. Increasing urbanization of MAYV outbreaks in the Americas has led to concerns for geographic expansion and spillover. Given the potential importance of this pathogen, we sought to fill critical gaps in knowledge regarding MAYV infectivity and geographic variation. This study describes the cytopathogenicity of MAYV in human dermal fibroblasts, human skeletal muscle satellite cells, human embryonic kidney cells (HEK), peripherally derived human ma
