PARP7-mediated mono-ADP-ribosylation stabilizes MYH9 to ensure actin cap integrity and chromosome segregation in mouse oocytes.

INTRODUCTION: Oocyte meiotic maturation requires highly asymmetric cell division, governed by spindle migration and actin cap formation. However, the upstream mechanisms that regulate the precise coordination of these events remain unclear. OBJECTIVES: This study aimed to elucidate the role of PARP7, a mono-ADP-ribosyltransferase, in regulating actin cytoskeletal dynamics and chromosomal stability during mouse oocyte meiosis. METHODS: Metabolomic profiling was performed to assess NAD+ dynamics during meiosis. PARP7 expression and localisation were analysed using genetic knockdown and pharmac

Thyme Oil Alleviates Cadmium-Induced Disturbances in Mitotic Activity, Cytoskeletal Organization and H3T3/H3S10 Phosphorylation in Vicia faba.

Cadmium (Cd) contamination, through induction of oxidative stress, severely impairs plant growth. Using primary roots of Vicia faba, we investigated how a 24 h incubation in CdCl2 solution (175 µM) affects mitotic progression in meristems and assessed whether thyme essential oil (TO; 0.03%, v/v), as a natural antioxidant, can protect proliferating cells during simultaneous Cd exposure. Cd strongly inhibited root growth, reduced mitotic index tenfold (to 0.6%), induced chromatin condensation, decreased CDKA protein levels and CycB transcripts and proteins, caused pronounced microtubule bundling

Higher frequency of homologous chromosome pairing in human adult aortic endothelial cells.

During mitosis, pairing of homologous chromosomes can be detrimental and has been correlated with gene misregulation, chromosomal aberrations, and various pathological diseases. We previously demonstrated that homologous chromosomes are spatially segregated, or antipaired, in neonatal human endothelial cells at metaphase/anaphase, which may help prevent abnormal recombination. However, it is unclear if this antipairing persists in adult endothelial cells. To test whether the antipairing, or one homolog per nuclear hemisphere motif, is conserved in adult endothelial cells, we examined human aor

Proteomic analysis of wanxi white goose testicles in different reproductive stages by data-independent acquisition (DIA) strategy.

Wanxi white goose is an important male parent in crossbreeding of Chinese geese, but its short reproductive cycle restricts its application in Northeast China. Therefore, understanding the potential mechanism of breeding period regulation in Wanxi white goose will help to provide more options for crossbreeding. In this study, the reproductive period was divided into prophase (T1), metaphase (T2) and anaphase (T3) according to the laying rhythm of geese. Based on this, testicular tissues were stained and further analyzed by DIA proteomics. A total of 69577 Unique spectra, 70325 Unique peptides

Elucidation of the molecular mechanism of the breakage-fusion-bridge (BFB) cycle using a CRISPR-dCas9 cellular model.

Chromosome instability (CIN) is frequently observed in many tumors. The breakage-fusion-bridge (BFB) cycle has been proposed to be one of the main drivers of CIN during tumorigenesis and tumor evolution. However, the detailed mechanisms for the individual steps of the BFB cycle warrants further investigation. Here, we demonstrated that a nuclease-dead Cas9 (dCas9) coupled with a telomere-specific single-guide RNA (sgTelo) can be used to model the BFB cycle. First, we showed that targeting dCas9 to telomeres using sgTelo impeded DNA replication at telomeres and induced a pronounced increase of

Age-dependent loss of cohesion protection in human oocytes.

Aneuploid human eggs (oocytes) are a major cause of infertility, miscarriage, and chromosomal disorders. Such aneuploidies increase greatly as women age, with defective linkages between sister chromatids (cohesion) in meiosis as a common cause. We found that loss of a specific pool of the cohesin protector protein, shugoshin 2 (SGO2), may contribute to this phenomenon. Our data indicate that SGO2 preserves sister chromatid cohesion in meiosis by protecting a "cohesin bridge" between sister chromatids. In human oocytes, SGO2 localizes to both sub-centromere cups and the pericentromeric bridge,

Molecular characterisation of a mosaicism with a complex chromosome rearrangement: evidence for coincident chromosome healing by telomere capture and neo-telomere formation.

BACKGROUND: Broken chromosomes must acquire new telomeric "caps" to be structurally stable. Chromosome healing can be mediated either by telomerase through neo-telomere synthesis or by telomere capture. AIM: To unravel the mechanism(s) generating complex chromosomal mosaicisms and healing broken chromosomes. METHODS: G banding, array comparative genomic hybridization (aCGH), fluorescence in-situ hybridisation (FISH) and short tandem repeat analysis (STR) was performed on a girl presenting with mental retardation, facial dysmorphism, urogenital malformations and limb anomalies carrying a comp

CDK2 Inhibition and Eradication of Aneuploid Cancers: Promise Meets Resistance.

Aneuploidy is a cancer hallmark that causes resistance to anticancer drugs and promotes aggressive tumors. Thus, aneuploidy is a consequential feature of human malignancy. This review addresses how cyclin-dependent kinase 2 (CDK2) inhibition targets a broad array of aneuploid cancers for proapoptotic death by engaging a death program called anaphase catastrophe. This program eliminates aneuploid cancers while sparing non-aneuploid epithelial cells, thereby providing a favorable therapeutic window. Despite CDK2 inhibition, a residual population of polyploid cancer cells persists in both in vitr

Fangchinoline inhibits mouse oocyte meiosis by disturbing MPF activity.

Fangchinoline (FA) is an alkaloid derived from the traditional Chinese medicine Fangji. Numerous studies have shown that FA has a toxic effect on various cancer cells, but little is known about its toxic effects on germ cells, especially oocytes. In this study, we investigated the effects of FA on mouse oocyte maturation and its potential mechanisms. Our results showed that FA did not affect meiosis resumption but inhibited the first polar body extrusion. This inhibition is not due to abnormalities at the organelle level, such as chromosomes and mitochondrial, which was proved by detection of

A homozygous loss-of-function mutation in FBXO43 causes human non-obstructive azoospermia.

Non-obstructive azoospermia (NOA) represents one of the most serious forms of male infertility caused by spermatogenic failure. Despite multiple genes found to be associated with human NOA, the genetic basis of this idiopathic disease remains largely unknown. FBXO43 is a direct inhibitor of the anaphase-promoting complex/cyclosome (APC/C) E3 ligase and crucially important in mouse spermatogenesis. In this study, for the first time, we identified a homozygous nonsense mutation in FBXO43 c.1747C > T:p.Gln583X in two NOA brothers from a Chinese consanguineous family via whole-exome sequencing. Cardiomyocyte binucleation is associated with aberrant mitotic microtubule distribution, mislocalization of RhoA and IQGAP3, as well as defective actomyosin ring anchorage and cleavage furrow ingression.

AIMS: After birth mammalian cardiomyocytes initiate a last cell cycle which results in binucleation due to cytokinesis failure. Despite its importance for cardiac regenerative therapies, this process is poorly understood. Here, we aimed at a better understanding of the difference between cardiomyocyte proliferation and binucleation and providing a new tool to distinguish these two processes. METHODS AND RESULTS: Monitoring of cell division by time-lapse imaging revealed that rat cardiomyocyte binucleation stems from a failure to properly ingress the cleavage furrow. Astral microtubule require

Prenatal diagnosis of 45,X/46,XY mosaicism with postnatal confirmation in a phenotypically normal male infant.

Prenatal detection of chromosome mosaicism has always been a diagnostic dilemma. In 21 reported cases of chromosomal mosaicism in cultured amniotic fluid cells, only two cases had cytogenetic confirmation of the mosaicism. All 21 pregnancies resulted in either phenotypically normal liveborns or grossly normal abortuses. We report a case of XO/XY mosaicism detected prenatally and confirmed postnatally in a grossly normal male infant. The indication for prenatal cytogenetic diagnosis was advanced maternal age (38 years). A diagnosis of XO/XY mosaicism was made from two separate culture flasks of

WEE1 opposes APC/CCDH1 activity to promote S phase entry.

WEE1 phosphorylates and inhibits the activity of cyclin-dependent kinases to prevent mitotic entry and establish a G2 DNA damage checkpoint1-3. Consequently, WEE1 inhibitors have been developed to be synthetically lethal in TP53-mutated tumours that become reliant on this WEE1-mediated checkpoint, and such inhibitors are undergoing clinical trials. However, WEE1 inhibitors also have efficacy in TP53-wild-type tumours, and many trials have been terminated owing to high levels of toxic side effects, suggesting that WEE1 has unknown functions. Here we show that WEE1 promotes cell cycle re-entry f

Assessment of Salvinia molesta for phytoremediation potential of copper and lead in contaminated water: Their impact on morpho-physio-cytological status.

This study evaluates the phytotoxic and cytogenetic effects of copper (Cu) and lead (Pb) on the aquatic fern Salvinia molesta, with emphasis on its potential for phytoremediation of heavy metal-contaminated water. Plants were exposed to graded concentrations (20, 40, 120 and 200 µmol/L) of Cu and Pb for 15 days. Pb exposure induced visible morphological alterations, while Cu had minimal structural impact. Both metals caused a dose-dependent reduction in chlorophyll (up to 60.82 % by Pb at 200 µmol/L) and protein content, accompanied by increased proline accumulation and elevated antioxidant en

RSK inhibitor BI-D1870 inhibits acute myeloid leukemia cell proliferation by targeting mitotic exit.

The 90 kDa Ribosomal S6 Kinase (RSK) drives cell proliferation and survival in cancers, although its oncogenic mechanism has not been well characterized. Phosphorylated level of RSK (T573) was increased in acute myeloid leukemia (AML) patients and associated with poor survival. To examine the role of RSK in AML, we analyzed apoptosis and the cell cycle profile following treatment with BI-D1870, a potent inhibitor of RSK. BI-D1870 treatment increased the G2/M population and induced apoptosis in AML cell lines and patient AML cells. Characterization of mitotic phases showed that the metaphase/an
