Tetrodotoxin-sensitive Navs contribute to early and delayed afterdepolarizations in long QT arrhythmia models.

Recent evidence suggests that neuronal Na+ channels (nNavs) contribute to catecholamine-promoted delayed afterdepolarizations (DADs) and cat … ine (4AP), whereas the L-type Ca2+ and late Na+ currents were augmented using Bay K 8644 (BayK) and anemone toxin II (ATX-II), respectively. The combination of 4AP and isoproterenol prolonged action potential duration (APD) and promoted aberrant Ca2+ release, EADs, and DADs in wild-type cardiomyocytes. Similarly, BayK in the absence of isoproterenol induc …

I(Ks) restricts excessive beat-to-beat variability of repolarization during beta-adrenergic receptor stimulation.

In vivo studies have suggested that increased beat-to-beat variability of ventricular repolarization duration (BVR) is a better predictor of … us pacing rates. I(Ks) was blocked by HMR1556, I(Kr) by dofetilide. Late I(Na) was augmented by sea anemone toxin-II. Isoproterenol was added for beta-adrenergic receptor stimulation. BAPTA-AM buffered intracellular Ca(2+). SEA0400 partially inhibited the Na(+)-Ca(2+) exchanger. BVR was quantified as variability of action-potential duration at 90% repolar …

Distribution and bioaccumulation of per- and polyfluoroalkyl substances (PFAS) in marine intertidal invertebrates - A multi-bioindicator study from Cape Town, South Africa.

Per- and polyfluoroalkyl substances (PFAS) are toxic, bioaccumulative, and highly persistent, raising global concerns over their environmental and health impacts. Effective biomonitoring is essential, particularly in regions that remain understudied. This study investigated PFAS bioaccumulation in five intertidal invertebrate species (mussels, limpets, periwinkles, sea urchins, and anemones) from False Bay, South Africa, a biodiverse coastal embayment of high ecological and socio-economic value but with a history of pollution from urbanisation and wastewater outfall. We investigated spatial va…

The role of exposed tryptophan residues in the activity of the cardiotonic polypeptide anthopleurin B.

Scorpion and sea anemone venoms contain several polypeptides that delay inactivation of voltage-sensitive sodium channels via interaction with a common site. In this report, we target exposed hydrophobic residues at positions 33 and 45 of anthopleurin B (ApB) by polymerase chain reaction mutagenesis to ascertain their contribution to toxin activity. Nonconservative replacements are not permitted at position 33, indicating that Trp-33 may play an important structural role. Strikingly, the relatively conservative substitution of Trp-33 by phenylalanine results in major reductions in binding affi…

Sticholysin II: a pore-forming toxin as a probe to recognize sphingomyelin in artificial and cellular membranes.

Sphingomyelin is a major component of membrane rafts, and also is a precursor of many bioactive molecules. The sphingomyelin plays important biological roles and alterations of its metabolism are the basis of some genetic disorders such as the Niemann Pick disease. A complete understanding of its biological role is frustrated by the lack of efficient tools for its recognition in the cell. Sticholysin II (StnII) is a 20 kDa protein from the sea-anemone Stichodactyla helianthus which shows a cytotoxic activity by forming oligomeric aqueous pores in the cell plasma membrane. A recent NMR analysis…

Voltage-dependent effect of a scorpion toxin on sodium current inactivation.

In voltage clamped nodes of Ranvier inactivation of the sodium permeability is slowed by toxin V from the scorpion Centruroides sculpturatus, by sea anemone toxin ATX II or by internally applied KIO3. The slow decay of the Na inward current is markedly accelerated if the test pulse is preceded by a depolarizing conditioning pulse followed by a 10-500 ms pause. This phenomenon was studied in detail, using conditioning pulses of varying amplitude and up to 15 s duration. In nodes treated with toxin V a 20 ms conditioning pulse to positive potentials was sufficient to produce a clear acceleration…

Tyrosine-driven hierarchical self-assembly unlocks robust non-Dopa wet adsorption in marine adhesive protein.

Many marine sessile organisms achieve robust underwater adsorption through 3,4-dihydroxyphenylalanine (Dopa)-independent strategies, yet the underlying self-assembly mechanisms remain elusive. Here, we decipher the Ca2+-triggered interfacial adsorption of a sea anemone thrombospondin-1 type I repeat-like (TSRL) protein, identifying its tyrosine-rich T3 subunit as the core mediator for coating formation. The fabricated T3 coatings exhibited strong wet adsorption stability, biocompatibility, and intrinsic antioxidant activity. Mechanistically, T3 self-assembly is driven by favorable enthalpy, in…

A Matter of Degrees: Latitudinal Variation in the Transcriptional Response to High and Low Temperatures in an Estuarine Cnidarian.

Populations of the same species inhabiting distinct geographical regions must meet the requirements of local thermal regimes to survive. While individuals integrate both deeply conserved and genotype-specific transcriptional responses to temperature shifts, unique local requirements may diversify the balance between these two mechanisms in distinct populations. The sea anemone Nematostella vectensis inhabits highly variable estuarine environments across a broad geographic range, providing an excellent system to investigate how local adaptations shape responses to temperature stress. While stud…

Raddeanin A induces ferroptosis by targeting VEPH1 in a cisplatin-resistant epithelial ovarian cancer cell line.

Ovarian cancer (OC) is a fatal female malignancy, and cisplatin resistance severely impacts the clinical management of epithelial ovarian cancer (EOC), the most prevalent histological subtype accounting for over 90% of cases. Exploration of novel effective targets and drugs for treatment of cisplatin-resistant EOC is urgently needed to address the tremendous challenge. In this study, we screened a Traditional Chinese Medicine (TCM) library including 978 monomers and found that Raddeanin A (RA) extracted from Anemone raddeana Regel displayed the powerful cytotoxic effect on cisplatin-resistant …

Augmentation of recovery from inactivation by site-3 Na channel toxins. A single-channel and whole-cell study of persistent currents.

Site-3 toxins isolated from several species of scorpion and sea anemone bind to voltage-gated Na channels and prolong the time course of INa by interfering with inactivation with little or no effect on activation, effects that have similarities to those produced by genetic diseases in skeletal muscle (myotonias and periodic paralysis) and heart (long QT syndrome). Some published reports have also reported the presence of a noninactivating persistent current in site-3 toxin-treated cells. We have used the high affinity site-3 toxin Anthopleurin B to study the kinetics of this current and to eva…

The sodium channel from rat brain. Reconstitution of neurotoxin-activated ion flux and scorpion toxin binding from purified components.

The rat brain Na+ channel was purified to homogeneity and reconstituted into pure egg phosphatidylcholine vesicles or vesicles composed of a … arallel loss of both the [3H]STX- and 125I-LqTx-binding activity measured after reconstitution. Sea anemone toxin II displaces bound 125I-LqTx with a KD 60-fold greater than that of unlabeled LqTx. Antillatoxin is a marine cyanobacterial toxin that potently activates voltage-gated sodium channels.

Antillatoxin (ATX) is a lipopeptide derived from the pantropical marine cyanobacterium Lyngbya majuscula. ATX is neurotoxic in primary cultures of rat cerebellar granule cells, and this neuronal death is prevented by either N-methyl-d-aspartate (NMDA) receptor antagonists or tetrodotoxin. To further explore the potential interaction of ATX with voltage-gated sodium channels, we assessed the influence of tetrodotoxin on ATX-induced Ca2+ influx in cerebellar granule cells. The rapid increase in intracellular Ca2+ produced by ATX (100 nM) was antagonized in a concentration-dependent manner by tet

Shared antigenicity between the polar filaments of myxosporeans and other Cnidaria.

Nematocysts containing coiled polar filaments are a distinguishing feature of members of the phylum Cnidaria. As a first step to characterizing the molecular structure of polar filaments, a polyclonal antiserum was raised in rabbits against a cyanogen bromide-resistant protein extract of mature cysts containing spores of Myxobolus pendula. The antiserum reacted only with proteins associated with extruded polar filaments. Western blot and whole-mount immunohistochemical analyses indicated a conservation of polar filament epitopes between M. pendula and 2 related cnidarians, i.e., the anthozoan,

Ultrastructural and immunocytochemical evidence of a colonial nervous system in hydroids.

BACKGROUND: As the sister group to all Bilateria, representatives of the phylum Cnidaria (sea anemones, corals, jellyfishes, and hydroids) possess a recognizable and well-developed nervous system and have attracted considerable attention over the years from neurobiologists and evo-devo researchers. Despite a long history of nervous system investigation in Cnidaria, most studies have been performed on unitary organisms. However, the majority of cnidarians are colonial (modular) organisms with unique and specific features of development and function. Nevertheless, data on the nervous system in c…

Animal venom studies: Current benefits and future developments.

Poisonous organisms are represented in many taxa, including kingdom Animalia. During evolution, animals have developed special organs for production and injection of venoms. Animal venoms are complex mixtures, compositions of which depend on species producing venom. The most known and studied poisonous terrestrial animals are snakes, scorpions and spiders. Among marine animals, these are jellyfishes, anemones and cone snails. The toxic substances in the venom of these animals are mainly of protein and peptide origin. Recent studies have indicated that the single venom may contain up to several…
