[Differential diagnosis of delayed encephalopathy induced by anthelmintic imidazoles].

Anthelmintic imidazoles induced delayed encephalopathy (AIIDE) is frequently encountered and has usually been misdiagnosed as sporadic encephalitis of unknown cause (SEU). This study was carried out to facilitate differentiation of the two diseases. The records of 202 cases of AIIDE were reviewed and the clinical features analyzed. As a results, 7 diagnostic criteria for AIIDE were proposed as follows: (1) history of exposure to anthelmintic before the onset; (2) presence of a latent period of of 2 to 5 weeks; (3) onset of the disease acute or subacute; (4) presence of neuropsychiatric symptom

Molecular modeling: a tool for predicting anthelmintic activity in vivo.

The structural and electronic features of the broad-spectrum benzimidazole anthelmintic mebendazole [MBZ, methyl 5-(benzoyl)-benzimidazole-2-carbamate] have been determined using a combination of quantum mechanics, molecular graphics, and molecular modeling techniques. Using conformational analyses and quantum mechanics, we found that the three-dimensional structure and electronic features of MBZ were consistent with those previously reported for highly active broad-spectrum benzimidazole anthelmintics and that in vivo drug efficacy against Hymenolepis diminuta depends upon the orientation of …

The anthelmintic effects of flubendazole on Trichuris trichiura and Ascaris lumbricoides.

Flubendazole is an effective anthelmintic against both Trichuris trichiura and Ascaris lumbricoides. Single doses of 200 mg, 500 mg and 600 mg flubendazole, or two doses of 300 mg flubendazole on two consecutive days, effectively reduced egg output of both helminths by over 90%. Cure rates of 90.0 to 97.0% were obtained in Ascaris infections but only 17.3 to 65.1% of Trichuris infections were completely cured with the various single-dose or two-dose regimes. Flubendazole did not cause erratic migration of Ascaris in mixed infections at any of the dosages used.

[Biological assay of anthelmintic drugs. Determination of DE 50].

Experiments on ivermectine, thiabendazole and levamisole were performed by using the Nippostrongylus brasiliensis rat model. The aim of the experiments was to study the 50% as well as 99.5% doses of the above anthelmintics. Besides this a relative evaluation of drugs was also carried out and different dose efficiencies were compared by keeping one anthelmintic as a standard. The results were obtained on the following experimental conditions: homogeneity of the group of rats (age, weight and sex), uniform infestation in all the animals (3000 larvae at day zero), administration of anthelmintics …

The efficacy of amidantel, a new anthelmintic, on hookworms and ascarids in dogs.

Amidantel is a new anthelmintic from a new chemical class with an interesting anthelmintic spectrum. In dogs amidantel is highly effective in a single oral dose of 25 mg/kg against both hookworm species, Ancylostoma caninum and Unicinaria stenocephala. In Toxascaris leonina infected dogs a complete cure rate was achieved with a single oral dose of 50 mg/kg. Similar results were obtained with 8 mg/kg administered three times per day. The most sensitive parasite to amidantel was found to be Toxocara canis with a 100 per cent cure rate after a single oral treatment with 10 mg/kg. In preliminary t

Levamisole: anthelmintic activity in calves following dermal application.

A series of seven experiments designed to evaluate the anthelmintic efficacy of levamisole (1-tetramisole) by dermal application is described. This work involved use of 181 artificially infested calves. The drug was formulated at a concentration of 10% m/v levamisole base in a solvent system promoting dermal absorption and was applied to both sides of the spine in the lumber region at a dose rate of 10 mg levamisole base per kg livemass. A consistent performance was achieved in these investigations, the results from all of which were based upon necropsy findings. Efficacy of treatment was asse

Clinical trials with mebendazole a new broad spectrum anthelmintic.

This paper presents our experience with mebendazole, a new broad-spectrum anthelmintic, in 91 cases with ascariasis, 71 cases with necatoriasis 54 cases with trichuriasis and 10 cases with T. saginata infection. Various dosage schedules were evaluated. In ascariasis, a single dose of 200 mg was effective in totally clearing the infection in 100 per cent cases. In necatoriasis 200 mg twice daily for four days was found to be the most effective dose. With this dose total eradication of the infection was seen in 92 per cent cases. In trichuriasis, 200 mg twice daily for four days totally eradicat

Evolutionary and epidemiological considerations for anthelmintic treatments in ruminant livestock.

Anthelmintic drug resistance is a concern for the sustained control of gastrointestinal nematodes (GINs) in ruminant livestock globally. Evolutionary-epidemiological modelling, which considers both parasite dynamics and resistance dynamics in response to interventions, can be useful in determining which anthelmintic resistance management (ARM) strategies may be effective without compromising parasite control. We address two key questions in ARM. First, how to improve the measurement of AR in populations. Second, identifying effective ARM strategies to slow the spread of AR while maintaining ef

Understanding anthelmintic resistance in livestock using "omics" approaches.

Widespread and improper use of various anthelmintics, genetic, and epidemiological factors has resulted in anthelmintic-resistant (AR) helminth populations in livestock. This is currently quite common globally in different livestock animals including sheep, goats, and cattle to gastrointestinal nematode (GIN) infections. Therefore, the mechanisms underlying AR in parasitic worm species have been the subject of ample research to tackle this challenge. Current and emerging technologies in the disciplines of genomics, transcriptomics, metabolomics, and proteomics in livestock species have advance…

Anthelmintic resistance in small ruminants in the Nordic-Baltic region.

Gastrointestinal nematodes (GIN) in small ruminants result in production losses, and consequently economic losses, and are an animal welfare problem in most countries in the Nordic-Baltic region. Intensive use of anthelmintics to control helminth infections has led to anthelmintic resistance (AR), which has become a major issue in many European countries. Several studies have been performed in countries in the Nordic-Baltic region (e.g. Denmark, Sweden, Norway and Lithuania) showing increasing/emerging levels of AR. The aim of this paper is to provide an overview of the problem of AR on sheep …

Natural variation in Caenorhabditis elegans responses to the anthelmintic emodepside.

Treatment of parasitic nematode infections depends primarily on the use of anthelmintics. However, this drug arsenal is limited, and resistance against most anthelmintics is widespread. Emodepside is a new anthelmintic drug effective against gastrointestinal and filarial nematodes. Nematodes that are resistant to other anthelmintic drug classes are susceptible to emodepside, indicating that the emodepside mode of action is distinct from previous anthelmintics. The laboratory-adapted Caenorhabditis elegans strain N2 is sensitive to emodepside, and genetic selection and in vitro experiments impl…
