USO1 Coordinates Centriolar Satellites to Regulate Male Germ Cell Proliferation and Cell Cycle Progression.

The endoplasmic reticulum-Golgi apparatus system is an important organelle regulating male reproduction. USO1 vesicle transport factor (USO1), as an important molecule in this system, is a general vesicular transport factor and regulates various biological processes in vivo. However, the potential role of USO1 in mammalian testis development and spermatogenesis has not been investigated. We documented the presence of USO1 in mouse germ cells and its functional roles by generating Uso1-knockout germ cell lines. Uso1 depletion suppressed cell proliferation and growth while stimulating apoptosis

Development of predictive retention-activity relationship models of tricyclic antidepressants by micellar liquid chromatography.

The distribution of tricyclic antidepressants from plasma to brain, where these drugs exert their main clinical action, and other organs is related to transport events across the cell membranes of the different tissues. It could be expected that all the molecular features that condition the transport processes (mainly hydrophobicity and molar total charge) also control the pharmacokinetic and biochemical behavior. Micellar liquid chromatography (MLC) has been proposed to emulate in vitro the partitioning process in the biomembranes. The use of micellar solutions of Brij35 as mobile phases in r

From random walks to thermal rides: universal anomalous transport in soaring flights.

Cross-country soaring flights rely on intermittent atmospheric updrafts to cover long distances, producing trajectories that alternate between rapid relocation and local exploration. From a large dataset of paraglider, hang glider and sailplane flights, we uncover a universal transport law: beyond short ballistic times, horizontal motion is persistently sub-ballistic, with a Hurst exponent ≈0.88 largely independent of aircraft type. Phase-resolved analysis using a probabilistic segmentation method shows that this scaling arises from the fundamentally intermittent, two-dimensional and direction

Development of synthetic chloride transporters using high-throughput screening and machine learning.

The development of synthetic compounds capable of transporting chloride anions across biological membranes has become an intensive research field in the last two decades. Progress is driven by the desire to develop treatments for chloride transport related diseases (e.g., cystic fibrosis), cancer or bacterial infections. In this manuscript, we use high-throughput screening and machine learning to identify novel scaffolds, and to find the molecular features needed to achieve potent chloride transport that can be generalized across diverse chemotypes. 1894 compounds were tested, 59 of which had

A comparison between transport and diffusion calculations using a finite element-spherical harmonics radiation transport method.

Most researchers choose the diffusion approximation to the transport equation as the model to describe photon migration in biological tissues. However, the applicability of this approximation is limited and, in certain cases, invalid. In this paper we introduce a two-dimensional, finite element-spherical harmonics (FE-P(N)) radiation transport method for the simulation of light propagation in tissue. The propagation of light is investigated first in a layered cylinder, which can be seen as a very simplistic approximation of a human head. Effects of the anisotropy factor g on the photon migrati

Bio-Inspired Retina by Regulating Ion-Confined Transport in Hydrogels.

The effective and precise processing of visual information by the human eye primarily relies on the diverse contrasting functions achieved through synaptic regulation of ion transport in the retina. Developing a bio-inspired retina that uses ions as information carriers can more accurately replicate retina's natural signal processing capabilities, enabling high-performance machine vision. Herein, an ion-confined transport strategy is proposed to construct a bio-inspired retina by developing artificial synapses with inhibitory and excitatory contrasting functions. By fine-tuning the ionic hydro

Epithelial morphogenesis, tubulogenesis and forces in organogenesis.

As multi-cellular organisms evolved from small clusters of cells to complex metazoans, biological tubes became essential for life. Tubes are typically thought of as mainly playing a role in transport, with the hollow space (lumen) acting as a conduit to distribute nutrients and waste, or for gas exchange. However, biological tubes also provide a platform for physiological, mechanical, and structural functions. Indeed, tubulogenesis is often a critical aspect of morphogenesis and organogenesis. C. elegans is made up of tubes that provide structural support and protection (the epidermis), perfor

Dynamic Heterogeneity and Stretched Exponential Distributions in Cargo Transport within Living Cells.

Stretched exponential distributions of nonequilibrium steady-state fluctuations are ubiquitous in living and nonliving systems alike, yet their microscopic origins have remained elusive. In this Letter, we track the motion of three types of vesicles across different cell types and find that their trajectories undergo random transitions between directed runs (on state) and diffusionlike jiggling (off state) in the highly dynamic, heterogeneous cytoplasm. The dwell times in these two states follow a stretched exponential distribution, which arises from a continuous weighted sum of simple exponen

Late-stage placental barrier model for transport studies of prescription drugs during pregnancy.

Throughout pregnancy, the placental barrier is crucial for fetal development, evolving continuously to meet the growing nutritional demands of the fetus. Although the placenta has the capacity to selectively filter compounds, harmful xenobiotic substances from the maternal blood can sometimes cross over into the fetal circulation. This drives the development of in vitro placental barrier models in the context of drug transport studies. In this work, we adapted our lab's previous placental barrier model to transplacental drug transport by transitioning from self-assembled vasculature to a simpl

Chitinase-1 Activity in Serum of Cats with FIP.

BACKGROUND: Chitotriosidase (chitinase 1 or CHIT1) is secreted by activated macrophages. Macrophages are involved in the pathogenesis of feline infectious peritonitis (FIP). No reports on CHIT1 activity in cats with FIP are available. OBJECTIVE: To preliminarily investigate the possible changes in serum CHIT1 activity in cats with FIP. METHODS: CHIT1 activity was measured in serum samples from clinically healthy cats (n = 17), cats with FIP (n = 19) and cats with diseases potentially characterized by macrophage activation (n = 20), after a preliminary assessment of the imprecision and linear

Mechanisms of uptake and transport of particulate formulations in the small intestine.

Micro- and nano-scale particulate formulations are widely investigated towards improving the oral bioavailability of both biologics and drugs with low solubility and/or low intestinal permeability. Particulate formulations harnessing physiological intestinal transport pathways have recently yielded remarkably high oral bioavailabilities, illustrating the need for better understanding the specific pathways underpinning particle small intestinal absorption and the relative role of intestinal cells. Mechanistic knowledge has been hampered by the well acknowledged limitations of current in vitro,

Fat caves: caveolae, lipid trafficking and lipid metabolism in adipocytes.

Caveolae are subdomains of the eukaryotic cell surface, so named because they resemble little caves, being small omega-shaped invaginations of the plasma membrane into the cytosol. They are present in many cell types, and are especially abundant in adipocytes, in which they have been implicated as playing a role in lipid metabolism. Thus, mice and humans lacking caveolae have small adipocytes and exhibit lipodystrophies along with other physiological abnormalities. In this review, we examine the evidence supporting the role of caveolae in adipocyte lipid metabolism in the context of the protei

[Interaction of iron, zinc and copper in the body of animals and humans].

In the light of the pertinent literature the sites and mechanisms of interactions of iron, zinc and copper in the human and animal organism are discussed. Both low and high intake of any of them may influence the utilization and metabolism of both remaining ones. E.G. dietary copper deficiency impairs the mobilization of iron reserves from the liver, and its excess inhibits the intestinal absorption of iron and zinc. Iron in excess may antagonize copper and zinc in the intestinal mucosa. Zinc excess has an unfavourable influence on iron balance by reducing the bioavailability of copper, and zi

Effects of high pressure on glucose transport in the human erythrocyte.

The effects of raised hydraulic pressure on D-glucose exit from human red cells at 25 degrees C were determined using light scattering measurements in a sealed pressurized spectrofluorimeter cuvette. The reduction in the rates of glucose exit with raised pressure provides an index of the activation volume, deltaV++ (delta ln k/deltaP)(T) = -deltaV++/RT. Raised pressure decreased the rate constant of glucose exit from 0.077 +/- 0.003 s(-1) to 0.050 +/- 0.002 s(-1) (n = 5, P < 0.003). The Ki for glucose binding to the external site was 2.7 +/- 0.4 mm (0.1 MPa) and was reduced to 1.45 +/- 0.15 mm

Phosphate Acquisition in Protozoan Parasites: Transport Systems, and Extracellular Phosphate-Releasing Enzymatic Activities.

Inorganic phosphate (Pi) is an essential nutrient required for energy metabolism, macromolecule biosynthesis, and signal transduction. Protozoan parasites are exposed to pronounced fluctuations in nutrient availability throughout their life cycles and therefore rely on adaptive strategies to secure phosphate from diverse host and environmental niches. This review summarizes current knowledge on phosphate acquisition mechanisms in protozoan parasites, with emphasis on membrane transport systems and extracellular phosphate-releasing enzymatic activities. Phosphate transport systems energized by

Matrix-free human 2D organoids recapitulate duodenal barrier and transport properties.

BACKGROUND: Traditionally, transformed cell line monolayers have been the standard model for studying epithelial barrier and transport function. Recently, intestinal organoids were proposed as superior in recapitulating the intestine. Typically, 3D organoids are digested and seeded as monolayers on gelatinous matrix pre-coated surfaces for anchorage. As this coat could potentially act as a diffusion barrier, we aimed to generate robust human duodenum-derived organoid monolayers that do not need a gelatinous matrix for anchorage to improve upon existing models to study epithelial transport and

Perivascular interactions and tissue properties modulate directional glymphatic transport in the brain.

The glymphatic theory suggests a convective transport mechanism through brain tissue, which has significant implications for both brain waste clearance and drug delivery. However, the existence and driving mechanisms of directional convection from periarterial to perivenous spaces remain debated. Additionally, the role of brain tissue stiffness in parenchymal transport remains unclear, as experiments have reported varying trends in stiffness changes in cases of aging and neurodegenerative diseases. Previous mechanistic models often simplify or neglect perivenous spaces and venous deformation,
