Cognitive impairment and increased brain neurosteroids in adult rats perinatally exposed to low millimolar blood alcohol concentrations.

Epidemiological evidence suggests that adolescents and adults perinatally exposed to alcohol, even at low doses, show high prevalence of cognitive impairment and social behavior deficits, which may be in part related to alcohol-induced changes of the gamma-aminobutyric acid (GABA)ergic neurotransmission. The endogenous neurosteroid 3alpha-hydroxy,5alpha-pregnan-20-one (3alpha,5alpha-tetrahydroprogesterone/3alpha,5alpha-THP), a potent positive allosteric modulator of GABA(A) receptor function, is implicated in the physiological tuning of GABA-mediated fast inhibition and in various alcohol's ac

Myocardial oxidative stress and toxicity induced by acute ethanol exposure in mice.

Alcoholic cardiomyopathy has been known for a long time, but there is little mechanistic insight into this important clinical problem. The present study was undertaken using a mouse model to test the hypothesis that alcohol exposure induces cardiac injury through induction of oxidative stress. Adult female Friend Virius B-type (FVB) mice were treated with ethanol by gavage at a dose of 5 g/kg. Six hours after the treatment, ethanol-induced myocardial injury was observed, as indicated by a significant increase in serum creatine phosphokinase activity, a common biomarker of myocardial injury, an

Blood acetone concentrations in forensic autopsies are related to the food residue in stomach.

Elevated blood acetone concentrations are used as an initial marker to identify ketoacidosis in forensic autopsies. However, blood acetone concentrations may not reflect the status of ketoacidosis. This study aimed to clarify the significance of measuring blood acetone concentrations in routine forensic autopsies. We studied 367 forensic autopsies to determine the relationships between blood acetone concentrations and blood ethanol concentrations, blood biochemical laboratory test values, as well as the physical factors that may affect blood acetone concentrations. Blood acetone concentrations

Liver size and indices of drug metabolism in alcoholics.

The role of liver size in drug metabolism was investigated in 34 chronic alcoholics and 28 controls by comparing antipyrine half-life with biopsy content and total amount of hepatic cytochrome P-450 (P-450) and liver weight. Liver size was significantly greater in alcoholics than in controls. Total P-450 was increased and antipyrine metabolism was enhanced in alcoholics with normal histology of the liver. In subjects with alcoholic hepatitis or cirrhosis, the antipyrine half-life was prolonged and P-450 was decreased. Alcoholics with fatty liver had a reduced P-450 content, but the total amoun

Analysis of risk factors for DUI and DWI crashes considering the built environment.

The risk level of alcohol-involved traffic crashes is closely related to alcohol consumption. However, research on different influencing factors for DUI (Driving Under Influence) and DWI (Driving While Intoxicated) remains limited. This study analyzed data from 3,365 alcohol-related traffic crashes in Tianjin, China. The crashes were categorized into DUI and DWI based on drivers' Blood Alcohol Concentration. Four machine learning models were enhanced and compared. The accuracy, precision, recall and F1-score were used to evaluate the performance of the models. Shapley additive explanations wer

Oral Administration of Alcohol-Tolerant Lactic Acid Bacteria Alleviates Blood Alcohol Concentration and Ethanol-Induced Liver Damage in Rodents.

Excessive alcohol consumption can have serious negative consequences on health, including addiction, liver damage, and other long-term effects. The causes of hangovers include dehydration, alcohol and alcohol metabolite toxicity, and nutrient deficiency due to absorption disorders. Additionally, alcohol consumption can slow reaction times, making it more difficult to rapidly respond to situations that require quick thinking. Exposure to a large amount of ethanol can also negatively affect a person's righting reflex and balance. In this study, we evaluated the potential of lactic acid bacteria

Impact of sex, strain, and age on blood ethanol concentration and behavioral signs of intoxication during ethanol vapor exposure.

Animal models of alcohol drinking and dependence are a critical resource for understanding the neurobiological mechanisms and development of more effective treatments for alcohol use disorder (AUD). Because most rat strains do not voluntarily consume large enough quantities of alcohol to adequately model heavy drinking, dependence, and withdrawal-related symptoms, researchers frequently turn to experimenter administered methods to investigate how prolonged and repeated exposure to large quantities of alcohol impacts brain and behavior. Vaporized ethanol is a common method used for chronically

Benefits of short educational programmes in preventing drink-driving recidivism: A ten-year follow-up randomised controlled trial.

BACKGROUND: One fifth of drivers convicted of drink-driving for the first time will be convicted again for the same offence in subsequent years. Lecture-based educational programmes are believed to reduce recidivism. Little is known about the modalities of enhancing the benefits of such programs. METHODS: This parallel randomised controlled trial measured the effects of the duration of lectures and the presence of a close relative 'in class' on rates of recidivism during the ten years following an initial drink-driving conviction. Of 1588 drivers in the Canton of Geneva convicted of a first-t

Ethanol concentrations in various biological specimens: Living and postmortem forensic toxicology analysis and comprehensive literature review.

Alcohol use upsurges the risk for many chronic ill-health consequences such as hepatitis, malignancies, and disastrous outcomes like road traffic accidents ending in fatal injuries. Biochemical and toxicological analysis of different body fluids is crucial for identifying the cause of death and postmortem interval in many forensic cases. Blood, urine, and vitreous fluid are the most valuable body fluids for detecting alcohol during any toxicological analysis. Alcohol is responsible for widespread morbidity and mortality worldwide. Blood alcohol concentration (BAC) is a necessary toxicological

Clinical and forensic evaluation of 66 cases of alcohol intoxication: Insights into substitute alcohols detected by HS-GC/MS.

Rising global alcohol consumption is a major public health and safety concern. Over the last decade, undeclared alcohol consumption has increasingly come into focus due to high inflation that has led many individuals to resort to unhealthy methods to access alcohol. In this context, the development of methods for alcohol detection in biological samples is of great importance in terms of rapid diagnosis, effective treatment, and obtaining reliable evidence in forensic processes. In this study, an HS-GC/MS method for the evaluation of alcohol intoxication in blood samples was developed and valid

Association between acute pre-injury alcohol use and 12-month health outcomes for survivors of major trauma: A registry-based study.

INTRODUCTION: Alcohol is commonly detected in patients presenting to hospital after major trauma and is a key preventable risk factor for injury. While it has been suggested that alcohol intoxication at the time of injury results in worse acute patient outcomes, there is currently limited knowledge on the impact of alcohol on health outcomes following hospital discharge. The aim of this study was to examine the relationship between acute pre-injury alcohol exposure and the self-reported health outcomes of survivors of major trauma 12-months post-injury. METHODS: Data from the Victorian State

Acute alcohol does not impair attentional inhibition as measured with Stroop interference scores but impairs Stroop performance.

RATIONALE: Inhibition is a core executive function and refers to the ability to deliberately suppress attention, behavior, thoughts, and/or emotions and instead act in a specific manner. While acute alcohol exposure has been shown to impair response inhibition in the stop-signal and Go/NoGo tasks, reported alcohol effects on attentional inhibition in the Stroop task are inconsistent. Notably, studies have operationalized attentional inhibition variably and there has been intra- and inter-individual variability in alcohol exposure. OBJECTIVE: This study aimed to examine the acute effects of al

Fatal acute intoxication after snorting cocaine and fluoxetine.

A case report of a fatal acute intoxication following cocaine and fluoxetine snorting is described here. A 44-year-old woman with an history of drug abuse was found dead at home. On-site evaluation conducted by police highlighted evidence of recent cocaine consumption whereas no tablets or drug blisters were spotted. In the hypothesis of a drug-related crime the local Prosecutor ordered a judicial autopsy. Pathological findings were consistent with an acute cardiac failure while toxicological analysis conducted on blood revealed the presence of fluoxetine and cocaine in reportedly fatal concen
