Cancer epidemiology, biomarkers & prevention : a publication of the American Association for Cancer Research, cosponsored by the American Society of Preventive Oncology2014eng
BACKGROUND: Mammographic density (MD) is one of the strongest known breast cancer risk factors. Twin studies have suggested that a large part of the variation in MD is genetically determined. We hypothesized that breast cancer susceptibility variants may affect MD, and that their effects may be modified by nongenetic factors.
METHODS: We assessed MD, using a computer-assisted method, on 2,348 postmenopausal Caucasian women (50-69 years) who participated in the Norwegian Breast Cancer Screening Program (NBCSP) in 2004 or 2006-07. We used linear regression (additive models) to determine the ass

PURPOSE: Mammographic breast density (BD) is associated with increased risk of breast cancer. This study asks which circulating metabolic and reproductive biomarkers are associated with BD, particularly dense breast area, in premenopausal women not taking exogenous hormones.
METHODS: In a cross-sectional study, 299 premenopausal women aged 40-49 completed questionnaires, provided a fasting blood sample, had height, weight, percentage body fat, waist and hip measurements taken, and attended a screening mammogram. Multivariate linear regression was used to calculate adjusted means for percentag…

A computational method was developed for the measurement of breast density using chest computed tomography (CT) images and the correlation between that and mammographic density. Sixty-nine asymptomatic Asian women (138 breasts) were studied. With the marked lung area and pectoralis muscle line in a template slice, demons algorithm was applied to the consecutive CT slices for automatically generating the defined breast area. The breast area was then analyzed using fuzzy c-mean clustering to separate fibroglandular tissue from fat tissues. The fibroglandular clusters obtained from all CT slices …

THE AIM OF THIS STUDY: To implement a spatial fuzzy C-means algorithm for image segmentation and breast tissue density quantification and compare it with BI-RADS breast density classes determined by radiologists.
MATERIAL AND METHODS: The analysis was based on 206 mammograms performed in 111 women with various breast abnormalities. Digitized mammographic films were independently double read by radiologists certified in breast diagnosis, followed by consensus with arbitration agreement (radiological ground truth). Reporting was done using the BI-RADS mammography lexicon. Using an algorithm bas…

OBJECTIVES: We examined breast density by dual energy X-ray absorptiometry (DXA) in relation to pubertal maturation and body fatness among girls of several ethnic groups, which may suggest important risk factors for future breast cancer.
METHODS: Recruiting 113 girls from Kaiser Permanente Hawaii, we determined ethnicity, age at menarche, weight, height; body mass index (BMI), BMI Z-scores, Tanner breast, and pubic hair pubertal stages (and classified stages for synchrony, thelarche or adrenarche). DXA scans of the whole body provided percent total and segmental body fat; and separate (specia…

We examined the association between new-onset breast tenderness and change in mammographic density after initiation of conjugated equine estrogens (CEE). We analyzed baseline, year 1 and 2 data from 695 participants of the Women's Health Initiative Estrogen + Progestin (daily CEE 0.625 mg + medroxyprogesterone acetate 2.5 mg [MPA] or placebo) and Estrogen-Alone (CEE 0.625 mg or placebo) trials who participated in the Mammogram Density Ancillary Study. Using multivariable repeated measures models, we analyzed the association between new-onset breast tenderness (i.e. absence of baseline tenderne

PURPOSE: Mammographic density and lobular involution are both significant risk factors for breast cancer, but whether these reflect the same biology is unknown. We examined the involution and density association in a large benign breast disease (BBD) cohort.
PATIENTS AND METHODS: Women in the Mayo Clinic BBD cohort who had a mammogram within 6 months of BBD diagnosis were eligible. The proportion of normal lobules that were involuted was categorized by an expert pathologist as no (0%), partial (1% to 74%), or complete involution (>or= 75%). Mammographic density was estimated as the four-categ

The association of adiposity with dense tissue area in the breast is unclear, but suggests a mechanism by which adiposity might increase breast cancer risk. We examined associations of body mass index (BMI), usual BMI from age 20 to 29, waist circumference and adult weight gain with breast density in a sample of premenopausal United States Chinese immigrant women. Analyses included 415 participants in a longitudinal breast density study in Philadelphia. In addition to detailed questionnaire information, data collection included measures of anthropometry, and assessment of mammographic breast d…

OBJECTIVES: Breast cancer prognosis is better for smaller tumours. Women with high breast density are at higher risk of breast cancer and have larger screen-detected and interval cancers in mammographic screening programmes. We assess which continuous measures of breast density are the strongest predictors of breast tumour size at detection and therefore the best measures to identify women who might benefit from more intensive mammographic screening or alternative screening strategies.
SETTING AND METHODS: We compared the association between breast density and tumour size for 1007 screen-dete…

INTRODUCTION: Bacterial colonization of breast implants can lead to infections and capsular contracture, making contamination prevention essential. While conventional UVC254 light is germicidal, it poses risks to human skin and eyes. In contrast, UVC222 effectively kills bacteria without harming mammalian cells due to limited stratum corneum penetration. This study investigates whether UVC222 irradiation can reduce bacterial density on breast implant shells and the epidermis of the submammary fold.
MATERIAL AND METHODS: This prospective, single-center study included 31 patients who underwent

BACKGROUND: Breast density is both an independent risk factor for breast cancer and a major determinant of mammographic diagnostic performance. However, its impact on overall screening performance within population-based screening programs is less well understood.
METHODS: This retrospective cohort study included women aged 50-74 years screened through BreastScreen NSW between 1 January 2016 and 31 December 2017, excluding first-time screeners above 70. Breast density was assessed using Lunit INSIGHT MMG (v1.1.7) generating an ordinal density score (1-10) and corresponding BI-RADS category (A…

OBJECTIVES: To assess the performance of breast cancer screening by category of breast density and age in a UK screening cohort.
METHODS: Raw full-field digital mammography data from a single site in the UK, forming a consecutive 3-year cohort of women aged 50 to 70 years from 2016 to 2018, were obtained retrospectively. Breast density was assessed using Volpara software. Examinations were grouped by density category and age group (50-60 and 61-70 years) to analyse screening performance. Statistical analysis was performed to determine the association between density categories and age groups.…

BACKGROUND: Mammographic breast density (MBD) is an independent risk factor for breast cancer. Information regarding the relationship of MBD and breast cancer biology in women with ductal carcinoma in situ (DCIS) is currently lacking. This study aimed to examine the clinicopathologic characteristics of DCIS in women stratified by MBD.
METHODS: A retrospective review was performed to identify women with pure DCIS who underwent preoperative mammography between 2010 and 2018. Clinicopathologic and demographic data were collected. For the purpose of analysis, MBD was categorized as "non-dense" (B…

Ultimately, the incorporation of automated quantitative measures of breast density will lead to more effective clinical care and more robust outcomes research than the current, subjective assignment of Breast Imaging and Reporting Data System density categories, by providing reproducible estimates of both the risk of masking a cancer as well as the risk of developing breast cancer—two important factors in determining personalized breast cancer screening algorithms.

BACKGROUND: Fully automated assessment of mammographic density (MD), a biomarker of breast cancer risk, is being increasingly performed in screening settings. However, data on body mass index (BMI), a confounder of the MD-risk association, are not routinely collected at screening. We investigated whether the amount of fat in the breast, as captured by the amount of mammographic non-dense tissue seen on the mammographic image, can be used as a proxy for BMI when data on the latter are unavailable.
METHODS: Data from a UK case control study (numbers of cases/controls: 414/685) and a Norwegian c

There has been considerable recent interest in the genetic, biological and epidemiological basis of mammographic density (MD), and the search for causative links between MD and breast cancer (BC) risk. This report will critically review the current literature on MD and summarize the current evidence for its association with BC. Keywords 'mammographic dens*', 'dense mammary tissue' or 'percent dens*' were used to search the existing literature in English on PubMed and Medline. All reports were critically analyzed. The data were assigned to one of the following aspects of MD: general association

INTRODUCTION: It is debated whether mammographic density gives rise to more aggressive cancers. We therefore aimed to study the influence of mammographic density on prognosis.
METHODS: This is a case-only study within a population-based case-control study. Cases were all postmenopausal women in Sweden with incident breast cancer, diagnosed 1993-1995, and aged 50-74 years. Women with pre-diagnostic/diagnostic mammograms were included (n = 1774). Mammographic density of the unaffected breast was assessed using a computer-assisted thresholding technique. The Cox proportional hazards model was us

Breast density is a significant predictor in the risk of developing breast cancer. Several methods are available for assessing breast density, but most are subject to intra-observer variability and are unable to assess the breast as a three-dimensional structure. Using Quantra(™) to quantify breast density, we have correlated this with risk factors to determine what impact these variables have on breast density. Women attending for full field digital mammography at the South West London Breast Screening Unit between December 2008 and March 2009 were invited to participate in the study by quest…

Up to 50% of recurrences of ductal carcinoma in situ of the breast are associated with invasive carcinoma but no pathological or molecular features have yet been found to predict for the development of invasive disease. For a tumour to invade, it requires the formation of new blood vessels. Previous studies have described a vascular rim around ducts involved by ductal carcinoma in situ, raising the possibility that the characteristics of periductal vascularisation may be important in determining transformation from in situ to invasive disease. Periductal vascular density and phenotype were det
