Uptake of this isotopic tracer was essentially the same as in cells derived from salivary glands of adult animals, both in the absence and in the presence of 1 microM acetylcholine, 10 microM monensin or 10 microM A23187. Tracer accumulation in resting cells was inhibited in the immature cells by 1 mM amiloride but the inhibition was significantly smaller than in fully mature cells (16 and 33%, respectively). The response of sodium, potassium-adenosine triphosphatase (Na,K-ATPase) to cyclic adenosine monophosphate (cAMP)-dependent protein kinase was examined in membranes obtained from rabbit iris-ciliary body. Both Hg2+ and CH3Hg+ exert a depressive action on the twitch amplitude elicited by nerve stimulations more rapidly than that by muscle stimulation. Cells isolated by enzymatic digestion of gland fragments were incubated in solutions with or without Ca2+ or Na+ and exposed to monensin (10 microM) or acetylcholine (1 microM). Effects on accumulation of 22Na and on cell Ca2+ (measured with fura-2) were compared.
