Immunogenicity and Reactogenicity of SARS-CoV-2 Vaccines in Patients With Cancer: The CANVAX Cohort Study.

PURPOSE: The immunogenicity and reactogenicity of SARS-CoV-2 vaccines in patients with cancer are poorly understood. METHODS: We performed a prospective cohort study of adults with solid-organ or hematologic cancers to evaluate anti-SARS-CoV-2 immunoglobulin A/M/G spike antibodies, neutralization, and reactogenicity ≥ 7 days following two doses of mRNA-1273, BNT162b2, or one dose of Ad26.COV2.S. We analyzed responses by multivariate regression and included data from 1,638 healthy controls, previously reported, for comparison. RESULTS: Between April and July 2021, we enrolled 1,001 patients;

Poly(propylacrylic acid)-peptide nanoplexes as a platform for enhancing the immunogenicity of neoantigen cancer vaccines.

Cancer vaccines targeting patient-specific tumor neoantigens have recently emerged as a promising component of the rapidly expanding immunotherapeutic armamentarium. However, neoantigenic peptides typically elicit weak CD8+ T cell responses, and so there is a need for universally applicable vaccine delivery strategies to enhance the immunogenicity of these peptides. Ideally, such vaccines could also be rapidly fabricated using chemically synthesized peptide antigens customized to an individual patient. Here, we describe a strategy for simple and rapid packaging of peptide antigens into pH-resp…

BCG: A throwback from the stone age of vaccines opened the path for bladder cancer immunotherapy.

INTRODUCTION: It is 40 years since the initial documentation of the efficacy of bacille Calmette-Guérin (BCG) in the management of non-muscle invasive bladder cancer (NMIBC) and probably an opportune a time as any to retrace the origins of this development and to reflect on the progress that has occurred on the use of immune modifiers in the treatment of NMIBC. MATERIALS AND METHODS: A PubMed search for publications on the history of BCG was conducted, and those related to the development of the vaccine for protection against tuberculosis as well as those published in the last 40 years relate

Strategies for Developing Oral Vaccines for Human Papillomavirus (HPV) Induced Cancer using Nanoparticle mediated Delivery System.

Human Papillomaviruses (HPV) are a diverse group of small non-enveloped DNA viruses. Some HPVs are classified as low-risk as they are very rarely associated with neoplasia or cancer in the general population, and cause lenient warts. Other HPVs are considered as high-risk types because they are responsible for several important human cancers, including cervical cancer, a large proportion of other anogenital cancers, and a growing number of head and neck cancers. Transmission of HPV occurs primarily by skin-to-skin contact. The risk of contracting genital HPV infection and cervical cancer is in

Immunotherapy for bladder cancer using recombinant bacillus Calmette-Guerin DNA vaccines and interleukin-12 DNA vaccine.

PURPOSE: We investigated the efficacy of recombinant bacillus Calmette-Guerin (BCG) DNA (poly-rBCG) and murine interleukin (IL)-12 (mIL-12) vaccines in inducing T helper 1 polarized cytokines and suppressing bladder tumor growth in mice. MATERIALS AND METHODS: Four mycobacteria candidate genes (Ag85A, Ag85B, Mpt64 and PstS3) were cloned, fused with ESAT6 and ligated into eukaryotic expression vectors. Combined poly-rBCG and mIL-12 vaccines were transferred into a murine bladder tumor model. The efficiency of gene expression was detected using Western blotting, flow cytometry and semiquantitat

Bovine papillomavirus prostate cancer antigen virus-like particle vaccines are efficacious in advanced cancers in the TRAMP mouse spontaneous prostate cancer model.

Prostate cancer is a candidate for immunotherapy because cancer cells express tissue-specific proteins that can be therapeutic targets. However, immune checkpoint inhibitors and active immunization have performed poorly in clinical trials. We developed a novel virus-like particle (VLP) vaccine composed of bovine papillomavirus L1 protein engineered to display surface docking sites. We decorated VLPs with peptides encoding T cell epitopes from two prostate cancer-associated tumor antigens, prostate stem cell antigen (PSCA), and prostatic acid phosphatase (PAP-1 and PAP-2), and a neo-antigen, st

Systemic Transcriptional Responses to Clinical Vaccine Formulations and Live-Attenuated Vaccination: Comparative Kinetics and Hypotheses for Therapeutic Cancer Vaccine Monitoring.

BACKGROUND/OBJECTIVES: Therapeutic cancer vaccination requires innate sensing, antigen presentation, lymphocyte priming, tumor access, cytotoxicity, and persistence. Prophylactic studies cannot demonstrate these antitumor functions but can compare systemic formulation responses. We tested whether blood transcription extended beyond innate responses to cancer-immunity-cycle programs. METHODS: From 385 GEO records, we retained six human blood cohorts (five bulk-RNA and one sorted-cell) and one mouse blood/lymph-node study. We percentile-ranked 23 modules and compared baseline changes across 17

Pre-vaccine immune profiling in cancer patients identifies correlates of COVID-19 vaccine responses.

mRNA vaccines have been highly effective against SARS-CoV-2-related severe illness and are currently undergoing investigation as anti-cancer therapeutics. Our prior work has shown that patients undergoing cancer therapy have reduced immune responses to mRNA-based vaccines. Therefore, further investigation into immunologic responses in the setting of immune-altered hosts is warranted. In this study, we investigated pre-vaccination peripheral immune cell repertoire from a cohort of 66 patients with cancer on active treatment. Immune cell repertoire was characterized by mass cytometry to identify

Pre-vaccination transcriptomic profiles of immune responders to the MUC1 peptide vaccine for colon cancer prevention.

INTRODUCTION: Self-antigens abnormally expressed on tumors, such as MUC1, have been targeted by therapeutic cancer vaccines. We recently assessed in two clinical trials in a preventative setting whether immunity induced with a MUC1 peptide vaccine could reduce high colon cancer risk in individuals with a history of premalignant colon adenomas. In both trials, there were immune responders and non-responders to the vaccine. METHODS: Here we used PBMC pre-vaccination and 2 weeks after the first vaccine of responders and non-responders selected from both trials to identify early biomarkers of imm…

Papillomavirus vaccine types 16 and 18: new drug. Cervical cancer: just another vaccine.

The papillomavirus 16, 18 vaccine, like the papillomavirus 6, 11, 16, 18 vaccine, reduces the risk of high-grade cervical dysplasia due to all viral genotypes by about 40% in young women who are not yet infected at the time of vaccination. The vaccine does not seem to provoke any serious adverse effects, but there is no proof that it reduces the risk of cancer.

HPV vaccine: the key to a powerful shield against cancer.

BACKGROUND: Human papillomavirus (HPV) is a major global public health concern associated with multiple cancers. Despite the proven effectiveness of HPV vaccines, disparities in knowledge and vaccination uptake persist. This study aimed to assess HPV-related knowledge, awareness, and vaccination behaviors in a large population-based sample in Türkiye and to identify key determinants influencing vaccination. METHODS: This descriptive cross-sectional study included 12,093 individuals aged 18–49 from all seven geographical regions of Türkiye. Data were collected using a structured questionnaire a

mRNA Vaccines as Novel Immunotherapeutic Approaches in Gastrointestinal Cancers.

Messenger RNA vaccines have moved rapidly from the infectious-disease area into oncology. They now represent an attractive therapeutic option for gastrointestinal (GI) cancers. Personalised constructs, designed to elicit tumour-specific T-cell immunity, have shown acceptable safety and clear immunogenicity in early-phase trials of colorectal and pancreatic cancers. These early signals are encouraging, but several obstacles still exist for routine clinical use: tumour heterogeneity, immune evasion, the difficulty of selecting the right target antigens, and the stability of the delivery platform

COVID-19 Booster Vaccine Equity for Patients With Cancer.

COVID-19 has caused greater than 300 million documented infections worldwide including over 5 million confirmed deaths. Patients with cancer are particularly vulnerable due to a combination of disease and therapy-related effects. Available vaccines were highly effective against the original viral strains in clinical trials. However, initial vaccination efforts in this vulnerable population were impacted by federal policy that created substantial vaccine scarcity and allocation difficulties by recommending prioritization of unmanageably large patient populations including the entire elderly pop

COVID-19 vaccination in cancer patients: a narrative review.

Coronavirus disease 2019 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has affected over 220 million individuals worldwide, and has been shown to cause increased disease severity and mortality in patients with active cancer versus healthy individuals. Vaccination is important in reducing COVID-19-associated morbidity and mortality. Thus, the aim of this article was to review the existing knowledge on effectiveness, immunogenicity and safety of COVID-19 vaccines in patients with cancer. Fifty-four articles were included following a search of PubMed and Goog

Vaccine-induced immune thrombotic thrombocytopenia and patients with cancer.

Vaccines to combat SARS-CoV-2 infection and the COVID-19 pandemic were quickly developed due to significant and combined efforts by the scientific community, government agencies, and private sector pharmaceutical and biotechnology companies. Following vaccine development, which took less than a year to accomplish, randomized placebo controlled clinical trials enrolled almost 100,000 people, demonstrating efficacy and no major safety signals. Vaccination programs were started, but shortly thereafter a small number of patients with a constellation of findings including thrombosis in unusual loca

COVID-19 vaccination and breast cancer surgery timing.

There have been recent reports in the breast imaging literature of unilateral axillary lymphadenopathy following COVID-19 vaccination. It is unclear whether the reactive lymphadenopathy may impact the sentinel lymph node biopsy procedure. In this article, we provide guidelines regarding the timing of the COVID-19 vaccine and breast cancer surgery which were formulated after a review of the available literature and in consultation with infectious disease specialists.

[Preventing cancer: The role of Papillomavirus vaccination in the general population].

Papillomavirus (HPV), the first sexually transmitted disease in the world, is the main infectious agent responsible for cancer (6300 per year, in France). The cycle of HPV infection - >precancerous lesions - >cancer is well documented with regard to the cervix (cf. Nobel Prize in 2008). While this area is the most frequent (3000), it is far from being the only one. Other cancers include the anus, oropharyngeal sphere, glans and vulva. The sum of these other induced HPV cancers is greater than the total number of cervical cancers and also concerns boys. Screening is essential but insufficient a

HPV vaccination and the effects on rates of HPV-related cancers.

Globally, human papillomavirus (HPV) infection is one of the most common sexually transmitted infection. HPV is linked to at least five malignancies including vulvar, vaginal, anal penile, oropharyngeal, and cervical cancer. Three HPV vaccines are currently available: bivalent (HPV 16,18), quadrivalent (HPV 6,11,16,18), and nonavalent (6,11,16,18,31,33,45,52,58) targeting between 2 and 7 oncogenic HPV serotypes. This review highlights the currently epidemiologic burden of HPV-related cancers, efficacy of current HPV vaccines, and speculates about the benefits of widespread HPV vaccination. At
