Humanin attenuates neuronal cellular senescence and preserves neurotransmitter release in differentiated PC12 cells.

Cellular senescence, characterized as a permanent arrest of cell proliferation, is triggered by various types of stress. Senescent cells are identified by markers, including senescence-associated β-galactosidase (SA-β-gal) activity and the secretion of senescence-associated secretory phenotype (SASP) factors. These senescent phenotypes have been observed in neurons, suggesting that cellular senescence may contribute to the pathogenesis of neurodegenerative diseases. Humanin (HN), an endogenous short peptide, mitigates Alzheimer's disease-associated pathologies, including amyloid β-induced neur…

Cellular senescence in brain aging and neurodegeneration: from molecular mechanisms to translational opportunities.

Aging remains the predominant risk factor for Alzheimer's disease (AD) and other neurodegenerative disorders, yet the mechanisms linking systemic aging to brain dysfunction remain incompletely understood. Cellular senescence, a state of stable cell-cycle arrest coupled with metabolic and secretory reprogramming, has emerged as a pivotal and context-dependent driver of brain aging. Accumulation of senescent glial cells (astrocytes, microglia, and oligodendrocyte progenitors) and emerging evidence of "neurescence" in post-mitotic neurons contribute to neuroinflammation, impaired proteostasis, an…

Acylglycerol Kinase Sensitizes Glioblastoma to Temozolomide via Limiting Mitochondrial Damage Related Cellular Senescence.

Temozolomide (TMZ) is still the first-line drug for glioblastoma (GBM) treatment though tumor cell resistance remains a major challenge. TMZ administration may induce cellular senescence (CSEN), which exerts a dual regulatory effect on tumor progression, and evidence has suggested that CSEN is closely associated with mitochondrial dysfunction. Acylglycerol kinase (AGK), a mitochondrial membrane kinase, has been reported to participate in regulating mitochondrial function and the production of reactive oxygen species (ROS). Here, we aimed to investigate the role of AGK in GBM progression and TM…

O-GlcNAcylation in cellular senescence: From molecular insights to anti-cancer therapeutic opportunities.

Cellular senescence is a dynamic stress response with broad implications in aging, tissue homeostasis, and disease. In anti-cancer therapies, senescence acts as a double-edged sword, initially suppressing tumor growth but contributing to therapy tolerance and minimal residual disease, later leading to tumor repopulation, and relapse. While senescent cells are the seat of a large-scale metabolic reprogramming, the specific contribution of O-GlcNAcylation, a nutrient-sensitive post-translational modification involved in cancer progression and therapy resistance, was only recently investigated. H…

Phytochemical Investigation of Origanum calcaratum Extracts and in vitro Study against Cellular Senescence.

Origanum calcaratum is an endemic Greek species found in Cyclades, the Eastern Aegean Sea, and Northeastern Crete. The aim of this study was the thorough phytochemical analysis of O. calcaratum extracts and the in vitro assessment against cellular senescence. Overall, 27 constituents were identified in methanolic and hydromethanolic extracts by LC-MS and isolation techniques, predominantly phenolic compounds. Both extracts and rosmarinic acid showed noncytotoxicity to healthy cells. In Li-Fraumeni cells, they exhibited concentration-dependent senolytic activity, with rosmarinic acid and the hy…

Cellular senescence in posterior segment neovascular diseases: A proposed feed-forward amplification model.

Neovascular age-related macular degeneration (nAMD) and proliferative diabetic retinopathy (PDR) continue to be significant contributors to permanent visual impairment. Although anti-vascular endothelial growth factor (anti-VEGF) therapy has substantially improved disease management, recurrent neovascularization, persistent leakage, incomplete treatment response, subretinal fibrosis in nAMD, and fibrovascular membrane formation or tractional complications in PDR indicate that disease progression involves mechanisms beyond VEGF signaling alone. Cellular senescence-a stress-induced condition mar…

Intermittent fasting mitigates kidney fibrosis following ischemia-reperfusion injury by suppressing cellular senescence.

Cellular senescence is recognized as a key mechanism contributing to maladaptive renal recovery and the development of fibrosis after acute kidney injury (AKI). Intermittent fasting (IF), a non-pharmacological dietary strategy, influences aging pathways and tissue homeostasis; however, its contribution to kidney fibrosis is still not well characterized. This study aimed to evaluate how IF influences renal fibrosis and cellular senescence using a murine model of AKI resulting from ischemia-reperfusion injury (IRI). Male C57BL/6 mice underwent unilateral IRI, followed by assignment of the IF gro…

Curcumin nanoparticles alleviate brain mitochondrial dysfunction and cellular senescence in γ-irradiated rats.

Despite the diverse applications of γ radiation in radiotherapy, industrial processes, and sterilization, it causes hazardous effects on living organisms, such as cellular senescence, persistent cell cycle arrest, and mitochondrial dysfunction. This study evaluated the efficacy of curcumin nanoparticles (CNPs) in mitigating mitochondrial dysfunction and cellular senescence induced by γ radiation in rat brain tissues. Four groups of male Wistar albino rats (n = 8 per group) were included: (Gr1) the control group; (Gr2) the CNPs group (healthy rats receiving oral administration of curcumin nanop…

Therapeutic effects of PDGF-AB/BB against cellular senescence in human intervertebral disc.

Cellular senescence, characterized by a permanent state of cell cycle arrest and a secretory phenotype contributing to inflammation and tissue deterioration, has emerged as a target for age-related interventions. Accumulation of senescent cells is closely linked with intervertebral disc (IVD) degeneration, a prevalent age-dependent chronic disorder causing low back pain. Previous studies have highlighted that platelet-derived growth factor (PDGF) mitigated IVD degeneration through anti-apoptosis, anti-inflammation, and pro-anabolism. However, its impact on IVD cell senescence remains elusive. …

While previous studies have highlighted that platelet-derived growth factor (PDGF) mitigated IVD degeneration through anti-apoptotic and pro-anabolic effects, its impact on IVD cell senescence remains elusive. In this study, human NP and AF cells derived from aged, degenerated IVDs were treated with recombinant human (rh) PDGF-AB/BB for 5 d. Transcriptome profiling by mRNA sequencing revealed that NP and AF cells responded to the treat

A novel miRNA-TF-mRNA regulatory network associated with cellular senescence in osteoporosis.

This study investigated the role of miR-22-3p/ESR1 axis in osteoporosis (OP) pathogenesis. Bioinformatics analysis of OP datasets and patient bone marrow samples revealed significant upregulation of miR-22-3p accompanied by downregulation of ESR1. Mechanistic validation via dual-luciferase reporter assays, RNA pull-down, and molecular docking confirmed that miR-22-3p directly targets and suppresses ESR1 expression. Functional in vitro assays in human bone marrow mesenchymal stem cells (hBMSCs) demonstrated that miR-22-3p overexpression accelerated both cellular senescence (CS) and adipogenic d…

Age-Related Macular Degeneration: A Disease of Cellular Senescence and Dysregulated Immune Homeostasis.

Age-related macular degeneration (AMD) is a degenerative ocular disease primarily affecting central vision in the elderly. Its pathogenesis is complex, involving cellular senescence and immune homeostasis dysregulation. This review investigates the interaction between these two critical biological processes in AMD pathogenesis and their impact on disease progression. Initially, cellular senescence is analyzed, with particular emphasis on retinal damage induced by senescent retinal pigment epithelial cells. Subsequently, the occurrence of immune homeostasis dysregulation within the retina and i…

Emerging role of cellular senescence in normal lung development and perinatal lung injury.

Cellular senescence is a status of irreversible growth arrest, which can be triggered by the p53/p21cip1 and p16INK4/Rb pathways via intrinsic and external factors. Senescent cells are typically enlarged and flattened, and characterized by numerous molecular features. The latter consists of increased surfaceome, increased residual lysosomal activity at pH 6.0 (manifested by increased activity of senescence-associated beta-galactosidase [SA-β-gal]), senescence-associated mitochondrial dysfunction, cytoplasmic chromatin fragment, nuclear lamin b1 exclusion, telomere-associated foci, and the sene…

Cellular senescence mediates retinal ganglion cell survival regulation post-optic nerve crush injury.

Traumatic optic neuropathy refers to optic nerve (ON) injury by trauma, including explosion and traffic accident. Retinal ganglion cell (RGC) death is the critical pathological cause of irreversible visual impairment and blindness in ON injury. We previously investigated the patterns of 11 modes of cell death in mouse retina post-ON injury. Here we aimed to identify additional signalling pathways regulating RGC survival in rodents post-ON injury. RNA sequencing analysis identified the upregulation of inflammation and cellular senescence-related genes in retina post-ON injury, which were confir…

Bortezomib induces cellular senescence in A549 lung cancer cells by stimulating telomere shortening.

Bortezomib (BTZ) is a first-generation proteasome inhibitor with anti-tumor properties for multiple myeloma and mantle cell lymphoma. Increasing evidence has shown that BTZ exhibits toxic effects on diverse tumor cells, including non-small cell lung cancer (NSCLC) cells. However, the mechanism has not been fully evaluated. Here, we examined the regulatory effect of BTZ on cellular senescence, a potent tumor suppressive mechanism, in NSCLC cell lines. SA-β-gal staining assay showed that BTZ caused a significant increase in β-Gal positive A549 cells. BTZ also induced cell cycle arrest on G0/G1 p…

DNA damage response and GATA4 signaling in cellular senescence and aging-related pathology.

Aging is the continuous degradation of biological function and structure with time, and cellular senescence lies at its core. DNA damage response (DDR) can activate Ataxia telangiectasia-mutated serine/threonine kinase (ATM) and Rad3-related serine/threonine kinase (ATR), after which p53 activates p21, stopping the cell cycle and inducing cell senescence. GATA4 is a transcription factor that plays an important role in the development of many organs, such as the heart, testis, ovary, foregut, liver, and ventral pancreas. Studies have shown that GATA4 can also contribute to the DDR, leading to a…
