Perioperative hypothermia is common clinically. The neural mechanisms underlying general anesthesia and the hypothermia it induces remain elusive. We found that lower core temperature (Tcore) deepened anesthesia and delayed emergence, which showed a logarithmic relationship below 31.9°C. Chemogenetic activation of brain-derived neurotrophic factor in the lateral preoptic area (LPOBDNF) neurons before isoflurane administration lowered Tcore and increased isoflurane induced burst-suppression ratio (BSR), δ power, and recovery time, while its optogenetic activation during isoflurane did not chang

Vasoactive intestinal peptide (VIP)-expressing neurons within the motor cortex (MC) exhibit diurnal variation in excitability and may participate in the modulation of sleep architecture. In this study, we found that MCVIP neurons displayed significantly higher excitability during the light phase. Using chemogenetic approaches, we examined their contribution to sleep-wake modulation. Given the sleep-modulating actions of the α2-adrenergic receptor agonist dexmedetomidine (Dex), we further tested whether Dex engages MCVIP neurons. Dex increased MCVIP neuronal excitability in slice recordings in

Alprazolam (Alp) is a clinically common benzodiazepine (BZD) with a potential risk of abuse, yet the mechanisms underlying its reinstatement remain unclear. In this study, a conditioned place preference (CPP) paradigm combined with forced swim stress (FSS) was employed to induce CPP reinstatement in male mice with prior Alp exposure. The results demonstrated that the activity of CaMKIIα neurons in the ventromedial hypothalamus (VMH) was significantly increased following FSS-induced reinstatement of Alp-CPP. The lateral hypothalamus (LH) receives innervation from the VMH, and GABAergic neurons

Reactive astrocytes are associated with neuroinflammation and cognitive decline in diverse neuropathologies; however, the underlying mechanisms are unclear. We used optogenetic and chemogenetic tools to identify the crucial roles of the hippocampal CA1 astrocytes in cognitive decline. Our results showed that repeated optogenetic stimulation of the hippocampal CA1 astrocytes induced cognitive impairment in mice and decreased synaptic long-term potentiation (LTP), which was accompanied by the appearance of inflammatory astrocytes. Mechanistic studies conducted using knockout animal models and hi

The medial prefrontal cortex (mPFC) plays a pivotal role in attention by exerting top-down control to allocate cognitive resources toward behaviorally relevant stimuli based on learned context and expectations. mPFC neurons project to multiple cortical and subcortical regions, including the locus coeruleus (LC)-the brain's primary source of norepinephrine (NE). The mPFC also receives inputs from the LC, which release NE to modulate mPFC neuronal activity and downstream cellular signaling. While enhanced functional connectivity between the mPFC and LC in mice during sustained attention tasks su

Precise control of protein function in living cells is essential for engineering programmable biological systems and therapeutic applications. However, most current strategies act indirectly by altering protein stability, localization, or proximity rather than directly modulating binding activity. Here we present VIPbodies, nanobodies engineered with self-cleaving viral proteases that convert protease inhibition into drug-dependent antigen recognition. Using anti-mCherry nanobodies as prototypes, we create variants responsive to orthogonal viral protease-inhibitor pairs and extend the design t

The advent of midazolam holds profound implications for modern clinical practice. The hypnotic and sedative effects of midazolam afford it broad clinical applicability. However, the specific mechanisms underlying the modulation of altered consciousness by midazolam remain elusive. Herein, using pharmacology, optogenetics, chemogenetics, fiber photometry, and gene knockdown, this in vivo research revealed the role of locus coeruleus (LC)-ventrolateral preoptic nucleus noradrenergic neural circuit in regulating midazolam-induced altered consciousness. This effect was mediated by α1 adrenergic re…

Ventral hippocampus (vHIP) and medial prefrontal cortex (mPFC) are both critical regions for social behaviors. However, how their interactions affect social behavior is not well understood. By viral tracing, optogenetics, chemogenetics, and fiber photometry, we demonstrated that inhibition of vHIP or direct projections from vHIP to mPFC impaired social memory expression. Via rabies retrograde tracing, we found that all three major GABAergic neurons in mPFC received direct inputs from vHIP. Activation of parvalbumin positive (PV+) neurons in mPFC but not somatostatin positive (SST+) neurons can…

Insomnia and anxiety are two common clinical diseases that threaten people's physical and mental health. Insomnia and anxiety may share some similar underlying neural circuit mechanisms in the brain. In this study, we combine techniques including chemo-fMRI, optogenetics, and chemogenetics to reveal that the glutamatergic neurons of the paraventricular hypothalamic nucleus (PVN) regulate both anxiety and arousal through two different downstream neural circuits. Optogenetic activation of the PVN-cingulate cortex (Cg) neural circuit triggers anxiety-like behaviors in mice without affecting the w…

Central cholinergic system plays a vital role in the pathogenesis of major depressive disorder. The cholinergic neurons in laterodorsal tegmental nucleus (LDTg) are the key components of the cholinergic system in brainstem. However, how the LDTg cholinergic neuronal activities are modulated in response to chronic stress remains largely unclear. We reported here that local inhibitory inputs from somatostatin-positive (SST+) interneurons modulated the cholinergic neuronal activities in the LDTg (LDTgSST-ChAT) and adjusted susceptibility to chronic stress in mice. Chronic unpredictable mild stres

The mesolimbic dopamine (DA) system has been implicated in pair bond formation. However, the involvements of DA release, real-time activities, and electrophysiological activities of D1/D2 medium spiny neurons (MSNs) in the nucleus accumbens (NAc) shell in pair bonding remain unclear. This work verified that male mandarin voles after pair bonding released higher levels of DA in the NAc shell and displayed higher levels of D1 MSNs activity and lower levels of D2 MSNs activity upon sniffing their partners compared to upon sniffing an unknown female. Moreover, pair bonding induced differential alt

Chronic visceral pain is frequently accompanied by changes in food preference. The paraventricular hypothalamus (PVH) and the anterior cingulate cortex (ACC) are well-known regions involved in pain processing and food intake. However, the underlying neural circuitry mechanisms remain unclear. Here, we showed that a circuit from cholecystokinin neurons in the PVH (PVHCCK) projecting to glutamatergic neurons in the ACC (ACCGlu) to regulate food preference in male mice with chronic visceral pain induced by neonatal colonic inflammation (NCI). The brain and immune system communicate through complex bidirectional pathways, but the specificity by which the brain perceives or even remembers alterations in immune homeostasis is still poorly understood. Recent data revealed that immune-related information under peripheral inflammatory conditions, termed as "immunengram", were represented in specific neuronal ensembles in the insular cortex (IC). Chemogenetic reactivation of these neuronal ensembles was sufficient to retrieve the inflammatory stages, indicating that the brain can store and retrieve specific immune responses. Against this

Hypersomnolence disorder (HD) is characterized by excessive sleep, which is a common sequela following stroke, infection, or tumorigenesis. HD is traditionally thought to be associated with lesions of wake-promoting nuclei. However, lesions of a single wake-promoting nucleus, or even two simultaneously, did not exert serious HD. Therefore, the specific nucleus and neural circuitry for HD remain unknown. Here, we observed that the paraventricular nucleus of the hypothalamus (PVH) exhibited higher c-fos expression during the active period (23:00) than during the inactive period (11:00) in mice.

The comorbidity of chronic pain and attention deficit complicates treatment, as each condition intensifies the other through mechanisms that … rbated hypersensitivity and attention deficit-like behaviors in CCI mice. Through neuronal tracing, chemogenetics, optogenetics, and electrophysiology, we discovered a monosynaptic dopaminergic pathway from the locus coeruleus to the thalamic reticular nucleus, influencing pain modulation in naive mice. In CCI mice, both projections were enhanced, and activatio …

Perioperative neurocognitive disorders (PND) contribute substantially to morbidity and mortality; however, no therapeutic target has yet been identified. We hypothesized that the neural circuit underlying this surgical complication involves corticotropin-releasing hormone (CRH) neurons in the paraventricular nucleus of the hypothalamus (PVN). Using optogenetics, chemogenetics, electrophysiology, and behavioral tests, we demonstrate that surgery activates PVN CRH neurons. These neurons, via CRH receptor 1 (CRHR1), innervate glutamatergic (Glu) neurons in the hippocampal subiculum (Sub), ultimat…

The full mechanism of action of propofol, a commonly administered intravenous anesthetic drug in clinical practice, remains elusive. The focus of this study was the role of GABAergic neurons which are the main neuron group in the ventral pallidum (VP) closely associated with anesthetic effects in propofol anesthesia. The activity of VP GABAergic neurons following propofol anesthesia in Vgat-Cre mice was observed via detecting c-Fos immunoreactivity by immunofluorescence and western blotting. Subsequently, chemogenetic techniques were employed in Vgat-Cre mice to regulate the activity of VP GAB

Migraine is commonly reported in patients with temporomandibular disorders (TMDs), but little is known about the mechanisms underlying the comorbid condition. Here, we prepared a mouse model to investigate this comorbidity, in which masseter muscle tendon ligation (MMTL) was performed to induce a myogenic TMD, and the pre-existing TMD enabled a subthreshold dose of nitroglycerin (NTG) to produce migraine-like pain in mice. RNA sequencing followed by real-time quantitative polymerase chain reaction confirmation showed that MMTL plus NTG treatment increased prodynorphin ( Pdyn ) mRNA expression

The prelimbic cortex (PL) and infralimbic cortex (IL) play a role in context-induced reinstatement of heroin seeking in an animal model of drug relapse. Both the PL and IL receive direct glutamatergic projections from the ventral CA1 (vCA1), which is also involved in context-induced reinstatement of cocaine and heroin seeking. Here we studied the role of vCA1-PL and vCA1-IL projections in context-induced reinstatement of heroin seeking by using electrophysiological, neuropharmacological, chemogenetic, and molecular methods. We showed that context-induced reinstatement of heroin seeking caused
