Eosinophil colony-stimulating factor induced by administration of interleukin-2 into the pleural cavity of patients with malignant pleurisy.

Systemic administration of interleukin (IL)-2 to patients with malignant diseases induces peripheral eosinophilia. In the present study, to … eurisy is due to the Eo-CSF activities of various components, including IL-5, IL-3, and GM-CSF, and chemotactic factors for eosinophils induced locally in the pleural cavity by IL-2.

Generation of leukotrienes from normal epidermis and their demonstration in cutaneous disease.

In order to evaluate the significance of chemotactic leukotrienes in cutaneous disease, synthetic leukotriene B4 and its metabolites were examined during in vitro chemotaxis. Leukotriene B4, and less so 20-OH-leukotriene B4, were chemotactic for neutrophils and eosinophils, with a preferential attraction of eosinophils. The responsiveness of human monocytes towards leukotriene B4 was relatively low. Normal cells and cells from different patients varied in their quantitative response. Cells from patients with eczema and T-cell lymphoma tended to migrate less than those from patients with other

Severe Fletcher factor (plasma prekallikrein) deficiency with partial deficiency of Hageman factor (factor XII): report of a case with observation on in vivo and in vitro leukocyte chemotaxis.

A case of cross-reacting material-negative Fletcher trait with additional partial deficiency of Hageman factor (HF, Factor XII) is described. Although the patient presented with a recent history of frequent epistaxis, he had no other personal or family history of a tendency toward bleeding or infection. Similar to other cases of Fletcher trait, his plasma showed a markedly prolonged partial thromboplastin time which could be corrected by prolonged incubation with the surface-activator kaolin. Surface-induced fibrinolysis, amidolysis of alpha-N-benzoyl-proline-L-phenylalanine-L-arginine-p-nitro

[Pyoderma gangrenosum and chronic myeloïd leukemia. Two new cases (author's transl)].

Two new cases of pyoderma gangrenosum with chronic myeloid leukemia are described and added to three others of the literature. Pyoderma gangrenosum is often bullous, painful and progresses quickly. It seems to have a bad prognosis in myeloid leukemia as in polycythemia vera, in which it appears lately as an evolution to myelofibrosis or acute leukemia transformation. Chemotactic disorders of polynuclear may be the main etiological factor in these cases.

Cell-body curvature modulates stall frequency to enhance Vibrio cholerae swimming and chemotaxis through hydrogels.

The swimming motility of Vibrio cholerae is a virulence factor that aids in breaching the mucus layer of the small intestine. V. cholerae cells have a curved cell shape and previous work demonstrated that loss of curvature decreases infectivity. Here we investigate the mechanism by which V. cholerae's curvature affects single-cell motility within mucus-mimicking environments. Using a multiscale chemotaxis assay, we compared the chemotactic performance of wild-type curved cells (O1 El Tor C6706) and straight mutants under linear chemical gradients in liquid solutions, viscous solutions, and sof

Dynamics of a protective avian inflammatory response: the role of an IL-8-like cytokine in the recruitment of heterophils to the site of organ invasion by Salmonella enteritidis.

Increased resistance to Salmonella enteritidis (SE) organ infectivity in chickens can be conferred by the prophylactic administration of SE-immune lymphokines (ILK). Resistance is associated with an enhanced heterophilic accumulation within 4 h of ILK injection. In these studies, the role of IL-8 in ILK-mediated heterophil recruitment during SE infections in young chickens was investigated. Heterophil accumulation was enhanced 2-4 h after the i.p. injection of both ILK and SE (ILK/SE) when compared to the control chicks. An i.p. injection of a rabbit polyclonal anti-human IL-8 antibody signifi

Histamine receptor 2-mediated growth-differentiation factor-15 expression is involved in histamine-induced melanogenesis.

Vitiligo is a progressive depigmenting disorder. Histamine has been shown to induce melanogenesis via histamine receptor 2, suggesting the possibility of histamine as a repigmenting agent for the treatment of vitiligo. However, the role and signaling mechanism of histamine are still unclear in melanogenesis, especially in relation to growth-differentiation factor-15, which is a protein belonging to transforming growth factor beta and found to be overexpressed in metastatic or malignant melanoma. We found that histamine induces growth-differentiation factor-15 in melanoma cell lines such as SK-

β-Defensin 19/119 mediates sperm chemotaxis and is associated with idiopathic infertility.

Sperm chemotaxis is required for guiding sperm toward the egg. However, the molecular identity of physiological chemoattractant and its involvement in infertility remain elusive. Here, we identify DEFB19/119 (mouse/human orthologs) as a physiological sperm chemoattractant. The epithelia of the female reproductive tract and the cumulus-oocyte complex secrete DEFB19/119 that elicits calcium mobilization via the CatSper channel and induces sperm chemotaxis in capacitated sperm. Manipulating the level of DEFB19 in mice determines the number of sperm arriving at the fertilization site. Importantly,

Regulatory mechanisms of neutrophil migration from the circulation to the airspace.

The neutrophil, a short-lived effector leukocyte of the innate immune system best known for its proteases and other degradative cargo, has unique, reciprocal physiological interactions with the lung. During health, large numbers of 'marginated' neutrophils reside within the pulmonary vasculature, where they patrol the endothelial surface for pathogens and complete their life cycle. Upon respiratory infection, rapid and sustained recruitment of neutrophils through the endothelial barrier, across the extravascular pulmonary interstitium, and again through the respiratory epithelium into the airs

Mosquito attractant blends to trap host seeking Aedes aegypti.

Aedes aegypti is the key vector of three important arboviral diseases -dengue, yellow fever and chikungunya. To identify volatile chemicals which could be used in odour based traps for Aedes mosquito surveillance, a few synthetic compounds and compound blends have been evaluated in an indigenously designed olfactometer. A total of 24 compounds and seven compound blends were screened against unfed adult female Ae. aegypti mosquitoes for attraction and compared with control group. The attractancy or repellency index of the test material to mosquitoes was calculated and rated them as class-1, cla

[How is the sense of smell connected? Cellular and molecular mechanisms guiding the development of the synaptic connections from the nose to the cortex (I)].

The physiological particularities that occur during the development of the olfactory system make it one of the most fascinating parts of the central nervous system and one of models that has been most widely studied in order to understand the mechanisms related with axonal growth and guidance towards the right targets. A variety of mechanisms are known, some mediated by contact (laminins, cell adhesion molecules, ephrins, etc.) and others that are secreted (semaphorins, slits, growth factors, etc.), to play diverse roles in establishing the synaptic interactions among the olfactory epithelium,

Immunohistochemical localization of eotaxin immunoreactivity in nasal polyps.

Eotaxin is a C-C chemokine that acts to selectively induce local accumulation of eosinophils and basophils. Eotaxin is also believed to be involved in the infiltration of eosinophils in the nasal polyps of patients with chronic sinusitis. However, only a few studies on eotaxin in nasal polyps have been performed. In this study, we investigated the localization of eotaxin in human nasal polyps and the identification of eotaxin-positive cells using immunohistochemistry. The distribution of eotaxin immunoreactivity in the nasal polyps of patients with chronic sinusitis was found to almost coincid

Lesional modulation of peripheral monocyte leucotactic responsiveness in leprosy.

Because the accumulation and activation of mononuclear phagocytes are critical to the host response to intracellular microbial pathogens, we evaluated mechanisms of peripheral monocyte leucotactic regulation in leprosy. Plasma from 53 of 67 patients was found to inhibit the locomotion of normal human monocytes. Neither the prevalence nor the magnitude of plasma leucotactic inhibitory activity correlated with disease histology or duration, type or duration of chemotherapy, or history of erythema nodosum leprosum. Plasma leucotactic inhibitory activity resided principally in a non-immunoglobulin

Mediators of hypersensitivity in exercise-induced asthma.

Neutrophil chemotactic factor (NCF) is a slightly acidic macromolecule which is released into the circulation of asthmatic individuals following an exercise task. Its appearance was basophil-independent, accompanied the development of airflow obstruction and its release could be inhibited by administration of disodium cromoglycate. There was no elaboration of NCF after isocapnic hyperventilation with cold air, although the same asthmatics produced NCF following treadmill exercise. Finally, NCF was also released during exercise-induced late asthmatic reactions, as in antigen-induced late respon

Chemotaxins in cotton mill dust: possible etiologic agent(s) in byssinosis.

Through the application of a simplified in vitro assay for chemotaxis of polymorphonuclear leukocytes, potent chemotaxins have been found to exist in cotton mill dust extracts. These substances may be involved in the recruitment of cells to pulmonary surfaces and in the etiology of byssinosis in textile workers who inhale cotton dust. The chemotaxins were extracted from cotton dust with alkaline aqueous solution and purified by chromatography on paper and Sephadex G-25. The active agents were readily water-soluble, stable to heat at 100 degrees C in water, inactivated by hydrolytic conditions,

Pregnant human peripheral leukocyte migration during several late pregnancy clinical conditions: a cross-sectional observational study.

BACKGROUND: Parturition at term and preterm is characterized by sterile inflammatory processes occurring in the absence of infection whereby peripheral leukocytes infiltrate gestational tissues in response to chemotactic signals. In response to a homing signal, recruited leukocytes undergo diapedesis and extravasate through capillaries, migrating into stromal tissue. There they interact with resident immune and stromal cells to produce a mixture of matrix metalloproteinases, prostaglandins and cytokines including interleukin-1β (IL-1β) and IL-6 that in turn transform the uterus from pregnancy

Alterations of macrophage functions by mediators from lymphocytes.

Sensitized lymphocytes were incubated in vitro with the specific antigen Supernatants from these cultures were chromatographed on Sephadex G-100 columns. Supernatant fractions containing MIF, chemotactic factor, and lymphotoxin, but free of antigen and antibody, were incubated with normal peritoneal exudate macrophages. Macrophage adherence, phagocytosis, spreading, motility, and direct hexose monophosphate oxidation were enhanced, while protein synthesis was unaffected. Thus, antigen-stimulated lymphocytes secrete a factor or factors which enhance certain macrophage functions. Implications fo
