A therapy for recurrent erosions of the cornea with trypsin-chymotrypsin ointment is described. Impressed by the efficiency of the digestive enzymes, a theory on the development of recurrent erosions is presented. In cases of recurrent erosions, Bowman's membrane becomes coated with a fine layer of proteinic material which prevents a firm adhesion of the new epithelium on the membrane. Therefore blisters can form. When the surface of the membrane is cleaned by the digestive enzymes, the advancing epithelium will firmly adhere to its base.

A proteolytic enzyme mixture, Orenzyme-Forte, containing trypsin, chymotrypsin, and ribonuclease was used to treat experimentally induced pulpitis in the teeth of three monkeys. Assessment by means of the criteria of Stanley showed that the enzyme mixture induced better healing than a zinc oxide--eugenol dressing. When the two were used simultaneously, a synergistic effect was seen. We suggest that this enzyme mixture may be useful in clinical practice in the treatment of pulpitis.

Bovine heart 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (PFK-2/FBPase-2) was phosphorylated by incubation with [gamma-32P]MgATP and cyclic AMP-dependent protein kinase (PKA) or protein kinase C (PKC). After digestion with chymotrypsin, the phosphorylation sites for the two protein kinases were identified by peptide mapping, and microsequencing. Evidence for new phosphorylation sites for PKA (Ser-483) and PKC (Ser-84 and Ser-466) was obtained.

Derivatives of benzo[b]thiophene-2-sulfonamide were prepared to investigate their potential utility as topically active inhibitors of ocular carbonic anhydrase. Such an agent would be useful in the treatment of glaucoma. Among the compounds described are 6-hydroxybenzo[b]thiophene-2-sulfonamide (16) and its acetate ester (23), which are among the most potent ocular hypotensive agents in this class, as assessed in the alpha-chymotrypsinized rabbit. The natural proteasome inhibitor salinosporamide A from the marine bacterium Salinispora tropica is a promising drug candidate for the treatment of multiple myeloma and mantle cell lymphoma. Using a comprehensive approach that combined chemical synthesis with metabolic engineering, we generated a series of salinosporamide analogues with altered proteasome binding affinity. One of the engineered compounds is equipotent to salinosporamide A in inhibition of the chymotrypsin-like activity of the proteasome yet exhibits superior activity in the cell-based HCT-116 assay.

This work describes a perfusion technique adapted to the isolated rabbit pancreas allowing investigation of both the endocrine and exocrine function. The pancreas-duodenum-stomach-spleen complex is removed and perfused with a modified Krebs Ringer Bicarbonate medium. The surgical steps leading to the removal of the complex are described. The endocrine response is studied by measuring insulin release when the pancreas is submitted to successive glucose stimulations and the exocrine function is evaluated by the alpha-chymotrypsin activity of the pancreatic juice harvested during the perfusion.
