Nile Tilapia (Oreochromis niloticus) Patched1 Mutations Disrupt Cardiovascular Development and Vascular Integrity through Smoothened Signaling.

Hedgehog (Hh) signaling is crucial in cardiovascular development and maintenance. However, the biological role of Patched1 (Ptch1), an inhibitory receptor of the Hh signaling pathway, remains elusive. In this study, a Ptch1 ortholog was characterized in Nile tilapia (Oreochromis niloticus), and its function was investigated through CRISPR/Cas9 gene knockout. When one-cell embryos were injected with CRISPR/Cas9 targeting ptch1, the mutation efficiency exceeded 70%. During 0-3 days post fertilization (dpf), no significant differences were observed between the ptch1 mutant group and the control g

Haemato-biochemical, mutagenic, and histopathological changes in Oreochromis niloticus exposed to BTX.

The study of the DNA damage response in erythrocytes after exposure to volatile organic compounds (VOCs) can present evidence for its potential effect as genotoxic- biomarkers for environmental pollution. Although VOCs are dangerous pollutants, still little is known about hemotoxic, cytotoxic, and genotoxic effects of such pollutants on fish. We optimized an assay method for apoptosis and DNA damage in erythrocytes of adult tilapia fish after 15 days exposure to benzene (0.762 ng/L), toluene (26.614 ng/L), and xylene (89.403 ng/L). The highest level of apoptosis and DNA damage were recorded in

Erythrocyte Recovery in Oreochromis niloticus Fish Exposed to Urban Effluents.

Urban activities pollute aquatic ecosystems, and the integrity of organisms such as fish. The use of cytological techniques, such as the analysis of blood cellular integrity using the Micronucleus test, can help detect mutagenic damage as a result to urban effluents exposure. In this context, this study aimed to evaluate the frequency of micronucleus and other nuclear abnormalities in Oreochromis niloticus fish environmentally exposed to urban effluents in relation to their erythrocyte recovery capacity when exposed to clean water (30 and 45 days). The results indicated high copper, dissolved

Isolation of potential zoonotic Mycobacterium spp. from diseased freshwater angelfish (Pterophyllum scalare) from an aquarium.

Nontuberculous mycobacteria infection is one of the most common chronic bacterial diseases in ornamental aquarium fish and appears to be directly related to stressful husbandry practices. Furthermore, it also represents zoonotic potential. Here we present the isolation and characterization of non-tuberculous mycobacteria from diseased freshwater angelfish (Pterophyllum scalare) in São Paulo, Brazil. Nine discarded breeding females with signs of disease were evaluated. The fish exhibited lethargy, loss of appetite, cachexia, skin ulcers, and exophthalmia. At necropsy, four fishes presented macr

First report of monogenean flatworms from Lake Tana, Ethiopia: gill parasites of the commercially important Clarias gariepinus (Teleostei: Clariidae) and Oreochromis niloticus tana (Teleostei: Cichlidae).

BACKGROUND: Lake Tana is the largest lake in Ethiopia and the source of the Blue Nile. The lake harbours unique endemic cyprinid fish species, as well as the commercially important endemic Nile tilapia subspecies Oreochromis niloticus tana and the North African catfish Clarias gariepinus. Its endemicity, especially within the Labeobarbus radiation, its conservation importance and its economic indispensability attract scientific interest to the lake's ichthyofauna. Fish parasites of Lake Tana, however, are hitherto poorly known, and no formal report exists on its monogenean flatworms. For susta

Philometra mirabilis sp. n. (Nematoda: Philometridae), a new gonad-infecting parasite from the freshwater fish Cichla mirianae (Cichlidae) in Brazilian Amazon.

A new nematode species, Philometra mirabilis sp. n. (Philometridae), is described based on a subgravid female specimen recovered from the ovary of the freshwater perciform fish Cichla mirianae Kullander and Ferreira (Cichlidae) in the Juruena River (Amazon River basin), State of Mato Grosso, Brazil. The new species is morphologically very different from congeners parasitizing fishes in South America, being mainly characterized by the markedly elongate, narrow body 171 mm long (maximum width/body length 1:598), the presence of three small cone-shaped oesophageal teeth protruding out of the mout

The opioid peptide β-endorphin interferes with the pituitary-testis axis in the Mozambique tilapia Oreochromis mossambicus.

Although the involvement of β-endorphin (β-ERP) in vertebrate reproduction has been suggested, its role in testicular activity is not clear in fish. We describe the influence of β-ERP on spermatogenesis in a cichlid fish in the present paper. In comparison to the control group, the administration of β-ERP (3 µg) caused a significant increase in the number of spermatogonia-A and spermatids. Following treatment with β-ERP (6 µg), a significant increase in the number of spermatogonia-A was observed, whereas the numbers of all the other germ cells, excluding spermatogonia-B, significantly decrease

Histology of the utricle in kinetotically swimming fish: a parabolic aircraft flight study.

OBJECTIVES: Humans taking part in parabolic aircraft flights (PAFs) may suffer from space motion sickness, which is a form of kinetosis. As it has been repeatedly shown that some fish in a given batch also reveal kinetotic behaviour (especially so-called spinning movements and looping responses) during PAFs, and as a result of the homology of the vestibular apparatus of all vertebrates, fish can be used as model systems to investigate the origin of susceptibility to motion sickness. Therefore. we were prompted to examine the utricular maculae, which are responsible for the internalization of g

Multiple corticosteroid receptors in a teleost fish: distinct sequences, expression patterns, and transcriptional activities.

Corticosteroid hormones, including the mineralocorticoids and the glucocorticoids, regulate diverse physiological functions in vertebrates. These hormones act through two classes of corticosteroid receptors (CR) that are ligand-dependent transcription factors: type I or mineralocorticoid receptor (MR) and type II or glucocorticoid receptor (GR). There is substantial overlap in the binding of these two receptor types to hormones and to DNA. In fish, the overlap in processes controlled by CRs may be different from that in other vertebrates, as fish are thought to synthesize only glucocorticoids,

Therapeutic potential of bioactive peptides against Lactococcus petauri in Nile tilapia.

Lactococcus petauri is an emerging pathogen causing piscine lactococcosis in various aquatic species. This disease has been increasingly reported as a cause of mortality in aquaculture. Due to the indiscriminate use of antimicrobials and to promote viable alternatives for their replacement, bioactive peptides with antimicrobial properties can be explored as tools against bacterial infections. Thus, the objective of this study was to identify the pathogen responsible for a mortality outbreak in cage-cultured Nile tilapia (Oreochromis niloticus), assess its pathogenicity, and evaluate the therap

A Multimodal Approach to Identify Metallothionein Metal Inducers in Nile Tilapia: Insights From Molecular Docking and Hepatocyte Exposure.

Many human activities contribute to the pollution of aquatic ecosystems, primarily through agricultural, industrial, mining, and domestic discharges into water bodies. Fish, being highly sensitive to environmental changes, serve as valuable models for monitoring the health of these ecosystems. Metallothionein (Mt), a biomarker for metal contamination, shows variable expression depending on the metal involved. Transcription of the mt gene is regulated by intracellular metal concentrations and mediated by interactions between metal-responsive transcription factors (Mtf) and metal response elemen
