Hijacking of host PCNA by circovirus replication-associated protein to recruit POLD1 drives viral DNA replication and is inhibited by R428.

Circoviruses are the smallest known mammalian viruses with a single-stranded circular genome, whose replication capacity is highly dependent on host factors. Although the virus-encoded replication-associated protein (Rep/Rep') plays a critical role in circovirus DNA replication, it lacks polymerase activity. Here, we uncovered a mechanism by which circoviruses hijack the host proliferating cell nuclear antigen (PCNA) via their Rep to recruit POLD1, the catalytic subunit of host DNA polymerase δ, thereby driving viral DNA synthesis. POLD1 was identified to be the major polymerase in the replica

Studies on syndemic infection of porcine reproductive and respiratory syndrome virus with porcine circovirus 2 in backyard pigs of Mizoram, India.

Porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus 2 (PCV2) are among one of the most prevalent and important infectious agents, which cause a severe economic impact on pig farming worldwide. Circulation of these viruses together in the same pig population may lead to the syndemic infection with altered pathogenicity and thereby pathology and clinical manifestation of the diseases. The present study evaluated syndemic infection between PRRSV and PCV2 in the pig population reared in the backyard farms in Mizoram state of India. The syndemicity between PRRSV and P

The replication protein of duck circovirus unwinds dsDNA in an ATP-driven and metal ion-dependent manner from 3' to 5'.

Duck circovirus (DuCV) causes immunosuppression, co-infection and increased mortality in ducks, and the prevalence and infection rate of DuCV have risen in recent years, resulting in significant economic losses to the duck industry. Studying the mechanism of virus replication is particularly important for controlling DuCV infection. The replication protein (Rep) encoded by the DuCV genome is a highly conserved enzyme and plays key roles in viral replication, making it an ideal target for antiviral drugs. However, the biochemical characteristics of DuCV Rep were still unknown. Here, we utilized

[Performance parameters and pathogen detection in pig groups differently vaccinated with respect to Porcine Circovirus Type 2 and Mycoplasma hyopneumoniae].

OBJECTIVE: Respiratory diseases, mostly multifactorial, cause problems in pig farms worldwide. Next to infectious agents, such as Porcine Circovirus Type 2 (PCV2) and Mycoplasma hyopneumoniae (M. hyopneumoniae) management, housing, and environmental factors are decisive for the development of disease. In a conventional, closed swine farm in Lower Saxony, Germany, which did not vaccinate against PCV2, the effect of an implementation of PCV2 vaccination (Suvaxyn® Circo + MH RTU) onto animal health was evaluated. In addition, the effect of this combination vaccine was assessed in comparison to si

Epidemiological investigation of porcine circovirus type 2 and its coinfection rate in Shandong province in China from 2015 to 2018.

BACKGROUND: Porcine circovirus type 2 (PCV2) is one of the crucial swine viral pathogens, caused porcine circovirus associated diseases (PCVAD). Shandong province is one of the most important pork producing areas and bears a considerable economic loss due to PCVAD. However, there is limited information on epidemiology and coinfection rate of PCV2 with other critical swine diseases in this area, such as porcine reproductive and respiratory syndrome virus (PRRSV), classical swine fever virus (CSFV), Pseudorabies virus (PRV), and porcine epidemic diarrhea virus (PEDV). RESULTS: Overall, 89.59% s

Coinfection with porcine circovirus type 2 and Streptococcus suis serotype 2 enhances pathogenicity by dysregulation of the immune responses in piglets.

Porcine circovirus type 2 (PCV-2) and Streptococcus suis (S. suis) are common pathogens in pigs. Both pathogens are associated with the porcine respiratory disease complex. Clinically, coinfection of PCV-2 and S. suis are often detected in pigs with respiratory symptoms, while interactions between the two pathogens during coinfection and the coinfection pathogenesis are poorly understood. In this study, a piglet model coinfected with PCV-2 and Streptococcus suis serotype 2 (SS2) was established; coinfection of piglets increased the contents of SS2 in blood, and piglets showed more severe pneum

Cytological Evidence of Telocyte Involvement in Skin Immune Regulation Following Jet Needle-Free Injection of an Inactivated Porcine Circovirus Vaccine.

A recently identified mesenchymal cell population, telocytes (TCs), has been found in many tissues of different animal species. Jet needle-free injection (JNFI) is a promising non-invasive drug-delivery method that can trigger effective immune responses in the skin. In this preliminary morphological study, an inactivated porcine circovirus vaccine was delivered into pig neck skin by JNFI, and untreated normal neck skin served as the control. Histopathology, immunohistochemistry (IHC), immunofluorescence (IF), and transmission electron microscopy (TEM) were used to evaluate TC distribution, ult

The course of pigeon circovirus infection in young pigeons experimentally kept under conditions mimicking the One Loft Race rearing system.

INTRODUCTION: Racing pigeon competitions are a popular sport where success depends on birds' ability to return fast to their loft of origin. However, many additional factors like differences in feeding, training, everyday care and even geographical loft location influence race outcomes, which has led to the development of the One Loft Race (OLR) system. The OLR system aims to eliminate these factors by housing pigeons from various lofts in equal conditions in one facility. This in turn, however, fosters inter-individual transmission of pathogens. MATERIAL AND METHODS: Fifteen young racing pig

DUSP1 mRNA modulation during porcine circovirus type 2 and porcine reproductive and respiratory syndrome virus co-infection regulates viruses replication.

The effects of porcine circovirus type 2b (PCV2b) and porcine reproductive and respiratory syndrome virus (PRRSV) co-infection in epithelial cells of the swine respiratory tract is unknown. In the present study, the newborn pig trachea cell line NPTr-CD163, which is permissive to both viruses, was persistently infected with PCV2b and then with PRRSV. Viral replication, cell viability, cytokines' mRNA expression, and modulation of cellular genes expression were evaluated in infected cells. In NPTr-CD163 co-infection model, PCV2b replication was enhanced while PRRSV replication was suppressed. C

Characterization of a gE/gI/TK gene-deleted pseudorabies virus variant expressing the Cap protein of porcine circovirus type 2d.

Porcine circovirus type 2 (PCV2) plays a key role in the etiology of PCV2-associated disease (PCVAD), and its predominant strain is PCV2d which is not completely controlled by most commercially available vaccines against PCV2a strains. Pseudorabies (PR) caused by pseudorabies virus (PRV) variants re-emerged in Bartha-K61 vaccine-immunized swine herds in late 2011, which brought considerable losses to the global pig husbandry. Therefore, it is significantly important to develop a safe and effective vaccine against both PCV2d and PRV infection. In the present study, the PCV2d ORF2 gene was ampli

The prevalence of porcine circovirus type 2e (PCV2e) in Korean slaughter pig lymph nodes when compared with other PCV2 genotypes.

The objective of this study was to determine the prevalence of porcine circovirus type 2e (PCV2e) over other PCV2 genotypes (a, b, c and d) from the lymph nodes of 1,550 randomly selected slaughter pigs. Samples were obtained at a rate of five samples per farm from 310 farms between January 2018 and May 2020. Of the 1,550 lymph node samples, PCV2 DNA was detected in 762 (49.20%) samples. Among the 762 PCV2 DNA-positive samples, a single PCV2 genotype was detected in 744 samples, while multiple PCV2 genotypes were only detected in 18 samples. Of the 744 single infection cases, PCV2d was the mos

Porcine circovirus type 2 exploits JNK-mediated disruption of tight junctions to facilitate Streptococcus suis translocation across the tracheal epithelium.

Porcine circovirus type 2 (PCV2) is considered as the primary pathogen of porcine circovirus-associated disease (PCVAD), which results in significant economic losses worldwide. Clinically, PCV2 often causes disease through coinfection with other bacterial pathogens, including Streptococcus suis (S. suis), and especially the highly prevalent S. suis serotype 2 (SS2). The present study determined that continuous PCV2 infection in piglets down-regulates tight junction proteins (TJ) ZO-1 and occludin in the lungs. Swine tracheal epithelial cells (STEC) were used to explore the mechanisms and conse

[Generation and characterization of recombinant classical swine fever virus C-strain expressing the cap protein of porcine circovirus type 2].

Classical swine fever (CSF), caused by classical swine fever virus (CSFV), is a devastating viral disease in swine, leading to significant economic losses to the pig husbandry. C-strain is one of the best modified live vaccines against CSF. The vaccine is highly safe and efficacious and can provide rapid and complete protection against essentially all genotypes of CSFV. Co-infections of pigs with CSFV and porcine circovirus type 2 (PCV2) occur frequently in the field, making it difficult to control the associated diseases. Here, a recombinant C-strain rHCLV-Cap expressing the Cap protein of PC

Immunopathological characterization of porcine circovirus type 2 infection-associated follicular changes in inguinal lymph nodes using high-throughput tissue microarray.

The immunopathogenesis of porcine circovirus type 2 (PCV2) infection in conventional pigs is complicated by various environmental factors and individual variation and is difficult to be completely reproduced experimentally. In the present field-based study, a tissue microarray (TMA) consisting of a series of lymphoid follicles having different PCV2-loads was constructed using formalin-fixed and paraffin-embedded superficial inguinal lymph nodes (LNs) from 102 pigs. Dietary aluminosilicate supplement enhances immune activity in mice and reinforces clearance of porcine circovirus type 2 in experimentally infected pigs.

Aluminosilicate is the major component of clay minerals such as zeolite, bentonite and clinoptilolite. The minerals possess a number of beneficial activities, especially in regulating the immune system. The aims of the present study were to evaluate immune enhancing effects of dietary aluminosilicate supplement (DAS) in mice, and to demonstrate clearance effects of DAS against porcine circovirus type 2 (PCV2) in experimentally infected pigs as an initial step towards the development of an antibiotic substitute for use in pigs. Effect of granulocyte-macrophage colony-stimulating factor on post-weaning multisystemic wasting syndrome in porcine circovirus type-2-transfected piglets.

Post-weaning multisystemic wasting syndrome (PMWS) is a complex disease syndrome in swine, affecting nursery and fattening pigs. Although ongoing evidence suggests that porcine circovirus type-2 (PCV2) is the causal agent of PMWS, the host immune system appears to have a crucial role in the PMWS pathogenesis of PCV2-affected pigs. Owing to difficulties in producing a biologically pure form of PCV2 devoid of the other viral agents commonly present in swine tissues, we decided to use a tandem-cloned PCV2 DNA providing highly pure grade reagent in order to monitor the virulence of PCV2 alone or w
