[Properties of phase I purified cellular antigen of Coxiella burnetii].

An attempt was made to purify phase I cell suspension of Coxiella burnetii used as an antigen in diagnostic serological tests. Homogenised suspension of chick embryos infected with phase I Henzerling and "Z" strains, after preliminary purification from host cell contaminants of chick embryos was subjected to consecutive centrifugation in sucrose/uropoline gradient and to continuous 20-45% uropoline gradient. The fractions obtained from uropoline gradient centrifugation were applied as phase I antigen C. burnetii in the following tests: complement fixation and microagglutination. Only fractions

Serum complement-fixing antibody to Pasteurella haemolytica A1 and its effect on experimentally induced pneumonic pasteurellosis in calves.

Ninety-three calves comprising 16 experimental groups were exposed to viral (bovine herpesvirus-1 or parainfluenza-3 virus) and Pasteurella haemolytica aerosols. Serum samples from these calves were tested before and after exposure for antibodies to P haemolytica by a modified direct complement-fixation test. At slaughter of the calves, the extent of pneumonia produced was estimated for each calf and compared with the results of the modified direct complement-fixation tests. The extent of pneumonia was not related (P greater than 0.05) to the amount of anti-P haemolytica antibody produced by e

Serological response of cattle to Brucella allergen after repeated intradermal applications of this allergen.

A study was conducted to determine whether an allergen that has been prepared from a mucoid strain of Brucella abortus triggers a serum antibody response that interferes with the interpretation of serologic tests results. Fifteen cattle seronegative for Brucella antigen were tested with the SDTH test several times. Blood samples were collected weekly and tested with the serum agglutination test and complement fixation test. Results show that some cattle tested seronegative after each of the SDTH tests while other cattle tested weakly positive with the serum agglutination test or the complement

Immunoglobulin responses in acute Q fever.

Knowledge of the development of different classes of antibody during the course of acute Q fever is important to the clinician for interpreting a patient's serological test results. In the present study, the appearance of antibodies to Coxiella burnetii phases I and II was determined for a period of 1 year. A total of 683 sera from 191 patients with symptomatic Q fever were evaluated by the complement fixation and indirect immunofluorescence (immunoglobulins M and G [IgM, IgG]) tests. These patients had contracted acute Q fever in the fall of 1983 during an epidemic that resulted in 415 serolo

[Contribution of immunofluorescent reactions in the cerebrospinal fluid to the study of neurocysticercosis].

The most important immunobiological aspects of neurocysticercosis and the cerebrospinal fluid (CSF) syndrome in this disease are reviewed. A study was made of 1.577 samples of CSF from 1.282 patients who suffered from several diseases of central nervous system. They were divided in 3 groups: 1- 459 patients with normal results of CSF tests; 2- 600 patients with pathological CSF, but with immunological reactions non-reactive; 3- 233 patients divided in 8 sub-groups according the results of the immunological reactions. CSF immunofluorescent reactions for syphilis (FTA-Abs). cysticercosis and tox

Immune response to Mycoplasma pneumoniae P1 and P116 in patients with atypical pneumonia analyzed by ELISA.

BACKGROUND: Serology is often used for the diagnosis of Mycoplasma pneumoniae. It is important to identify specific antigens that can distinguish between the presence or absence of antibodies against M. pneumoniae. The two proteins, P116 and P1, are found to be immunogenic. By using these in ELISA it is possible to identify an immune response against M. pneumoniae in serum samples. RESULTS: A recombinant protein derived from the P116 protein and one from the P1 protein were used in two ELISA tests, rP116-ELISA and rP1-ELISA. Human serum samples from patients with atypical pneumonia were teste

An epizootic of measles in captive silvered leaf-monkeys (Presbytis cristatus) in Malaysia.

An epizootic of measles occurred in a group of 31 silvered leaf-monkeys (Presbytis cristatus) that had been in captivity for 4-12 months. Twenty-four of the monkeys exhibited a maculopapular rash that persisted for 6-9 days. A serous to mucopurulent nasal discharge and conjunctivitis were seen in some animals. Eight monkeys died during the epizootic; however, their deaths could not be directly attributed to measles. Serum samples from the surviving monkeys collected 1-2 months prior to, and 5 weeks after, the epizootic were examined by the complement-fixation and hemagglutination-inhibition te

Leptospira interrogans serovar hardjo infection of cattle.

A survey of normal cattle in the Southern Victorian statistical divisions revealed that microscopic agglutination titres to L. hardjo occur at high frequency and are distributed throughout the cattle population. These titres are difficult to interpret as they may represent recent or old exposure, with or without disease. L. hardjo infection of dairy cattle was studied in 4 herds using the microscopic agglutination and complement-fixation tests. Statistical comparisons of individual titres obtained indicated that the sensitivity of the complement-fixation test was satisfactory for diagnostic pu

[The complement fixation reaction: a traditional, updated method for the detection of antibodies in the diagnosis of infections].

Complement-fixation (CF) is still an important basic serologic test for the diagnosis of infectious diseases. In several areas of microbiology (viral, bacterial, parasitic and fungal) it has served as a reference standard against which other methods have been compared. Its partial displacement by other techniques, as advocated in some recent literature, is often unfounded and uncritical; displacement is mostly due not to problems inherent in the method, but rather to the lack of reagents of satisfactory quality. The CF technique has been greatly improved in recent years. Improvements include t

Serological reactions of fractionated hamster immunoglobulins with California group viruses.

Immunoglobulin M and immunoglobulin G (early and late) were separated by sucrose density gradient centrifugation from single inoculation hamster immune sera prepared against La Crosse, California encephalitis and snowshoe hare viruses. Fractionated antibodies were tested for differences in serological specificity by hemagglutination-inhibition, neutralization, and complement-fixation reactions. None of the immunoglobulins demonstrated a consistent pattern of superior specificity in serological reactions with closely related California group viruses. Results were most uniform in the neutralizat

Specificity dependence between serological tests for diagnosing bovine brucellosis in Brucella-free farms showing false positive serological reactions due to Yersinia enterocolitica O:9.

When brucellosis false positive serological reactions happen in cattle, the serial use of pairs of specificity-correlated serological tests (rose bengal, complement fixation, competitive ELISA) results in specificities lower than expected. In this situation, highly specific tests, such as the indirect ELISA used alone, may be more adequate than serial testing.

IMMUNOFLUORESCENCE AS AN AID IN THE EARLY DIAGNOSIS OF TRICHINOSIS.

The established serological tests for trichinosis are often negative during the period when laboratory investigation is most likely to be us … est was positive two weeks (the earliest blood samples available) after onset, while precipitin and complement fixation tests did not become positive until the end of the fourth week. The immunofluorescence test thus becomes positive at least two weeks earlier than the other two, a factor which undoubtedly increases its value in diagnosis.
