Ventromedial prefrontal cortex activity during extinction recall suggests successful extinction learning via mental imagery.

Mental imagery is an important element of psychological therapies for fear-related disorders. The present study evaluated the efficacy of imagery extinction using a within-subject design allowing for the comparison of a conditioned stimulus extinguished via mental imagery (CS+Ei) to an unextinguished conditioned stimulus (CS+U) during extinction recall. Twenty-eight human participants underwent differential fear conditioning during functional magnetic resonance imagining. Successful acquisition of differential fear conditioning was confirmed by self-reported fear, skin conductance response (SC

The Effects of Early Life Pain and Juvenile Fear Conditioning on CRF-Receptor Expression in the Amygdala and Hypothalamus.

Early life pain and stress have lasting consequences on nervous system development that can interact with later stress or trauma to create a susceptibility to fear, anxiety, depression and chronic pain among other psychological disorders. Recent work has identified changes in corticotropin releasing factor signaling in limbic system structures, such as the amygdala and hypothalamus, as a key mechanism behind these changes - albeit in a sex-dependent manner. CRF has two major receptors, CRHR1 and CRHR2 which have also been shown to play key roles in fear and pain expression. The current work ex

Estradiol is associated with altered cognitive and physiological responses during fear conditioning and extinction in healthy and spider phobic women.

The first-line psychological treatment for anxiety disorders is exposure therapy, which can be modeled in the laboratory using fear extinction. In healthy women, estradiol levels predict return of fear following extinction, whereas low levels are associated with greater return of fear. Investigating whether estradiol is similarly associated with extinction in clinically anxious women may provide insight to mechanisms underlying symptom relapse following exposure therapy. In the present study, women with spider phobia and healthy women participated in a 2-day fear conditioning and extinction pr

Female rats express habitual behavior earlier in operant training than males.

Habitual behavior can be advantageous by increasing the availability of cognitive resources for use in other tasks. However, habitual behaviors are problematic when they are coopted to prolong the maladaptive responding present in several psychopathologies such as substance abuse, dysregulated fear responding in posttraumatic stress disorder, and obsessive-compulsive disorder. Although sex differences exist in the occurrence or progression of these psychopathologies, there are no studies that compare the development of habitual behavior systematically in male and female animals. In the present

Phobias and preparedness: the selective, automatic, and encapsulated nature of fear.

We describe evidence for an evolved module for fear elicitation and fear learning with four primary characteristics. First, it is preferentially activated by stimuli related to survival threats in evolutionary history. Thus, fear-relevant stimuli lead to superior conditioning of aversive associations compared with fear-irrelevant stimuli. Second, the module is automatically activated by fear-relevant stimuli, meaning that fear activation occurs before conscious cognitive analysis of the stimulus can occur. Third, the fear module is relatively impenetrable to conscious cognitive control, and fe

[A psychosomatic theory of allergic sensitization; allergy as a quasi conditioned reaction].

A psychosomatic theory concerning the sensitization process of immediate type allergy is presented. According to this theory the allergy systems from a deficient local defense function of the respiratory tract's mucous membranes. It is assumed that persons, with a specific disposition and learning experiences, in certain conflict situations where emotional impulses cannot be expressed, tend so suffer an obstructive hyperfunction of the nasal and/or bronchial mucosa including hyperemia, swelling, hypersecretion (and spasms of the bronchial muscular system). These modifications of mocous functio

Inhibition of delay a century later: A systematic review and Meta-analysis.

Inhibition of delay has received relatively little attention compared with other Pavlovian phenomena, despite its early description by Pavlov. This review examined the conditions under which inhibition of delay develops and evaluated evidence for it both as a phenomenon, reflected in temporal discrimination of the conditioned stimulus, and as a mechanism involving inhibitory processes acting early in the stimulus. Inhibition of delay has been observed across many species and procedures, typically after extensive training and most often with aversive unconditioned stimuli. Although this diversi

Effect of Tryptophan Depletion on Conditioned Threat Memory Expression: Role of Intolerance of Uncertainty.

BACKGROUND: Responding emotionally to danger is critical for survival. Normal functioning also requires flexible alteration of emotional responses when a threat becomes safe. Aberrant threat and safety learning occur in many psychiatric disorders, including posttraumatic stress disorder, obsessive-compulsive disorder, and schizophrenia, in which emotional responses can persist pathologically. While there is evidence that threat and safety learning can be modulated by the serotonin systems, there have been few studies in humans. We addressed a critical clinically relevant question: How does low

Hippocampus and contextual fear conditioning: recent controversies and advances.

Dorsal hippocampal (DH) lesions produce a severe deficit in recently, but not remotely, acquired contextual fear without impairing memory of discrete training stimuli, i.e., DH lesions produce an anterograde and time-limited retrograde amnesia specific to contextual memory. These data are consistent with the standard model which posits temporary involvement of the hippocampus in recent memory maintenance. However, three recent controversies apparently weaken the case for a selective mnemonic role for the hippocampus in contextual fear. First, although retrograde amnesia (from posttraining lesi

Inactivation of ventral hippocampus projections promotes sensitivity to changes in contingency.

The loss of behavioral flexibility is common across a number of neuropsychiatric illnesses. This may be in part due to the loss of the ability to detect or use changes in action-outcome contingencies to guide behavior. There is growing evidence that the ventral hippocampus plays a critical role in the regulation of flexible behavior and reward-related decision making. Here, we investigated the role of glutamatergic projections from the ventral hippocampus in the expression of contingency-mediated reward seeking. We demonstrate that selectively silencing ventral hippocampus projections can rest

Emotions and skin (II)-the conditioning of scratch responses in cases of lichen simplex.

Lichen simplex is generally regarded as a condition initiated and perpetuated by scratching and emotional tension. It was felt that the scratching might partly be a conditional response to itching and other signals, and that feelings of guilt, anxiety and hostility would be prominent features in these patients. Conditioning experiments designed to establish scratch responses to an itch stimulus (UCS) and a tone (CS) showed that lichen simplex patients conditioned more readily and extinguished more slowly than controls. These finding were more marked when the itch stimulus was applied to affect

[Rehabilitation after total laryngectomy--vision and realization].

Universality, early initiation, complexity and continuity - should be the main attributes of rehabilitation in patients after laryngectomy. The authors discuss the problem of universality and accessibility of rehabilitation in Poland. A great role in realization of this attributes played since 20 years the Polish Society of Laryngectomees. Till now the governmental help and the participation of the National Health Fund for the patients after laryngectomy is very limited. The early rehabilitation should be started before the operation and a particular note must be taken to the patient's changed

Individual threat-relevance accelerates belief-updating in conditioned hallucinations.

Threat-related learning is implicated in psychopathological hallucinations; however, its role in experimentally-induced hallucinations remains unclear. Here, we investigated how threat-relevant stimuli (spiders vs. flowers) influence conditioned hallucinations (i.e., illusory auditory tones triggered by learned associations with images). We found that, for conditioning with spiders (but not flowers), higher spider-phobia was related to time-dependent variations in conditioned hallucinations: increasing in early phases (where spiders reliably predicted tones) and decreasing in later phases (as

Learning Mechanisms in Alcohol Use Disorders.

Pavlovian and operant learning mechanisms promote and maintain alcohol intake despite aversive consequences. Here, we review the evidence for drug-associated modifications of basic learning mechanisms during the initiation and maintenance of alcohol use disorders in humans. We discuss how neuroadaptive effects elicited by chronic alcohol intake in the so-called brain reward system can bias learning and decision making towards drug consumption and contribute to the relapse risk after detoxification. Robustness of results and limitations of the respective neurobiological or imaging techniques ar

Polydipsia induced in the rat by a second-order schedule.

Drinking was studied in rats pressing a bar on a second-order schedule in which every third completion of a 1-min fixed interval was followed by food presentation. A brief flash of light signaled the completion of each fixed-interval component. The rats drank not only after the food presentations but also after presentations of the light flash alone. A high rate of steady drinking followed intervals terminated by a food presentation. Drinking that followed intervals terminated by a light flash alone was of comparable rate, but characteristically interrupted by bar pressing. When 250-mg food pe

Effects of vagus nerve stimulation on extinction of conditioned fear and post-traumatic stress disorder symptoms in rats.

Exposure-based therapies help patients with post-traumatic stress disorder (PTSD) to extinguish conditioned fear of trauma reminders. However, controlled laboratory studies indicate that PTSD patients do not extinguish conditioned fear as well as healthy controls, and exposure therapy has high failure and dropout rates. The present study examined whether vagus nerve stimulation (VNS) augments extinction of conditioned fear and attenuates PTSD-like symptoms in an animal model of PTSD. To model PTSD, rats were subjected to a single prolonged stress (SPS) protocol, which consisted of restraint, f

[Placebo Effects in Acute Pain Therapy].

Placebo and nocebo effects are currently within the focus of clinical and experimental pain research. Neurophysiological and psychophysiological mechanisms might play an important role. Placebo and nocebo effects might be mediated through the descendent inhibitory pain pathway via endogenous opioids. Apart from that three different psychological mechanisms might be relevant for placebo and nocebo effects: classical conditioning, social learning and pain expectation. An open analgesic drug application, detailed patient information about efficacy and possible adverse events (without an extensive

The placebo response in functional bowel disorders: perspectives and putative mechanisms.

The nature and determinants of the placebo response are widely unknown, as are the underlying psychological and biological mechanisms. High placebo response rates in functional bowel disorders (functional dyspepsia, irritable bowel syndrome) are similar to those in non-intestinal diseases (depression, pain, Parkinson's disease) and not too dissimilar to other organic gastrointestinal diseases (duodenal ulcer, inflammatory bowel diseases). Methodological reasons (regression to the mean, shift in signal detection through manipulation of expectations) and psycho-biological mechanisms (Pavlovian c

Hippocampus and retrograde amnesia in the rat model: a modest proposal for the situation of systems consolidation.

The properties of retrograde amnesia after damage to the hippocampus have been explicated with some success using a rat model of human medial temporal lobe amnesia. We review the results of this experimental work with rats focusing on several areas of consensus in this growing literature. We evaluate the theoretically significant hypothesis that hippocampal retrograde amnesia normally exhibits a temporal gradient, affecting recent, but sparing remote memories. Surprisingly, the evidence does not provide much support for the idea that there is a lengthy process of systems consolidation followin
