Delayed loss of corneal epithelial stem cells in a chemical injury model associated with limbal stem cell deficiency in rabbits.

PURPOSE: Ocular injuries following exposure to the chemical agent sulfur mustard (SM) are characterized by acute corneal erosions and inflammation of the anterior segment that may be followed by delayed Partial Limbal Stem Cell Deficiency (LSCD), expressed clinically by corneal neovascularization and epithelial defects. LSCD may derive from direct destruction of limbal stem cells or indirectly from altered limbal stromal niche. The aim of this study was to investigate the mechanism underlying LSCD in SM injuries, focusing on the effects of the chemical on limbal epithelium. METHODS: Rabbit ey

Effects of keratinocyte growth factor-2 on corneal epithelial wound healing in a rabbit model of carbon dioxide laser injury.

Keratinocyte growth factor-2 (KGF-2), also called fibroblast growth factor-10 (FGF-10), is a member of the fibroblast growth factor family. It plays a critical role in epithelial development and exerts its biological activities in a paracrine manner on the receptor FGFR2-IIIb. This study examined the function of topically applied KGF-2 in vivo on wound healing using a CO(2) laser, corneal epithelial wounded, rabbit model. Topically applied 25 microg/ml KGF-2 accelerated corneal epithelial wound healing, in contrast to the control, and reduced inflammation, stromal edema, and fibrosis. In addit

Flt-1 intraceptor induces the unfolded protein response, apoptotic factors, and regression of murine injury-induced corneal neovascularization.

PURPOSE: To determine whether Flt24K, a recombinant construct of domains 2 to 4 of VEGFR-1 (Flt) coupled with an endoplasmic reticulum retention signal (KDEL) can bind VEGFR-2 and induce apoptosis, unfolded protein response (UPR), and regression of injury-induced corneal neovascularization. METHODS: Human microvascular endothelial cells were transfected with pCMV.Flt24K and subjected to hypoxia. Cell lysates underwent Western blot analysis with anti-XBP-1 antibody and RT-PCR for CHOP. Human malignant melanoma cells (which express VEGFR-2 but not Flt), were transfected with pCMV.Flt24K, and ly

Ability of in vitro (corneal injury--eye organ--and chorioallantoic membrane) tests to represent histopathological features of acute eye inflammation.

The spontaneous removal of substances from the eye in vivo and histopathological responses to mild or moderate irritants were examined to define the main features that may be reflected in a valid in vitro test for irritant potential. Solutions or fine particulate suspensions applied to the rabbit eye are quickly removed from the central corneal surface by the blink reflex, gravity and drainage. The histological changes in the first 24 hours of mild-to-moderate irritation show some thinning of, but no severe irreversible damage to, the corneal epithelium. Other changes are oedema, congestion, s

Growth factor receptor-bound protein 2-mediated corneal epithelial injury drives depression-associated dry eye disease.

Depression is increasingly recognized as a comorbidity of dry eye disease (DED), yet its underlying mechanisms remain unclear. This study investigates the therapeutic effects of fluoxetine on depression-associated corneal injury and explores the molecular pathways involved. Chronic unpredictable mild stress (CUMS) mice received fluoxetine treatment. Behavioral tests, corneal wound healing, tear secretion, and 4D-DIA proteomics were performed. GRB2 expression was validated via immunohistochemistry, qRT-PCR, and western blot. Human corneal epithelial cells (hCECs) under hyperosmotic stress (450

Primary cultures of rabbit corneal epithelial cells as an experimental model to evaluate ocular toxicity and explore modes of action of toxic injury.

In vivo and in vitro animal models are used to investigate the toxicological modes of action of a substance as surrogates for humans since it is not ethical to conduct certain experiments in humans. The toxic actions of many compounds are manifested in specific organs, known as target organs of toxicity. Thus, in vitro systems that use cells derived from that target organ are best used to understand toxicological mechanisms. This article reviews the development of primary cultures of rabbit corneal epithelial cells which retain tissue specific and functional properties of in vivo cells as an e

A bioactivity-enhanced thermosensitive amnion-derived hydrogel with sustained IGF-1 release: A multitargeted and efficient strategy for corneal injury repair.

Corneal alkali burn (CAB) remains difficult to treat because rapid tear clearance limits topical drug exposure, while the injured cornea develops a complex microenvironment involving inflammation, neovascularization, fibrosis, epithelial barrier loss, and impaired nerve repair. Here, we developed a human amniotic membrane-derived thermosensitive hydrogel (hAMgel) and loaded it with insulin-like growth factor-1 (IGF-1) to create a bioactive sustained-release system (IGF1@hAMgel) for CAB repair. The hAMgel transformed from a flowable precursor into a semi-transparent film on the ocular surface w

Differential expression of corneal and limbal cytokines and chemokines throughout the clinical course of sulfur mustard induced ocular injury in the rabbit model.

PURPOSE: The sight threatening sulfur mustard (SM) induced ocular injury presents specific symptoms for each clinical stage. The acute injury develops in all of the exposed eyes and is characterized by erosions and severe inflammation. The irreversible late pathology develops only in part of the eyes, and is clinically expressed by chronic inflammation and corneal neovascularization (NV). The mechanisms underlying this injury are still in research and treatment is insufficient. Aiming to shed light on pathological mechanisms and improve the therapeutic measures, we studied the expression patte

Mesenchymal Stem Cell-Derived Exosomes as Drug Carriers for Delivering miRNA-29b to Ameliorate Inflammation in Corneal Injury Via Activating Autophagy.

PURPOSE: Corneal injury (CI) resulting in corneal opacity remains a clinical challenge. Exosomes (Exos) derived from bone marrow mesenchymal stem cells (BMSCs) have been proven effective in repairing various tissue injuries and are also considered excellent drug carriers due to their biological properties. Recently, microRNA-29b (miR-29b) was found to play an important role in the autophagy regulation which correlates with cell inflammation and fibrosis. However, the effects of miR-29b and autophagy on CI remain unclear. To find better treatments for CI, we used Exos to carry miR-29b and inves
