Host Genetic Background Effect on Body Weight Changes Influenced by Heterozygous Smad4 Knockout Using Collaborative Cross Mouse Population.

Obesity and its attendant conditions have become major health problems worldwide, and obesity is currently ranked as the fifth most common cause of death globally. Complex environmental and genetic factors are causes of the current obesity epidemic. Diet, lifestyle, chemical exposure, and other confounding factors are difficult to manage in humans. The mice model is helpful in researching genetic BW gain because genetic and environmental risk factors can be controlled in mice. Studies in mouse strains with various genetic backgrounds and established genetic structures provide unparalleled oppo

Crossed pulmonary arteries: An underestimated cardiovascular variant with a strong association with genetic syndromes-A report of 74 cases with systematic review of the literature.

Crossed pulmonary arteries (CPAs) represent an uncommon anatomic variant, usually associated with some specific syndromes and conotruncal defects. This finding has been described in 22q11.2 Deletion Syndrome (22q11.2DS). We evaluated the correlation between CPAs and genetic diseases, in order to better define the characteristics of this variant, considered a rare anatomic pattern. An in-depth analysis of CPAs genotype-phenotype correlations was performed via a literature review. We detected 74 CPAs patients through echocardiography. Of these 74 patients, 35.1% of patients showed additional int

Genetic and peripheral markers of the oxytocin system and parental care jointly support the cross-generational transmission of bonding across three generations.

BACKGROUND: Human and animal research indicates that oxytocin (OT) plays a key role in the cross-generational transmission of parental bonding, and human studies suggest that allelic variations on the oxytocin receptor gene (OXTR) and circulating OT levels interact with patterns of parental care to shape children's social-affiliative competencies. Yet, no study to date has tested the joint contribution of OT and parental care across three generations. METHODS: The study included 345 participants comprising 115 family lines of grandmothers, mothers, and their infants. Salivary OT and allelic v

Genetic ancestry of patients with porphyria cutanea tarda in a country with mixed races: a cross-sectional study (Rio de Janeiro - Brazil).

Porphyria cutanea tarda has a complex etiology with genetic factors not completely elucidated. The miscegenation of the Brazilian population has important implications in the predisposition to diseases. There are no studies concerning the genetic ancestry of patients with porphyria cutanea tarda from a mixed population. Thirty patients living in Rio de Janeiro with sporadic porphyria cutanea tarda were studied for the genetic ancestry through informative markers - INDELS. There was a significant predominance of European ancestry across the sample of patients with porphyria cutanea tarda (70.2%

Identification of new genetic variants of HLA-DQB1 associated with human longevity and lipid homeostasis-a cross-sectional study in a Chinese population.

Healthy longevity has been an unremitting pursuit of human, but its genetic and the environment causes are still unclear. As longevity population is a good healthy aging model for understanding how the body begin aging and the process of aging, and plasma lipids metabolism and balance is a very important to life maintain and physiologic functional turnover. It is important to explore how the effect of genetic variants associated long-life individuals on lipids metabolism and balance. Therefore, we developed a comparative study based population which contains 2816 longevity and 2819 control. Th

Genetic variants in 8-oxoguanine DNA glycosylase 1 (OGG1) and X-ray repair cross-complementing protein 1 (XRCC1) and non-genetic factors are associated with bladder cancer susceptibility and prognosis in a Brazilian population.

Cellular responses to DNA damage play a crucial role in carcinogenesis, particularly in environmentally related cancers such as bladder cancer (BC). This is the ninth most frequently occurring cancer globally and most frequent in the urinary system. Polymorphic variants in genes encoding DNA repair proteins may interfere with their expression and/or activity. Consequently, modulation in an individual's gene response to exposure might occur resulting in greater susceptibility and lower survival rates in BC. Taking this into account, the study evaluated associations of single-nucleotide polymorp

Shared genetic architecture between constipation and aging-related phenotypes: Insights from large-scale genome-wide cross-trait analysis.

While the frequency of constipation increases sharply with age, the genetic links between constipation and aging-related phenotypes remain largely unexplored. So far, research has typically focused only on individual characteristics while neglecting broader genetic connections. In this study, we evaluated the shared genetic architecture of constipation and aging-related phenotypes in order to better understand the causes of aging-related intestinal dysfunction. Using publicly available genome-wide association study summary statistics, we developed a multi-stage analysis pipeline to identify co

Identification of Key Indicators in the Adverse Outcome Pathway for Genetic Damage Induced by Chromate Exposure: A Machine Learning-Based Cross-Sectional Study.

How chronic inflammation and immune dysregulation drive genetic damage upon environmental exposure is a pivotal question in toxicology. Focusing on hexavalent chromium [Cr-(VI)], a human carcinogen, we dissected this link using a machine-learning approach in an occupational population. We discovered that specific immune checkpoint molecules (PD-L1, PD-1, LAG-3) and regulatory patterns mediated the effect of blood Cr on key immune populations, including PD-1-(+) B cells and CD8-(+) T cells. Furthermore, we defined a core set of four inflammatory-immune indicators (NLRP3, positive/negative immun

A novel genetically-encoded bicyclic peptide inhibitor of human urokinase-type plasminogen activator with better cross-reactivity toward the murine orthologue.

The inhibition of human urokinase-type plasminogen activator (huPA), a serine protease that plays an important role in pericellular proteolysis, is a promising strategy to decrease the invasive and metastatic activity of tumour cells. However, the generation of selective small molecule huPA inhibitors has proven to be challenging due to the high structural similarity of huPA to other paralogue serine proteases. Efforts to generate more specific therapies have led to the development of cyclic peptide-based inhibitors with much higher selectivity against huPA. While this latter property is desir
