Expression and loss of alleles in cultured mouse embryonic fibroblasts and stem cells carrying allelic fluorescent protein genes.

BACKGROUND: Loss of heterozygosity (LOH) contributes to many cancers, but the rate at which these events occur in normal cells of the body is not clear. LOH would be detectable in diverse cell types in the body if this event were to confer an obvious cellular phenotype. Mice that carry two different fluorescent protein genes as alleles of a locus would seem to be a useful tool for addressing this issue because LOH would change a cell's phenotype from dichromatic to monochromatic. In addition, LOH caused by mitotic crossing over might be discernable in tissues because this event produces a pair

Genotoxicity assay of oil dispersants in bacteria (mutation, differential lethality, SOS DNA-repair) and yeast (mitotic crossing-over).

5 oil dispersants and a sample of paraffin were devoid of mutagenic activity in the Ames reversion test, with and without S9 mix, using 7 his- S. typhimurium strains (TA1535, TA1537, TA1538, TA97, TA98, TA100, TA102). However, 3 dispersants produced direct DNA damage in E. coli WP2, which was not repairable in repair-deficient strains (WP2uvrA, CM871, TM1080), as shown by two different DNA-repair test procedures. The uvrA excision-repair system was in all cases the most important mechanism involved in repairing the DNA damage produced by oil dispersants, while the combination of uvrA with othe

The food additive microbial transglutaminase is a potential new environmental inducer of autoimmune diseases.

Microbial transglutaminase (mTG) is a frequently used processed food additive, and the consumption of its cross-linked complexes is rapidly expanding. Despite numerous reports concerning its public safety, it is designated as a processing aid and classified as safe for use. mTG and/or its cross-linked complexes can compromise human health. They represent non-self peptides, resulting in non-immune-tolerable neoepitopes. They are proinflammatory, allergenic, immunogenic, pathogenic, human immune system suppressors, and potentially toxic, hence raising concerns for public health. mTG functionally

Longitudinal dynamics of gene expression and metabolomics in an aging population cohort.

Multiomic profiling provides a comprehensive physiological overview at the molecular level, but understanding of its spatiotemporal dynamics remains limited in human populations. We profiled longitudinal whole-blood gene expression and metabolite levels in 335 females over 8 years. Levels of 5061 genes and 181 metabolites changed over time, with individual trajectories often diverging from population-level trends. Longitudinally variable genes showed cell type specificity and enrichment for aging-relevant pathways, including cardiometabolic and neurodegenerative disorders. Longitudinal traject

Natural history of the severe subtype of MYH9-related disease (Epstein syndrome).

OBJECTIVES: This study aimed to describe the natural history and genotype-phenotype associations concerning nonhematological complications in Epstein syndrome, a high-risk subtype of MYH9-related disease (MYH9-RD). METHODS: We conducted a nationwide cross-sectional study using data from congenital thrombocytopenia registries in Japan. Immunofluorescence analysis and gene testing identified patients with Epstein syndrome carrying high-risk MYH9 variants (R702C/H, S96L). We analyzed the onset and progression of nonhematological complications using Kaplan-Meier survival curves. RESULTS: Among 1

Strain difference in transgene-induced tumorigenesis and suppressive effect of ionizing radiation.

Transgenic expression in medaka of the Xiphophorus oncogene xmrk, under a pigment cell specific mitf promoter, induces hyperpigmentation and pigment cell tumors. In this study, we crossed the Hd-rR and HNI inbred strains because complete genome information is readily available for molecular and genetic analysis. We prepared an Hd-rR (p53+/-, p53-/-) and Hd-rR HNI hybrid (p53+/-) fish-based xmrk model system to study the progression of pigment cells from hyperpigmentation to malignant tumors on different genetic backgrounds. In all strains examined, most of the initial hyperpigmentation occurre

Multiplexed microsatellite loci in American crow (Corvus brachyrhynchos): a severely affected natural host of West Nile virus.

Recent advances in high throughput molecular techniques have allowed the development of cost- and time-effective libraries of molecular markers, such as microsatellites, for population genetic studies in non-model species. The American crow, Corvus brachyrhynchos, is recognized to be one of the species that has been most negatively affected by the emergence of West Nile virus (WNV) in North America in 1999. Genetic monitoring of the process of a declining population after the introduction of an infectious disease can provide insights into the demographic and evolutionary impact of a pathogen i

Screening of copy number variants in the 22q11.2 region of congenital heart disease patients from the São Miguel Island, Azores, revealed the second patient with a triplication.

BACKGROUND: The rearrangements in the 22q11.2 chromosomal region, responsible for the 22q11.2 deletion and microduplication syndromes, are frequently associated with congenital heart disease (CHD). The present work aimed to identify the genetic basis of CHD in 87 patients from the São Miguel Island, Azores, through the detection of copy number variants (CNVs) in the 22q11.2 region. These structural variants were searched using multiplex ligation-dependent probe amplification (MLPA). In patients with CNVs, we additionally performed fluorescent in situ hybridization (FISH) for the assessment of

[RYR1 myopathies in childhood: phenotype-genotype correlation and incidence].

INTRODUCTION: Ryanodine receptor type 1-related myopathies (RYR1-RM) represent the most prevalent category of congenital myopathies. The introduction of genetic techniques has shifted the diagnostic paradigm, suggesting the prioritization of molecular studies over biopsies. This study aims to explore the clinical and epidemiological characteristics of patients with RYR1 gene variants in a tertiary pediatric hospital, intending to enhance the understanding of the genotype-phenotype correlation in RYR1-RM. PATIENTS AND METHODS: An observational, descriptive, and cross-sectional study was conduc

Hypersensitivity to mutation and sister-chromatid-exchange induction in CHO cell mutants defective in incising DNA containing UV lesions.

Five UV-sensitive mutant strains of CHO cells representing different genetic complementation groups were analyzed for their ability to perform the incision step of nucleotide excision repair after UV exposure. The assay utilized inhibitors of DNA synthesis to accumulate the short-lived strand breaks resulting from repair incisions. After 6 J/m2, each of the mutants showed less than 10% of the incision rate of the parental AA8 cells. After 50 J/m2, the rate in AA8 was similar to that at 6 J/m2, but the rates in the mutants were significantly higher (approximately 20% of the rate of AA8). Thus b

UV-light induced sister chromatid exchanges in xeroderma pigmentosum lymphocytes.

Cultured lymphocytes from 9 patients with clinically different types of xeroderma pigmentosum were exposed to ultraviolet light at 24 h. An increased rate of sister chromatid exchanges were observed in 6 patients (128--148% increase in three, 34--51% in three), but not in three patients with deSanctis-Cacchione syndrome (xeroderma pigmentosum with mental defect), compared to simultaneously cultured controls. A positive result could be useful as preliminary cytogenetic diagnostic test. The results are interpreted as an expression of UV-light induced chromosomal instability due to impaired DNA r

Reverse Breeding: A Breakthrough Technique for Genetic Fixation and Hybrid Reconstruction.

Reverse breeding (RB), a novel plant breeding approach, helps to identify the parental homozygous lines for any uncharacterized but useful heterozygous plant that a plant breeder needs utmost. For this, genetic engineering at meiotic cell division, particularly meiosis-I, must achieve the right combination and complement homozygous parental lines for recreating the desired hybrid. In this technology, meiotic cross-over is suppressed to stop the recombination at the chromosome level in the selected heterozygous plants. The success of RB in crops depends on the efficiency and effectiveness of do

Mapping quantitative trait loci underlying body weight changes that act at different times during high-fat diet challenge in collaborative cross mice.

Over one billion people worldwide suffer from obesity, and the number is continually rising. This epidemic is partly caused by the modern lifestyle. Animal models, especially mouse models, are crucial to identifying the genetic components of complex disorders and exploring the potential applications of these genetic findings. The body weight of the animals used in research is often measured regularly to monitor their health. Only endpoint measurements, such as ultimate body weight, are frequently examined in quantitative trait locus (QTL) studies; time series data, including weekly or biweekly

Epistatic Features and Machine Learning Improve Alzheimer's Disease Risk Prediction Over Polygenic Risk Scores.

BACKGROUND: Polygenic risk scores (PRS) are linear combinations of genetic markers weighted by effect size that are commonly used to predict disease risk. For complex heritable diseases such as late-onset Alzheimer's disease (LOAD), PRS models fail to capture much of the heritability. Additionally, PRS models are highly dependent on the population structure of the data on which effect sizes are assessed and have poor generalizability to new data. OBJECTIVE: The goal of this study is to construct a paragenic risk score that, in addition to single genetic marker data used in PRS, incorporates e

Highly diverse dynamics of Pseudomonas aeruginosa colonization from initial detection in cystic fibrosis patients: A 7-year longitudinal genetic diversity study.

In cystic fibrosis (CF), Pseudomonas aeruginosa (Pa) is a major pathogen that can persistently colonize patients. Genetic studies showed a high diversity of Pa, the success of widespread or 'international' clones and described epidemic clones in CF and Epidemic High-Risk (ERH) clones. Here, we characterized Pa genetic diversity over time after first colonization in CF patients, with the aim of accurately describing the dynamics of colonization in a context of scarce longitudinal studies including the first isolated Pa strain. Results represent the first genotyping data available for CF Pa in F

Complex interplay between PRRSV-1 genetic diversity, coinfections and antimicrobial use influences performance parameters in post-weaning pigs.

Porcine reproductive and respiratory syndrome (PRRS) is one of the main diseases of pigs, leading to large economic losses in swine production worldwide. PRRSV high mutation rate and low cross-protection between strains make PRRS control challenging. Through a semi-longitudinal approach, we analysed the relationships among performance parameters, PRRSV-1 genetic diversity, coinfections and antimicrobial use (AMU) in pig nurseries. We collected data over the course of five years in five PRRS-positive nurseries belonging to an Italian multisite operation, for a total of 86 batches and over 200,0
