Mixtures of bacterial nucleic acids can often be detected in synovial joints affected with arthritis. We investigated the potential role of such mixtures of bacterial nucleic acids in the pathogenesis of arthritis in a naturally occurring canine model. Dogs with a common inflammatory knee arthritis in which associated pathological degenerative anterior cruciate ligament (ACL) rupture often develops were studied. Synovial biopsies were obtained from 43 dogs with the naturally occurring ACL rupture arthropathy, 12 dogs with normal knees and intact ACL, and 16 dogs with normal knees and experimen

AIM: To investigate the Coxiella burnetii DNA content in environmental samples that may contribute to the transmission of C. burnetii.
METHODS AND RESULTS: During a large Q fever outbreak in the Netherlands, surface swabs and aerosol samples were collected inside stables and around six Q fever-affected ruminant farms, which are located in municipalities varying in Q fever incidence. After the outbreak in 2010, aerosol samples were collected in the same geographical areas. The use of an optimized multiplex qPCR for the detection of C. burnetii DNA revealed that all samples obtained inside stab

Genetic damage is an early effect of altered cell function and tumorigenesis. Analysis of epigenetic alterations in genetic damage in the context of environmental heavy metal-induced cytotoxicity highlights the importance of environment-epigenetics-genetics interactions. The mechanism through which circular RNA (circRNA), an important molecule in epigenetics, regulates cadmium (Cd)-induced DNA damage is unknown. Herein, we report that circRNA circZNF462 regulates chromatin accessibility and inhibits DNA damage after Cd exposure. Downregulation of circZNF462 was associated with DNA damage in a

BACKGROUND: Non-Hodgkin lymphoma in humans is associated with environmental chemical exposures, and risk is enhanced by genetic variants in glutathione S-transferases (GST) enzymes.
OBJECTIVE: We hypothesized that boxer dogs, a breed at risk for lymphoma, would have a higher prevalence of GST variants with predicted low activity, and greater accumulated DNA damage, compared to other breeds. We also hypothesized that lymphoma in boxers would be associated with specific environmental exposures and a higher prevalence of canine GST variants.
ANIMALS: Fifty-four healthy boxers and 56 age-matched

Urban freshwater ecosystems are increasingly exposed to mixtures of pollutants, including per- and polyfluoroalkyl substances (PFAS). However, their ecological effects remain poorly understood, especially in tropical regions with limited sanitation infrastructure. This study investigated PFAS contamination, water quality, and microbial community structure in Pampulha Lake, a tropical urban reservoir in southeastern Brazil. Spatiotemporal patterns in prokaryotic and eukaryotic diversity were assessed over a year (2022-2023) using environmental DNA metabarcoding of 16S and 18S rRNA genes by inte

The thyroid is exposed to DNA damage induced by normal physiological processes (eg, oxidative stress resulting from thyroid hormone synthesis or mitochondrial respiration) or through environmental insults (eg, environmental pollutants, ionizing radiation exposure). Robust antioxidative stress defense and DNA repair mechanisms protect thyrocyte genome integrity, but defective or dysregulated DNA repair pathways have been implicated in thyroid pathology, including autoimmune thyroid disease and thyroid malignancy. In thyroid cancer, disturbed antioxidative stress defense, Mismatch Repair, Non-Ho

CVDs arise from the interplay of multiple factors, with genetics and environmental exposures representing key drivers. Accumulating evidence over recent decades has unequivocally established that epigenetic modifications, most prominently DNA methylation, play a pivotal, non-redundant role in the initiation, development, and progression of CVDs. As a critical molecular bridge linking genetic predisposition, environmental insults, and the pathogenesis of CVDs, DNA methylation dynamically mediates crosstalk among these three components, thereby emerging as a core focus for dissecting the underly

Adult and young herring gulls (Larus argentatus) in Sweden and Iceland were investigated with respect to DNA adducts, analysed with the nuclease-P1 version of the (32)P-postlabelling method, and micronucleated erythrocytes. Three important aims were: (1) to estimate the degree of exposure to genotoxic environmental pollutants in the Baltic Sea area and Iceland, (2) to evaluate the utility of the investigated biomarkers in birds, and (3) to investigate if there was any relationship between genotoxic effects and thiamine deficiency. The results demonstrate that both Swedish and Icelandic herring

Inner-city, minority populations are high-risk groups for adverse birth outcomes and also are more likely to be exposed to environmental contaminants, including environmental tobacco smoke (ETS), benzo[a]pyrene (BaP), and other polycyclic aromatic hydrocarbons (PAHs) found in urban air. In a sample of nonsmoking African-American and Dominican women, we evaluated the effects on birth outcomes of prenatal exposure to ETS, using questionnaire data and plasma cotinine as a biomarker of exposure, and environmental PAHs using BaP-DNA adducts as a molecular dosimeter. We previously reported that amon

BACKGROUND: Epidemiological investigations of Legionella infections are based, since recently, on molecular techniques that are more sensitive and specific than phenotypic traits. We were interested in these methods for subtyping isolates of L. pneumophila serogroup 1 and confirm the epidemic spread of an outbreak of legionnaires' disease at the Universitary Hospital Germans Trias i Pujol (HUGTiP) in Badalona.
METHODS: Environmental samples taken from domestic water, heating and cooling water systems and oxygen humidifiers were examined. Clinical and environmental isolates of L. pneumophila s

Accuracy, sensitivity, simplicity, reproducibility, and low-cost are desirable requirements for genotoxicity assessment techniques. Here we describe a simple electrophoretic assay for genomic DNA lesions quantification (EAsy-GeL) based on subjecting DNA samples to rapid unwinding/renaturation treatments and neutral agarose gel electrophoresis. The experiments performed in this work involved different biological samples exposed to increasing environmental-simulated doses of ultraviolet-B (UVB) radiation, such as Escherichia coli, human leukocytes, and isolated human genomic DNA. DNA extraction

The objective of this study was to develop polymerase chain reaction (PCR) assays for detection of Baylisascaris procyonis eggs and larvae in fecal, environmental, and tissue samples. We have optimized conventional and real-time PCR assays for B. procyonis using the mitochondrial cytochrome oxidase 2 gene as the target for amplification. The lower limit of detection of the parasite genomic DNA was 10 pg in the conventional PCR and 100 fg in the real-time PCR. In both PCR assays, specific amplification of a 146 bp product was achieved with DNA extracted from a single in vitro hatched B. procyon

Pathogens in the environment may pose a threat to our ecosystem and public health by causing infectious disease outbreaks. Early detection and identification are crucial for effective surveillance, outbreak prevention, and source attribution. However, analyzing environmental samples (e.g., air, soil, water, biowaste) is challenging due to their complex composition. Testing for each pathogen, known and undiscovered ones, is not possible yet. Metagenomic shotgun sequencing offers a promising approach for pathogen-agnostic DNA detection in these matrices. This review provides guidance and recomme

Between July 1994 and March 1995, 64 isolates of Pseudomonas aeruginosa were implicated in bacteraemia in 25 cancer patients in five wards of two hospitals. These, together with 24 environmental isolates and one isolate from a bacteraemia in a non-cancer patient were examined by three PCR-based DNA fingerprinting methods: random amplified polymorphic DNA (RAPD), enterobacterial-repetitive intergenic consensus (ERIC)-PCR, and 16S-23S spacer region-based RAPD. These methods were reproducible, discriminatory and showed close agreement; all indicated that 47 isolates that had caused bacteraemia in

BACKGROUND: Trachoma, a conjunctivitis caused by repeated infections with Chlamydia trachomatis, remains a significant cause of blindness worldwide. While mass treatments with azithromycin decreases disease and infection, re-emergence occurs, indicating that elimination may require other sustainable interventions. Environmental changes largely focus on facial hygiene and latrines, but further work to identify other possible transmission targets are needed. We sought to determine, in a cross-sectional survey of households of children with active trachoma, if we could detect the presence of Chla

Many critical aquatic habitats are in close proximity to human activity (i.e., adjacent to residences, docks, marinas, etc.), and it is vital to monitor biodiversity in these and similar areas that are subject to ongoing urbanization, pollution, and other environmental disruptions. Environmental DNA (eDNA) metabarcoding is an accessible, non-invasive genetic technique used to detect and monitor species diversity and is a particularly useful approach in areas where traditional biodiversity monitoring methods (e.g., visual surveys or video surveillance) are challenging to conduct. In this study,

Anthrax is a zoonotic disease caused by Bacillus anthracis, and well recognized as a potential agent for bioterrorism. B. anthracis can be identified by detecting the virulence factors genes located on two plasmids, pXO1 and pXO2. The aim of the present study was to determine the presence of virulence genes in 27 isolates of B. anthracis isolated from clinical and environmental samples. For this purpose, multiplex PCR and DNA probes were designed to detect protective antigen ( pag), edema factor (cya), lethal factor (lef ), and capsule (cap) genes. Our results indicated that all the isolates c

The genetic relatedness of clinical and environmental Cryptococcus neoformans strains in the Maltese Islands was investigated by randomly amplified polymorphic DNA fingerprinting with four primers. The clinical strains isolate over the course of 1 year from AIDS patients showed identical fingerprints. The electrophoretic patterns of the two clinical strains were also the most common patterns among the environmental strains, but the patterns among the environmental strains showed a wide variability and no correlation with the site of isolation.

Workflow for waterborne parasites DNA detection in surface water. Screening for environmental surveillance in monitoring and resource-limited settings. Zoonotic protozoa and microsporidia are waterborne pathogens with implications for animal and human health. Their resistant stages can persist in freshwater. Standard reference methods for water analysis are often limited to specific parasites, require large water volumes, and depends on expensive equipment. Practical alternatives are needed to facilitate parasite detection in water. This study aimed to develop a pellet-based workflow to allow
