Effects of gene therapy on muscle 18S rRNA expression in mouse model of ALS.

BACKGROUND: The efficiency of gene therapy experiments is frequently evaluated by measuring the impact of the treatment on the expression of genes of interest by quantitative real time PCR (qRT-PCR) and by normalizing these values to those of housekeeping (HK) genes constitutively expressed throughout the experiment. The objective of this work was to study the effects of muscle gene therapy on the expression of 18 S ribosomal RNA (Rn18S), a commonly used HK gene. FINDINGS: Mouse model of motor neuron disease (SOD1-G93A) was injected intramuscularly with Brain-derived neurotrophic factor (BDNF

DESCRIPTION AND PHYLOGENETIC AFFINITIES OF A NEW SPECIES OF NEOPSILOTREMA (DIGENEA: PSILOSTOMIDAE) FROM LESSER SCAUP, AYTHYA AFFINIS (ANSERIFORMES: ANATIDAE).

Neopsilotrema is a small genus of psilostomid digeneans parasitic in the intestine of birds in the Palearctic and Nearctic. At present, the genus includes 4 species: Neopsilotrema lisitsynae from the Palearctic and Neopsilotrema affine, Neopsilotrema lakotae, and Neopsilotrema marilae from the Nearctic. Herein, we describe a new species, Neopsilotrema itascae n. sp., from lesser scaup Aythya affinis collected in Minnesota. The species can be distinguished from congeners on the basis of the ventral sucker:oral sucker width ratio, body width:length ratio, and cirrus sac size, along with other ch

Molecular evidence for a novel encapsulated genotype of Trichinella from Patagonia, Argentina.

At present, Trichinella spiralis is the only species of this genus reported from South America. Herein, we detail a molecular analysis of a new encapsulated isolate of muscle larvae of Trichinella, found in a mountain lion (Puma concolor) coming from the Patagonia, Argentina. We studied three DNA regions previously probed to be useful for the identification of all eleven recognized Trichinella genotypes: expansion segment 5 (ES5), cytochrome c-oxidase subunit I (COI) and 5S ribosomal DNA intergenic spacer region (5S ISR). BLAST searches with these DNA sequences showed that the mitochondrial an

p53 in the Molecular Circuitry of Bone Marrow Failure Syndromes.

Mice with a constitutive increase in p53 activity exhibited features of dyskeratosis congenita (DC), a bone marrow failure syndrome (BMFS) caused by defective telomere maintenance. Further studies confirmed, in humans and mice, that germline mutations affecting TP53 or its regulator MDM4 may cause short telomeres and alter hematopoiesis, but also revealed features of Diamond-Blackfan anemia (DBA) or Fanconi anemia (FA), two BMFSs, respectively, caused by defects in ribosomal function or DNA repair. p53 downregulates several genes mutated in DC, either by binding to promoter sequences (DKC1) or

Diversity profiling of xenic cultures of Dientamoeba fragilis following systematic antibiotic treatment and prospects for genome sequencing.

The presence of bacterial DNA in Dientamoeba fragilis DNA extracts from culture poses a substantial challenge to sequencing the D. fragilis genome. However, elimination of bacteria from D. fragilis cultures has proven difficult in the past, presumably due to its dependence on some unknown prokaryote/s. This study explored options for removal of bacteria from D. fragilis cultures and for the generation of genome sequence data from D. fragilis. DNA was extracted from human faecal samples and xenic D. fragilis cultures. Extracts were subjected to 16S ribosomal DNA bacterial diversity profiling. X

Molecular analysis of sarcoidosis lymph nodes for microorganisms: a case-control study with clinical correlates.

INTRODUCTION: Sarcoidosis is an incurable, chronic granulomatous disease primarily involving the lungs and lymph nodes of unknown aetiology, treated with non-specific anti-inflammatory/immunosuppressive drugs. Persistently symptomatic patients worsen with a disabling, potentially fatal clinical course. To determine a possible infectious cause, we correlated in a case-control study the clinical information with the presence of bacterial DNA in sarcoidosis mediastinal lymph nodes compared with control lymph nodes resected during cancer surgery. METHODS: We retrospectively studied formalin-fixed

Circular RNA circERBB2 promotes gallbladder cancer progression by regulating PA2G4-dependent rDNA transcription.

BACKGROUND: CircRNAs are found to affect initiation and progression of several cancer types. However, whether circRNAs are implicated in gallbladder cancer (GBC) progression remains obscure. METHODS: We perform RNA sequencing in 10 pairs of GBC and para-cancer tissues. CCK8 and clone formation assays are used to evaluate proliferation ability of GBC cells. qPCR and Western blot are used to determine expression of RNAs and proteins, respectively. CircRNA-protein interaction is confirmed by RNA pulldown, RNA immunoprecipitation, and fluorescence in situ hybridization. RESULTS: We find that cir

Identification of two DNA helicases UvrD and DinG as suppressors for lethality caused by mutant cspA mRNAs.

CspA is a major cold shock-inducible protein (70 aa), and its major role in the cold shock response was shown to be as an RNA chaperone destabilizing secondary structure of mRNAs at low temperature. Previously, we showed that the overexpression of mutant cspA containing premature non-sense codons at various positions led to stalled ribosomes on mutant cspA transcripts, ultimately leading to cell death. This lethality is primarily due to the highly translatable cspA 5'-UTR that recruits most of the ribosomes from other mRNAs, which are then stalled at the abnormal stop codon. This was called th

Ribotyping and automated ribotyping of Listeria monocytogenes.

Ribotyping is a molecular technique that allows identification and typing of bacteria to the strain level. It is based on restriction endonuclease cleavage of total genomic DNA followed by electrophoretic separation, Southern blot transfer, and hybridization of transferred DNA fragments with a radiolabeled ribosomal operon probe. Following autoradiography, only those bands containing a portion of the ribosomal operon are visualized. The number of fragments generated by ribotyping is a reflection of the multiplicity of rRNA operons present in a bacterial species.Automated Ribotyping-AR (RiboPri

Nocardia veterana isolated from ascitic fluid of a patient with human immunodeficiency virus infection.

Nocardia veterana is a recently characterized species within the genus Nocardia, and only three human clinical isolates have been reported for this species. We describe a case of ascitic fluid infection in an immunocompromised patient due to N. veterana. To our knowledge, this is the first report of a Nocardia sp. strain from ascitic fluid and the fourth report of N. veterana isolated from human samples. Chemotaxonomic methods showed the strain to belong to the genus Nocardia, and identification to the species level was done by 16S ribosomal DNA gene sequencing. The antibiotic susceptibility p

Identification of the Echinococcus (hydatid disease) organisms using cloned DNA markers.

Cloned DNA fragments of the ribosomal RNA gene of Schistosoma mansoni hybridise strongly to Echinococcus DNA following restriction endonuclease and Southern transfer analysis. Individuals within a strain of E. granulosus exhibit identical patterns of hybridisation. However, the hybridisation patterns show significant differences between E. granulosus and E. multilocularis, and between the horse and sheep strains of E. granulosus. This technique represents a powerful, additional method for the identification and characterisation of new isolates of E. granulosus and E. multilocularis.

Characterization of Metarhizium viride Mycosis in Veiled Chameleons (Chamaeleo calyptratus), Panther Chameleons (Furcifer pardalis), and Inland Bearded Dragons (Pogona vitticeps).

Metarhizium viride has been associated with fatal systemic mycoses in chameleons, but subsequent data on mycoses caused by this fungus in reptiles are lacking. The aim of this investigation was therefore to obtain information on the presence of M. viride in reptiles kept as pets in captivity and its association with clinical signs and pathological findings as well as improvement of diagnostic procedures. Beside 18S ribosomal DNA (rDNA) (small subunit [SSU]) and internal transcribed spacer region 1 (ITS-1), a fragment of the large subunit (LSU) of 28S rDNA, including domain 1 (D1) and D2, was s

Unravelling the consortium of the cercarial dermatitis in lake from a basin of Argentinian Central Andes: histological and phylogenetic insights of Chilina snail and their fluke partner.

In the context of a research program dealing with the symbiotic associations between aquatic snails and trematodes, we studied a host-parasite consortium in Lake El Nihuil (Mendoza, Argentina), a water body from Atuel river basin (Central Andes) in which recent outbreaks of cercarial dermatitis ('swimmer's itch') occurred. According to the morphological traits and molecular phylogenetic analyses (mitochondrial COI and 16S rRNA genes), the gastropod host was assigned to the endemic genus Chilina. The snails hosted and released a schistosomatid brevifurcate apharingeate cercaria, whose phylogene

The functions of DNA methyltransferases during the feeding and development of Haemaphysalis longicornis are potentially associated with lysosome pathways.

BACKGROUND: DNA methylation is an epigenetic modification that plays an important role in animal and plant development. Among the diverse types of DNA methylation modifications, methylation of cytosines catalyzed by DNA cytosine methyltransferases (DNMTs) is the most common. Recently, we characterized DNA methyltransferase genes including HlDnmt1 and HlDnmt from the Asian longhorned tick, Haemaphysalis longicornis. However, the dynamic expression and functions of these DNMTs at different developmental stages and feeding statuses of the important vector tick H. longicornis remain unknown. RESU

Three new species of Prosorhynchoides (Digenea: Bucephalidae) from Tylosurus gavialoides (Belonidae) in Moreton Bay, Queensland, Australia.

We surveyed 30 individuals of Tylosurus gavialoides (Castelnau) (Belonidae) collected from Moreton Bay, Queensland, Australia, and describe three new species of Prosorhynchoides Dollfus, 1929 from them. The new species are morphologically distinct from existing Prosorhynchoides spp. and 28S and ITS-2 ribosomal DNA data further supports our morphological findings. We also conduct the first mitochondrial DNA analysis of species of Prosorhynchoides. The new species from T. gavialoides form a strongly supported clade on the basis of the two ribosomal markers, further supporting the emerging hypoth

The divergence pattern of Trichinella spiralis isolates from China revealed by mitochondrial and ribosomal markers.

Trichinella spiralis is the main etiological agent of human trichinellosis. In China, trichinellosis remains a serious food-borne parasitic zoonosis and poses a serious threat to human health. However, the genetic structure of Chinese T. spiralis population is still little known. In this study, we used three molecular markers to analyze phylogeographic structure of the Chinese T. spiralis population. A total of 11 T. spiralis isolates were collected from 10 geographical locations in mainland China. The cytochrome c-oxidase gene (COI), large subunit ribosomal DNA (mt-lsrDNA) and 5S ribosomal DN

Detection of Babesia bigemina infection in strains of Rhipicephalus (Boophilus) microplus collected from outbreaks in south Texas.

The sudden death of several cattle infested experimentally with Rhipicephalus (Boophilus) microplus led to a clinical investigation into the reasons for the unexpected mortality. Microscopic evidence for Babesia bigemina infection was found in blood smears from the affected animals and a PCR assay was designed to detect the presence of B. bigemina and Babesia bovis in all R. microplus strains received and propagated at the laboratory. The assay utilizes a nested PCR approach with the first PCR amplifying a well-conserved segment from the Babesia 18S ribosomal RNA gene followed by a nested PCR

Taxonomy and molecular phylogeny of nassulid ciliates (Alveolata, Ciliophora) with descriptions of one new genus and three new species.

BACKGROUND: As unicellular eukaryotes, ciliates are indispensable components of ecosystems, serving as an intermediate nutrition link between bacteria or algae and meiofauna. The class Nassophorea Small and Lynn, 1981 is a group of morphologically specialized ciliates characterized by a basket-like cytopharynx supported by thick microtubular rods, which are algivorous and thus act as primary consumers in ecosystems. In the present study, the morphology and molecular phylogeny of three new nassulid ciliates were studied based on morphological and small subunit ribosomal ribonucleic acid gene se
