AquaTox-Predictor: An MMoE-Enhanced Multimodal Deep Learning Framework for Aquatic Ecotoxicity Prediction.

As the concentration of compounds in aquatic environments rises, the degradation of aquatic habitats underscores the increasing importance of studying the impact of chemicals on different aquatic populations. In this study, we developed AquaTox-Predictor, a multitask, multimodal classification model that integrates molecular fingerprints, molecular graphs, and a Multi-gate Mixture-of-Experts (MMoE) framework for prediction of seven aquatic ecotoxicities of chemicals. The results of the validation sets demonstrated that our model outperformed other methods, achieving the highest average AUC wit

A framework for risk assessment of plastic additives - Part 1: fundamental elements.

There is a growing worldwide focus on evaluating the risk potential of substances associated with the manufacture and use of plastic, including the intentionally added constituents (additives) that impart specific technical functions as well as their transformation products. To enhance the consistent and effective evaluation of plastic additive uses, the European Centre for Ecotoxicology and Toxicology of Chemicals (ECETOC) is proposing a framework for risk assessment of plastic additives in complex scenarios. Here, the fundamental elements of the framework are presented, including an overview

Characterization of Springtail (Arrhopalites caecus) for Use in Soil Ecotoxicity Testing.

Springtails (subclass: Collembola) represent one of the most extensively studied invertebrate groups in soil ecotoxicology. This is because of their ease of laboratory culture, significant ecological role, and sensitivity to environmental contaminants. Folsomia candida (family: Isotomidae) is a globally widespread parthenogenetic species that is prevalent in laboratory toxicity testing with springtails. Conversely, Arrhopalites caecus (family: Arrhopalitidae), a parthenogenic globular springtail species, remains untested in soil ecotoxicology. This species is found in diverse habitats, includi

Locomotor activity and physiological responses of parasite-infected Gammarus fossarum exposed to the herbicide metazachlor.

Herbicides are among the most commonly found contaminants in freshwater ecosystems. Standard tests are frequently employed to assess their ecotoxicological impacts, but sublethal endpoints in non-target species are often not considered. In addition, ecotoxicological investigations rarely take into account that many species from field populations are naturally infected with parasites. To overcome these gaps, our study aimed to investigate how environmentally relevant concentrations of the herbicide metazachlor affect the locomotor activity and selected physiological responses of Gammarus fossar

Microplastics and copper impacts on feeding, oxidative stress, antioxidant enzyme activity, and dimethylated sulfur compounds production in Manila clam Ruditapes philippinarum.

Microplastics (MPs) are widespread ocean pollutants and many studies have explored their effects. However, research on MPs combined impact with copper (Cu) on dimethylated sulfur compound production is limited. Dimethyl sulfide (DMS) is an important biogenic sulfur compound related to global temperatures. This study examined the ecotoxicological effects of polyamide 6 MPs and Cu on dimethylsulfoniopropionate (DMSP), DMS, and dimethyl sulfoxide (DMSO) production in Manila clams (Ruditapes philippinarum). Our findings showed that MPs and Cu increased oxidative stress, indicated by higher superox

The toxicity of arsenic(III), chromium(VI) and zinc to groundwater copepods.

Groundwater ecosystems globally are threatened by anthropogenic contamination, yet there are few ecotoxicological data using obligate groundwater biota on which to base risk assessments. Copepods are found inhabiting aquifers of different geologies around the world and so are a useful taxon for use in ecotoxicological studies of groundwater. The aim of this study was to test the sensitivity of obligate groundwater copepods to metal contaminants (arsenic(III), chromium(VI) and zinc) in groundwater in static 96 h, 14 days and 28 days exposure tests. The copepods were variably sensitive to As, Cr

First experimental assessment of copper and cadmium toxicity and sublethal responses in Clibanarius erythropus (Latreille, 1818).

Trace elements are a persistent threat to Mediterranean coastal ecosystems, where anthropogenic pressures continue to increase. Although biomarker-based approaches are widely used in marine biomonitoring, species-specific ecotoxicological data remain limited. The hermit crab Clibanarius erythropus (Latreille, 1818), a widespread Mediterranean detritivore chronically exposed to sediment-associated trace elements, has never been ecotoxicologically investigated. In this study, the first integrated assessment of the sensitivity of C. erythropus to copper (Cu) and cadmium (Cd) was conducted. Acute

Biomarkers at different levels of organisation after atrazine formulation (SIPTRAN 500SC®) exposure in Rhinella schineideri (Anura: Bufonidae) Neotropical tadpoles.

Brazil is an important consumer of herbicides. In sugarcane cultivation-the country's most extensive agricultural crop-atrazine-based formulations are the principal form of weed control. Several studies have investigated adverse effects of atrazine or their formulations on anurans, but not specifically on Brazilian species. Our aim was therefore to investigate the lethal and sublethal effects of an atrazine-based herbicide in Rhinella schneideri tadpoles and, in particular, effects on the pigmentation system as a new endpoint in ecotoxicological studies. Rhinella schneideri tadpoles at the Gos

Persistent, Bioaccumulative, and Toxic Chemicals in Wild Alpine Insects: A Methodological Case Study.

With their high persistence in the environment and their potential for long-range atmospheric transport, persistent, bioaccumulative, and toxic chemicals (PBTs) may be among the numerous anthropogenic threats to insect populations worldwide. The effects of PBTs on insects have been investigated in the laboratory, but topical field studies are scarce. A reason might be the multiple challenges faced by PBT-related field studies on wild insects. We studied two species of bumblebees (Bombus spp.) and of ants (Formica spp.) in two high-elevation locations in the Austrian and German Alps to tackle t

Environmental Risk Assessment of Nanomaterials in the Light of New Obligations Under the REACH Regulation: Which Challenges Remain and How to Approach Them?

Within the European regulation on the Registration, Evaluation, Authorisation and Restriction of Chemicals (REACH, EC No 1907/2006) specific provisions for nanomaterials were included, which have become effective on 1 January 2020. Although knowledge on the peculiarities of testing and assessing fate and effects of nanomaterials in the environment strongly increased in the last years, uncertainties about how to perform a reliable and robust environmental risk assessment for nanomaterials still remain. These uncertainties are of special relevance in a regulatory context, challenging both indust

Assessing the impact of antineoplastic drugs in the aquatic environment: State of the art and future perspective for freshwater organisms.

Since the late 70s, the continuous pharmaceuticals` input into the environment has raised concerns regarding the eventual risk posed by such compounds to human and environmental health. A major group of pharmaceuticals in terms of environmental impact are the antineoplastic agents (AAs). Herein, we followed a systematic review method to retrieve antineoplastic agents (AAs') ecotoxicological information regarding freshwater species. In this analysis, data from diverse taxonomic groups, from microorganisms to vertebrate species, looked at different levels of biological organization, including ce

A review on microplastics and nanoplastics in the environment: Their occurrence, exposure routes, toxic studies, and potential effects on human health.

Microplastics (MPs) and nanoplastics (NPs) are emerging environmental pollutants, having a major ecotoxicological concern to humans and many other biotas, especially aquatic animals. The physical and chemical compositions of MPs majorly determine their ecotoxicological risks. However, comprehensive knowledge about the exposure routes and toxic effects of MPs/NPs on animals and human health is not fully known. Here this review focuses on the potential exposure routes, human health impacts, and toxicity response of MPs/NPs on human health, through reviewing the literature on studies conducted in

Potential adverse effect of tyrosinase inhibitors on teleosts:A review.

Coloration plays a crucial role in the social communication and survival of organisms. Multidisciplinary studies have been conducted to elucidate the correlation between coloration and melanin biosynthesis (referred as melanogenesis). The multi-copper enzyme tyrosinase catalyzes the first two steps of melanogenesis for coloration in teleosts. Due to the increasing demand of tyrosinase inhibitors for the production of skin whitening cosmetics, hypopigmentation pharmaceuticals, and anti-browning agents, a large number of natural and synthetic inhibitors have been developed over the past few deca

Pesticides in the rivers and streams of two river basins in northern Greece.

The pollution caused by pesticides, and their ecotoxicological implications were investigated in water samples from the Strymonas and Nestos river basins (Northern Greece). Chlorpyrifos was the most frequently detected pesticide in both basins (42 and 37% in the Strymonas and Nestos basins, respectively), followed by fluometuron and terbuthylazine (25 and 12%, Strymonas), and bentazone and boscalid (24 and 10%, Nestos). The Annual Average and the Maximum Allowable Concentration of Environmental Quality Standards set in European Union Directives were exceeded in several cases by alphamethrin an

Toxic effects of chlorpromazine on Carassius auratus and its oxidative stress.

Under laboratory conditions, ecotoxicological effects of chlorpromazine (CPZ) on freshwater goldfish (Carassius auratus) were examined using the toxic culture experiment. The results showed that the median lethal concentration (LC(50)) of CPZ toxic to Carassius auratus in 24, 48 and 96 h was 1.11, 0.43 and 0.32 mg/L, respectively. Thus, CPZ is an extreme toxicant to goldfish. Furthermore, there were significantly positive correlations between the ecotoxicological effects of CPZ and its concentrations, and the toxicity became higher as the exposure time increased. The activity of superoxide dis

Ecotoxicity evaluation of thymol in tunneller dung beetles (Coleoptera: Geotrupidae) as a natural alternative to ecotoxic anthelmintics.

Synthetic anthelmintics widely used in livestock production are largely excreted unchanged in faeces, where they can exert sublethal and lethal effects on non-target organisms such as dung beetles. Phytochemical compounds, including thymol (THY), have been proposed as environmentally compatible alternatives; however, information on their ecotoxicological safety across different dung beetle taxa remains limited. We assessed the sublethal effects of dietary thymol on Thorectes lusitanicus (Coleoptera: Geotrupidae), a flightless tunneller dung beetle considered functionally sensitive to veterinar

Toxicology of chemical biocides: Anticoagulant rodenticides - Beyond hemostasis disturbance.

The use of anticoagulant rodenticides (ARs) is one of the most commonly employed management methods for pest rodents. ARs compete with vitamin K (VK) required for the synthesis of blood clotting factors in the liver, resulting in inhibition of blood coagulation and often animal death due to hemorrhage. Besides rodents (target species), ARs may affect non-target animal species and humans. Out of hemostasis disturbance, the effects of ARs may be related to the inhibition of proteins that require VK for their synthesis but are not involved in the coagulation process, to their direct cytotoxicity,

The stereoisomerism of second generation anticoagulant rodenticides: a way to improve this class of molecules to meet the requirements of society?

Second generation anticoagulant rodenticides (SGAR) are generally highly efficient for rodent management even towards warfarin-resistant rodents. Nevertheless, because of their long tissue-persistence, they are very associated with non-target exposure of wildlife and have been identified as 'Candidates for Substitution' by the European Union's competent authority. A promising way to reduce ecotoxicity issues associated to SGAR could be the improvement of SGAR based on their stereoisomery, and due to this improvement, positioning about SGAR might be reconsidered. © 2018 Society of Chemical Indu
