Depolarization-induced suppression of inhibition (DSI) in central neurons is mediated by a transient reduction of [gamma]-aminobutyric acid (GABA) release from interneurons. DSI is induced by a retrograde signal emitted from principal cells. We used electrophysiological recordings from CA1 neurons of the rat hippocampal slice to test the hypothesis that only certain classes of interneurons are susceptible to DSI. DSI of action potential-dependent, spontaneous, inhibitory postsynaptic currents (sIPSCs) in hippocampus is facilitated by carbachol (3 microM), which increases the occurrence of larg

BACKGROUND: Epilepsy is a disease caused by paroxysmal abnormal supersynchronous electrical activity of brain neurons, and it is also one of the most common illnesses in neurology. Among the causes, hippocampal sclerosis may be one of the main causes of temporal lobe epilepsy. However, the pathogenesis of hippocampal sclerosis in epilepsy remains unclear.
METHODS: We established an epilepsy model by intraperitoneal injection of pentetrazol (PTZ) into Sprague-Dawley rats, and applied isobaric tags for relative and absolute quantitation (iTRAQ) technology to identify differentially expressed pr

During normal development of the nervous system, a major reduction occurs in the initially excessive number of neurons and synapses. This "pruning" process is heavily influenced by patterns of electrical activity in the synaptic circuits being pruned. Many of the cell biological and molecular mechanisms involved in this activity-dependent modification of nervous system structure and function have been explicated, and the area is one of intense study. Similarly, an explosive increase has occurred in knowledge about the molecular pathogenesis of Alzheimer disease (AD). There are significant mech

BACKGROUND/AIMS: Enteric neuroimmune communication in gastrointestinal hypersensitivity responses includes antigen detection by mast cells and release of chemical messages to the enteric nervous system. The aim of this study was to analyze the electrical and synaptic behavior of neurons in the colonic submucous plexus during exposure to Trichinella spiralis antigen in animals infected earlier with the parasite.
METHODS: Microelectrodes were used to record in submucous neurons of guinea pig distal colon during application of Trichinella antigen.
RESULTS: Neurons in sensitized animals were mor

Slow synaptic excitation and inhibition were studied with intracellular microelectrodes in submucous ganglion cells of the guinea pig ileum. Elevation of adenosine 3',5'-cyclic monophosphate (cAMP) after application of forskolin or the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX) resulted in slowly activating depolarization of the membrane potential. The depolarization was associated with increased input resistance, enhanced excitability, and suppression of hyperpolarizing afterpotentials. This occurred in AH/type 2 but not S/type 1 neurons. The action of forskolin or IBMX mi

The gamma-aminobutyric acid(A) (GABAA) receptor is a ligand-gated ionophore involved in synaptic inhibition. Biochemical and molecular biological studies indicate that considerable receptor heterogeneity exists, but physiological differences between inhibitory GABAA synaptic responses have not been identified in the brain. The present report describes two anatomically segregated GABAA-mediated synaptic currents in the hippocampal CA1 region that have distinct physiological, pharmacological, and functional properties. GABAA,fast enters at or near the cell body, decays rapidly (3-8 ms), is block

Mechanical ventilation (MV) causes a wide range of cognitive impairments. Unfortunately, to date, we are lacking knowledge about its underlying neural mechanisms and significant treatment options for the condition. Here, we show that MV-induced memory impairment in rats stems from dysfunctions in the olfactory bulb-medial prefrontal cortex-ventral hippocampus network and hippocampal synaptic currents imbalance. More importantly, we introduce a novel treatment approach, namely olfactory epithelium electrical stimulation (OEES) that shows promising preclinical results in mitigating the mentioned

Transverse slices from guinea pig hippocampi were exposed to micromolar concentrations of sodium cyanide while neural and synaptic function were monitored in the CA1 region. Cyanide concentrations between 10 and 200 microM rapidly depressed synaptic transmission between Schaffer collateral-commissural fibers and CA1 pyramidal cells. Analysis of input/output curves revealed that the suppression had two components, a decrease in EPSP generation and an increase in action potential threshold. Direct electrical excitability of axons was not affected. At concentrations to 500 microM, cyanide had no

1. Each abdominal ganglion of the crayfish contains peripheral inhibitors of the fast flexor muscles. These flexor inhibitors (FIs), which can effectively inhibit tension development in the tailflip powerstroke muscles, are excited by a delayed central pathway from the same giant axons which trigger escape (33). The FIs also received sensory input, which increases in efficacy in the more posterior segments (4), but until now neither the origin of the input nor its central pathways had been well described. We have used intracellular recording and staining techniques to investigate the afferent

Putative command neurones of snail's escape behaviour reacted with excitatory postsynaptic potentials and spikes to tactile, vestibular, light-off and thermal stimuli evoking behavioural escape reactions. Presented data confirm the command function of identified neurones. No monosynaptic connections have been revealed between intracellularly recorded primary cells of the statocyst and command neurones, but it has been shown that command neurones reacted to strong stimulation of the acoustic nerve. Presence of second-order sensory neurones in the vestibular pathway was assumed.

Responses of motoneurons supplying muscles of the forelimbs, hindlimbs, and back to stimulation of the interstitial nucleus of Cajal (INC) were recorded intracellularly in cats under chloralose anesthesia. Stimulation of the ipsilateral and contralateral INC evoked predominantly excitatory postsynaptic potentials in these motoneurons. Response latencies and properties of responses to multiple shock stimuli indicated that the responses were evoked by a di- or polysynaptic pathway. Stimulation of the anterior MLF (P2), which should have activated the entire interstitiospinal tract, but few retic

Neuronal gap junctions, or electrical synapses, are extensively expressed in the mammalian forebrain and play a key role in synchronizing network activity. Connexin 36 (Cx36) is the primary gap junction protein in mature GABAergic neurons, but its contribution to thalamocortical oscillations and cognitive processes remains unclear. Here, we examined the effects of Cx36 deletion on sleep/wake regulation, spontaneous and evoked EEG activity, and behavior in mice. While Cx36 knockout (KO) mice displayed largely intact sleep architecture, spectral analysis revealed impaired gamma and beta band act…

Inspired by avian visual systems, wide-spectrum bidirectional optoelectronic synaptic transistors (OSTs) have emerged as a transformative platform for neuromorphic vision processing. However, current artificial synaptic technologies remain constrained by fragmented control paradigms that rely on wavelength-selective or electrical polarity-dependent modulation, restricting their capacity for complex visual information processing. In this study, an organic 2D molecular crystal (2DMC) heterojunction is developed to achieve high-performance full-spectrum bidirectional OSTs. The high-quality 2DMC h

Mammalian cochlear hair cells convert acoustic stimuli into electrical signals, which are relayed to the central auditory pathway via auditory neurons. Progressive loss of hair cells and their synapses is a characteristic of age-related hearing loss (ARHL), but the underlying mechanisms remain poorly understood. In this study, we tested the hypothesis that hair cell physiological properties change with age in CBA/CaJ mice, a strain known for its slow ARHL progression. We found that hair cell size began to decrease from 10 months of age, well before detectable increase in hearing thresholds fro

Epilepsy is an important problem in neurological disorders. Recent studies claimed that gap junctions have a critical role in epileptic neur … nd by attenuating the epileptiform behavior. The results of this study suggest that the blockade of electrical synapses may contribute to the prevention and amelioration of epileptic activity.

1. In albino rats the botulinum poisoned gastrocnemius muscle was supplied with an accessory motor nerve in order to investigate whether muscle fibres with structurally intact but non-transmitting synapses would accept additional innervation. As a control similar operations were made in unpoisoned rats.2. One to three months after nerve implantation the muscles were examined histologically for the presence of a new end-plate zone. In botulinum treated muscles 1662 +/- 165 (mean +/- S.D.) of new end-plates were found. In the control animals only a few (90 +/- 13) were observed in the immediate

Ampullary sense organs are distributed in groups over the head of Hydrolagus colliei with their pores in clusters and innervated by the buccal, hyomandibular and superficial ophthalmic branches of the anterior lateral line nerve. The ampullae contain ciliated sense cells in an alveolate-shaped epithelium, which communicates to the surface through a jelly-filled tube. The sense cells synapse at their bases with the afferent nerve fibers that terminate in the dorsal nucleus of the anterior lateral line lobe of the medulla. The anatomy and ultrastructure support the homology with the ampullae of

A few seconds of tetanic preganglionic stimulation of rat sympathetic ganglia results in potentiation of cholinergic synaptic transmission lasting several hours. However, ganglia from aged (28-32 months) rats did not develop as much potentiation as did ganglia from young (3-6 months) rats. This potentiation appears to decay exponentially but in two phases. The early component decays in 15 minutes, a time course consistent with the phenomenon of post-tetanic potentiation (PTP). The later component decays over several hours and is a form of long-term potentiation (LTP). A double exponential deca

High-frequency vibration or electrical stimulation of cutaneous afferents may produce long-lasting hypaesthesia. Such stimulation alters the excitability of axons in the peripheral nerve but there is evidence that this does not completely explain the hypaesthesia. The present study was undertaken to determine whether a prolonged afferent barrage results in depression of synaptic transmission at a central site. Changes in central excitability to cutaneous inputs were examined in normal subjects by measuring the cerebral evoked potential at different stages after high-frequency conditioning stim

Voltage-gated K+ channels of the Kv2 family coassemble with electrically silent KvS subunits in specific subpopulations of brain neurons, forming heteromeric Kv2/KvS channels with distinct functional properties. Little is known about the composition and function of Kv2 channels in spinal cord neurons, however. Here, we show that while Kv2.1 is broadly expressed in multiple classes of spinal cord neurons, the Kv6.4 "electrically silent" subunit is specifically expressed in motoneurons. In motoneurons, we find that Kv6.4 protein is coclustered with Kv2.1 and Kv2.2 subunits at endoplasmic reticul
