Reproductive physiology and endocrinology responses of cows exposed to environmental heat stress - Experiences from the past and lessons for the present.

The effects of environmental heat-stress on production and reproduction in dairy cows have been intensively studied throughout the past few decades. In light of climate changes and global warming, this issue has gained attention worldwide. So far, most of the documentations are related to warmer-climate regions, however, environmental thermal stress has recently been reported in cooler regions, such as Europe. The review attempts to present the experiences from the past years and lessons for the present. The review highlights some of the environmental characterizations and provides some practi

Behavioral and patho-physiological response as possible signs of pain in dairy cows during Escherichia coli mastitis: A pilot study.

Bovine mastitis is one of the most common diseases in the dairy industry and it is a major welfare problem. Pain during mastitis is generally assessed through behavior but a combination of indicators would increase the chances of detecting pain and assessing its intensity. The aim of this study was to assess behavioral and patho-physiological responses as possible signs of pain experienced by cows after experimental intramammary challenge (mastitis) with Escherichia coli. Six Holstein-Friesian cows received an inoculation of E. coli P4 in one healthy quarter. Evolution of the disease was asses

Effects of intratesticular vs intraepididymal calcium chloride sterilant on testicular morphology and fertility in dogs.

BACKGROUND: Both stray and free-roaming owned dogs contribute to the serious global dog overpopulation problem. Many dog owners are unwilling to have their pet castrated for various reasons, including a reluctance to have their dog's behavior changed. A non-surgical method of sterilizing both stray and owned dogs would help to prevent unwanted litters. Previous studies have shown that intratesticular injection of calcium chloride dihydrate (CaCl2) in alcohol is a promising and cost-effective alternative to surgery for stray dogs, with testosterone significantly decreased and sexual activity el

The Energetic Value of Land-Based Foods in Western Hudson Bay and Their Potential to Alleviate Energy Deficits of Starving Adult Male Polar Bears.

Climate change is predicted to expand the ice-free season in western Hudson Bay and when it grows to 180 days, 28-48% of adult male polar bears are projected to starve unless nutritional deficits can be offset by foods consumed on land. We updated a dynamic energy budget model developed by Molnar et al. to allow influx of additional energy from novel terrestrial foods (lesser snow geese, eggs, caribou) that polar bears currently consume as part of a mixed diet while on land. We calculated the units of each prey, alone and in combination, needed to alleviate these lethal energy deficits under c

THE BEHAVIOR IN VIVO OF CERTAIN RELATIVELY PURE ANTIGENS.

The experiments are of interest in several respects. It is clear that crystallized egg albumen is rapidly eliminated from the circulation and in the experiments cited it could no longer be detected after 18 or 19 hours. A considerable portion of it rapidly passes through the kidney in an apparently unaltered state. Evidently this passage begins almost at once and may continue for a day or two. In an experiment not reported in this paper, egg albumen appeared in naturally voided urine 2 hours following its injection into the peritoneal cavity. In the experiments reported no urine was voided unt

Results of a New Patch to Externally Reinforce Colorectal Anastomosis: An Experimental Study.

INTRODUCTION: Anastomotic leak (AL) is a serious complication in colorectal surgery, associated with increased morbidity, mortality, and healthcare costs. Technological advances have yet to eliminate AL, which occurs from 5% to 14% of cases involving anastomosis. This study introduces and evaluates a new collagen-cyanoacrylate patch designed to externally reinforce colonic anastomoses with the aim to contain anastomotic leaks. The objective of the study is to evaluate the feasibility and safety of a novel collagen-cyanoacrylate patch to reinforce colonic anastomosis in a porcine model. METHOD

The pesticide chlorpyrifos increases the risk of Parkinson's disease.

BACKGROUND: Pesticides as a class have been associated with an increased risk of Parkinson’s disease (PD), but it is unclear which specific pesticides contribute to this association and whether it is causal. Since chlorpyrifos (CPF) exposure has been implicated as a risk factor for PD, we investigated its association to incident PD and if this association is biologically plausible using human, rodent, and zebrafish (ZF) studies. METHODS: The association of CPF with PD was performed using the UCLA PEG cohort (829 PD and 824 control subjects), the pesticide use report and geocoding the residence

Human cancer xenografts in immunocompromised mice provide an advanced genuine tumor model for research and drug development-A revisit of murine models for human cancers.

Molecular and cell biology studies have proven that human cancers are an enormously heterogenous disease, even if they originate from the same organ and tissue with identical morphological characteristics. Cancer cells in tumors from different individuals exhibit somewhat different characteristics on multiple levels, such as with respect to 1) their genetic polymorphism; 2) epigenetic mechanisms; 3) group gene activation/inactivation; 4) cell metabolism behavior; 5) aberrant incomplete terminal differentiation; 6) proliferative potential; and 7) hierarchical structure. These multiple parameter

Measuring discrimination- and reversal learning in mouse models within 4 days and without prior food deprivation.

Many neurological and psychiatric disorders are characterized by deficits in cognitive flexibility. Modeling cognitive flexibility in mice enables the investigation of mechanisms underlying these deficits. The majority of currently available behavioral tests targeting this cognitive domain are reversal learning tasks that require scheduled food restriction, extended training periods and labor-intensive, and stress-inducing animal handling. Here, we describe a novel 4-day (4-d) continuously running task measuring discrimination- and reversal learning in an automated home cage (CognitionWall DL/

Role of adrenomedullin in the cerebrospinal fluid-contacting nucleus in the modulation of immobilization stress.

The contribution of the cerebrospinal fluid-contacting nucleus (CSF-contacting nucleus) and adrenomedullin (ADM) to the developmental modulation of stressful events remains controversial. This study explored the effects of endogenous ADM in the CSF-contacting nucleus on immobilization of stress-induced physiological parameter disorders and glucocorticoid hormone releasing hormone (CRH), rat plasma corticosterone expression, and verification of such effects by artificially lowering ADM expression in the CSF-contacting nucleus by targeted ablation of the nucleus. Immunohistochemical experiments

Mating triggers a physiological switch to reproduction in field crickets to mitigate the flight-oogenesis tradeoff.

Organisms must reallocate limited resources to meet changing physiological demands, yet the cues that trigger these shifts and the physiological programs that mediate them remain poorly understood. For example, in some winged animals, females cannot simultaneously invest in energetically expensive flight musculature and egg production, requiring physiological transitions between dispersal and reproductive phases of their life cycle (the flight-oogenesis trade-off). However, the mechanisms that allow individuals to transition between dispersal and reproductive states remain unclear. Here, we te

Multiplex In Vivo Imaging of Biodistribution and Pharmacokinetics of CoMoCAT Single-Walled Carbon Nanotubes.

Semiconducting single-walled carbon nanotubes (SWCNTs) are emerging as powerful near-infrared probes for deep-tissue imaging. While SWCNT samples involving multiple chiralities can be seen as a collection of spectrally resolved fluorophores, their multiplex imaging capabilities remain underexplored. This work presents a filter-resolved evaluation of DSPE-mPEG-coated CoMoCAT SWCNTs integrating optical characterization, biodistribution, pharmacokinetics, and toxicity studies in BALB/c mice. Band-pass filters centered at 1000, 1050, and 1100 nm are used to isolate emission from the dominant (6,5)

Pharmacokinetics of letrozole and effects of its increasing doses on gonadotropins in ewes during the breeding season.

Letrozole is a non-steroidal, third-generation aromatase inhibitor used in humans. Although letrozole is not approved for use in animals, it is used off-label in cases of synchronization and infertility. The aim of this study was to determine the pharmacokinetics of letrozole after a single intravenous administration at three different doses in ewes during the breeding season and its effect on gonadotropins (luteinizing hormone (LH) and follicle-stimulating hormone (FSH)) at the beginning of proestrus. The study was carried out on 24 healthy Merino ewes. Ewes were randomly divided into four gr

Effects of bumetanide on neurobehavioral function in children and adolescents with autism spectrum disorders.

In animal models of autism spectrum disorder (ASD), the NKCC1 chloride-importer inhibitor bumetanide restores physiological (Cl-)i levels, enhances GABAergic inhibition and attenuates electrical and behavioral symptoms of ASD. In an earlier phase 2 trial; bumetanide reduced the severity of ASD in children and adolescents (3-11 years old). Here we report the results of a multicenter phase 2B study primarily to assess dose/response and safety effects of bumetanide. Efficacy outcome measures included the Childhood Autism Rating Scale (CARS), the Social Responsive Scale (SRS) and the Clinical Glob
