Emodin alleviates intestinal ischemia-reperfusion injury through antioxidant stress, anti-inflammatory responses and anti-apoptosis effects via Akt-mediated HO-1 upregulation.

BACKGROUND: Intestinal ischemia-reperfusion (I/R) injury is a severe vascular emergency. Previous research indicated the protective effects of Emodin on I/R injury. Our study aims to explore the effect of Emodin on intestinal I/R (II/R) injury and elucidate the underlying mechanisms. METHODS: C57BL/6 mice and Caco-2 cells were used for in vivo and in vitro studies. We established an animal model of II/R injury by temporarily occluding superior mesenteric artery. We constructed an oxygen-glucose deprivation/reoxygenation (OGD/R) cell model using a hypoxia-reoxygenation incubator. Different dos…

Emodin Inhibitsg Hepatocellular Carcinoma Invasion and Migration by Regulating the HDAC4/NF-κB (p65)/CXCL12 Signaling Axis and Suppressing M2 Macrophage Polarization.

This study aims to investigate how emodin, an active component of Polygonum multiflorum, regulates macrophage polarization through the HDAC4/NF-κB/CXCL12 pathway and impacts the progression of hepatocellular carcinoma (HCC). The inhibitory effect of emodin on M2 polarization was assessed by collecting conditioned cell culture medium from HepG2 cells and treating macrophages with emodin. Network pharmacology and other experiments were used to identify HDAC4 as a key target, and this was further validated through molecular docking, surface plasmon resonance (SPR), and Western blot analysis. Addi…

Emodin, a Potent Anthraquinone Mitigates MPTP-Induced Parkinsons' Disease Pathology by Regulating Nrf2 and Its Downstream Targets: In Silico and In Vivo Approach.

Parkinson's disease (PD) is marked by neurodegeneration that follows the destruction of dopaminergic neurons, mainly localized to the substantia nigra. It results in debilitating motor as well as non-motor symptoms. The current study investigated the neuroprotective potential of emodin, a naturally occurring anthraquinone derivative, in a well-established model of PD in mice induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). The key focus is the Nrf2 signaling pathway, the major defense mechanism of the cells against oxidative damage and neuroinflammation, both exacerbated in the …

Molecular spectroscopic studies on the interactions of rhein and emodin with thioglycolic acid-capped core/shell CdTe/CdS quantum dots and their analytical applications.

Water-soluble thioglycolic acid (TGA)-capped core/shell CdTe/CdS quantum dots (QDs) were synthesized. The interactions of rhein and emodin with TGA-CdTe/CdS QDs were evaluated by fluorescence and ultraviolet-visible absorption spectroscopy. Experimental results showed that the high fluorescence intensity of TGA-CdTe/CdS QDs could be effectively quenched in the presence of rhein (or emodin) at 570 nm, which may have resulted from an electron transfer process from excited TGA-CdTe/CdS QDs to rhein (or emodin). The quenching intensity was in proportion to the concentration of both rhein and emodi…

Discovery of Emodin as a novel competitive KPC-2 inhibitor through virtual screening: Synergistic efficacy with Meropenem against Carbapenem-resistant Klebsiella pneumoniae infection.

Carbapenem-resistant Klebsiella pneumoniae produces the carbapenemase KPC-2, which rapidly confers resistance and poses a significant threat to public health worldwide. Natural compounds have broad application potential, novel structures, and are not easily recognized by existing bacterial resistance mechanisms. Therefore, through virtual screening based on the KPC-2 protein structure and in vitro antimicrobial assessment, the natural anthraquinone compound emodin was identified as a KPC-2 inhibitor (IC50 = 24.4 μM) and a synergist of meropenem. Molecular dynamics simulation and interaction an…

Emodin inhibits lipid accumulation and inflammation in adipose tissue of high-fat diet-fed mice by inducing M2 polarization of adipose tissue macrophages.

Adipose tissue macrophages (ATMs) represent the most abundant leukocytes in adipose tissue (AT). An increase in number and a phenotypical switch of ATMs during the development of obesity contribute to chronic inflammation and metabolic disorders, which have been regarded as potential therapeutic targets to restore AT homeostasis. Emodin has been shown to exert strong anti-inflammatory property via acting on macrophages in a range of disease models. However, whether emodin exerts a beneficial effect on obesity via modulating ATMs has not been reported. In high-fat diet (HFD)-induced obese mice,…

Emodin inhibits Newcastle disease virus replication via regulating the phosphorylation levels of eEF2K/eEF2 and targeting viral N protein to reduce viral protein production.

Newcastle disease (ND) caused by Newcastle disease virus (NDV) infection is a highly contagious avian disease that causes substantial threat to the global poultry industry. Emodin (EMO), a member of the free anthraquinone compounds derived from traditional Chinese medicines, is known for its antibacterial, antiviral, antitumor, and antioxidant activity. However, the anti-NDV activity and potential mechanism of action of EMO remains unknown. In this study, EMO was found to exert a remarkable anti-NDV activity in a dose- and phase-dependent manner. In addition, EMO restricted NDV replication mai…

Mechanistic study of the role of emodin in targeting and inhibiting the Rap1 signaling pathway to regulate epithelial-mesenchymal transition and reverse cisplatin resistance in hepatocellular carcinoma.

BACKGROUND: Chemotherapy plays a crucial role in the treatment of advanced hepatocellular carcinoma (HCC), but the therapeutic efficacy of cisplatin, a commonly used chemotherapeutic agent, is frequently compromised by drug resistance in advanced stages of the disease. A previous study demonstrated that the combination of emodin and cisplatin alleviates cisplatin resistance by inhibiting epithelial-mesenchymal transition (EMT), but the underlying molecular mechanism remains unclear. This study aimed to systematically elucidate the mechanism of action of emodin in enhancing the antitumor activi…
