Enkephalin immunoreactivity in iris nerves: distribution in normal and grafted irides, persistence and enhanced fluorescence after denervations.

The morphology and distribution of nerve fibers showing enkephalin-like immunoreactivity was studied in rat and mouse iris whole mounts. In adult rat, a relatively dense network of varicose fibers was seen throughout the iris. Individual, long, usually smooth fibers were observed running together with non-fluorescent fibers in bundles. Positive nerve fibers were also seen in the ciliary body and the choroid membrane. The fluorescence intensity was normally low. No enkephalin-positive fibers were detected in adult mouse iris. Extirpation or lesioning either one or all the three ganglia known to

Quantitation and localization of Met-enkephalin-Arg-Gly-Leu in rat brain using highly sensitive antibodies.

Met-enkephalin-Arg-Gly-Leu is an endogenous opioid peptide recently identified in bovine adrenal medulla. In the present study, we describe the production of highly sensitive and specific antibodies against this octapeptide. The sensibility of the radioimmunoassay procedure allows us to quantify at the femtomole level, the Met-enkephalin-Arg-Gly-Leu in individual parts of the brain without prior purification or concentration. The antibodies are highly specific for the C terminal part of the molecule, and did not cross-react with the other opioid peptides. Immunochemical techniques were used al

MIF-1 does not act like naloxone in antagonizing the cardiovascular activity of leucine-enkephalin in the conscious dog.

MIF-1 (Pro-Leu-Gly-NH2), a hypothalamic tripeptide, has been demonstrated to stimulate naloxone in antagonizing the effects of opioid peptides in a number of experimental systems including enkephalin-induced analgesia in the tail-flick assay, beta-endorphin induced hypothermia and hypomotility, deprivation-induced drinking, and analgesia in goldfish. MIF-1, however, has no effect upon the activity of enkephalins in the mouse vas deferens or enkephalin binding in the rat striatum. We have studied the interactions of MIF-1 with Leu5-enkephalin (Leu5-ENK) in the conscious, chronically instrumente…

A met-enkephalin analog inhibits adrenocorticotropin secretion by cultured pituitary cells from a patient with Nelson's syndrome.

ACTH excretion by cultured nonenzymatically dispersed pituitary tumor cells from a patient with Nelson's syndrome was studied. Hormone release was suppressed by 74 +/- 6% by the addition of 1 microM of the met-enkephalin analog FK 33824, while naloxone (1 microM) stimulated ACTH release by 70 +/- 5%. Somatostatin, dexamethasone, bromocriptine, and cyproheptadine in a concentration of 1 microM each inhibited ACTH release by 25 +/- 2%, 35 +/- 2%, 52 +/- 2%, and 61 +/- 4%, respectively, while lysine vasopressin (0.1 microM) and dibutyryl cAMP (5 mM) stimulated ACTH release by 112 +/- 8% and 220 +

Effects of Restraint Stress on Circulating Corticosterone and Met Enkephalin in Chickens: Induction of Shifts in Insulin Secretion and Carbohydrate Metabolism.

This study examined the effects of acute restraint stress in the presence or absence of naltrexone on the circulating concentrations of insulin, glucose, Met-enkephalin and corticosterone in 14-week-old chickens [design: 2 sex × 2 stress/non-stress × 2 +/- naltrexone]. In chickens (five male and five females per treatment) subjected to restraint for 30 min, there were increases in the plasma concentrations of corticosterone and Met-enkephalin. The plasma concentrations of insulin and glucose were also increased in the chickens during restraint. Moreover, there were increases in the plasma conc

Striatal pre-enkephalin overexpression improves Huntington's disease symptoms in the R6/2 mouse model of Huntington's disease.

The reduction of pre-enkephalin (pENK) mRNA expression might be an early sign of striatal neuronal dysfunction in Huntington's disease (HD), due to mutated huntingtin protein. Indeed, striatopallidal (pENK-containing) neurodegeneration occurs at earlier stage of the disease, compare to the loss of striatonigral neurons. However, no data are available about the functional role of striatal pENK in HD. According to the neuroprotective properties of opioids that have been recognized recently, the objective of this study was to investigate whether striatal overexpression of pENK at early stage of H

In vivo effects of chronic treatment with [MET5]-enkephalin on hematological values and natural killer cell activity in athymic mice.

The role of endogenous opioids in immunological mechanisms was examined by subjecting athymic (nu/nu) mice to chronic injections of the opioid agonist [Met5]-enkephalin (MET) or continuous opioid receptor blockade with naltrexone (NTX). After 8 days of treatment, neither excess peptide nor deprivation of opioids from receptors had any effect on body weight, spleen index (spleen to body weight ratio), total and differential white blood cell counts, and natural killer (NK) cell activity in peripheral blood or splenic lymphocytes. At 28 days, chronic treatment with MET or NTX had no effect on any

Expression of enkephalin and dynorphin precursor mRNAs in brain areas of hypo-and hyperthyroid rat: effect of kainic acid injection.

An abnormal thyroid status induces morphological and neurochemical modifications in the adult brain. In this study we have analyzed the expression of enkephalin (ENK) and dynorphin (DYN) precursor mRNAs by means of in situ hybridization in the brain of hypothyroid and hyperthyroid rats. The influence of thyroid hormones on kainic acid (KA)-induced expression of ENK and DYN mRNAs in the granule cells of the dentate gyrus was also studied. Our results can be summarized as follows: (1) hypothyroidism induces an up-regulation of ENK mRNA in the granule cells of the dentate gyrus and layers V/VI of

Ultrastructure of Met5-enkephalin terminals in the caudate-putamen nuclei of adult rats receiving neonatal intranigral 6-hydroxydopamine.

Destruction of dopamine neurons of the nigrostriatal pathway in the early postnatal rat enhances the levels of Met5-enkephalin in the adult dorsal striatum (caudate-putamen nuclei) and may contribute to the abnormal self-injurious behavior seen in humans with Lesch-Nyhan disease. We examined whether there were ultrastructural changes in Met5-enkephalin immunoreactive terminals in the rat model that might reflect cellular sites for enhanced activity of these opioid neurons. At 3 days postnatal, 10-20 nl injections of a 1% solution of the dopamine neurotoxin, 6-hydroxydopamine (6-OHDA), or vehic

Galanin-, neuropeptide Y- and enkephalin-like immunoreactivities in catecholamine-storing paraganglia of the fetal guinea pig and newborn pig.

The occurrence and distribution of several neuropeptides and transmitter enzymes have been investigated by means of indirect immunofluorescence histochemistry in preaortal and carotid body-like paraganglia of the fetal guinea pig and the newborn pig. Preaortal paraganglia from the celiac and inferior mesenteric ganglion regions in fetal guinea pigs showed cell bodies immunoreactive (IR) for tyrosine hydroxylase (TH), dopamine beta-hydroxylase (DBH), neuropeptide Y (NPY), galanin (GAL) and metenkephalin (ENK). Almost all cells were IR for TH and DBH, whereas NPY-like immunoreactivity (-LI), GAL

Distribution of substance P-, methionine-enkephalin-, somatostatin- and serotonin-immunoreactive nerve elements of the large bowel in Hirschsprung's disease.

The distribution of substance P-, methionine-enkephalin-, somatostatin- and 5-hydroxytryptamine-immunoreactive nerve elements in Hirschsprung's disease was studied. It was compared to the distribution of the comparable nerve elements in the sigmoid colon and rectum of control children. A reduction of substance P-, methionine-enkephalin- and 5-hydroxytryptamine-immunoreactive fibers as well as a higher density of somatostatin-immunoreactive fibers were found in the aganglionic segment in Hirschsprung gut as compared to those in the ganglionic segment in Hirschsprung children and the large bowel

Nerve elements containing Met-enkephalin-Arg6-Gly7-Leu8-like immunoreactivity in canine pancreas - a histochemical study.

Canine pancreas was examined by Sternberger's peroxidase-antiperoxidase method using an antiserum against Met-enkephalin-Arg6-Gly7-Leu8 (Met-Enk-Arg-Gly-Leu), an opioid octapeptide included in preproenkephalin A. Immunoreactive Met-Enk-Arg-Gly-Leu-containing nerve terminals with beaded appearance were demonstrated to be juxtaposed to small blood vessels of the exocrine and endocrine pancreas. Eighty-five out of 174 ganglion cell somas in the pancreas were immunopositive. The specific immunostaining was absorbed by preincubation of the antiserum (dilution, 1:2000) with Met-Enk-Arg-Gly-Leu (50 m

Lack of effect by substance P at sites in the substantia gelatinosa where Met-enkephalin reduces the transmission of nociceptive impulses.

In alpha-chloralose anaesthetized spinal cats, Met-enkephalin amide (M-ENKA) and substance P were administered electrophoretically in the substantia gelatinosa while studying the excitation of deeper spinal neurones by noxious and non-noxious cutaneous stimuli. At sites where Met-enkephalin selectively reduced excitation by noxious skin stimuli, substance P was without effect. This result does not support the hypothesis that enkephalins are released at axo-axonic synapses on the terminals of substance P releasing primary afferent fibres.

Peptide chirality and opioid receptor modulation: Hepatoprotective effect of d-Met-enkephalin in acetaminophen-induced liver injury.

l-Met-enkephalin is a neuropeptide known to exert protective effects in various experimental models of autoimmune and inflammatory diseases. These effects are mediated through opioid receptors and can be abolished by the opioid receptor antagonist naltrexone. Investigation of peptide enantiomerism and the incorporation of d-amino acids are crucial for designing novel peptides with altered structural and biological properties compared with their native l-forms. Since no data are currently available on the properties or biological activity of the d-Met-enkephalin enantiomer, we evaluated its hep

Methionine enkephalin upregulates toll-like receptors in macrophages to suppress severe fever with thrombocytopenia syndrome virus infection.

BACKGROUND: Severe fever with thrombocytopenia syndrome virus (SFTSV) is a newly identified tick-borne virus with a high case fatality rate. Currently, no specific antiviral drugs are available for its treatment. Methionine enkephalin (MENK) has the ability to enhance immune cell function nonspecifically. This study investigated the mechanism by which MENK regulates macrophages to exert antiviral effects against SFTSV infection. METHODS: An SFTSV-infected RAW264.7 macrophage model pretreated with MENK was developed. RNA sequencing was employed to analyze transcriptomic alterations and identif

The influence of experimental inflammation and axotomy on leucine enkephalin (leuENK) distribution in intramural nervous structures of the porcine descending colon.

BACKGROUND: The enteric nervous system (ENS), located in the intestinal wall and characterized by considerable independence from the central nervous system, consists of millions of cells. Enteric neurons control the majority of functions of the gastrointestinal tract using a wide range of substances, which are neuromediators and/or neuromodulators. One of them is leucine-enkephalin (leuENK), which belongs to the endogenous opioid family. It is known that opioids in the gastrointestinal tract have various functions, including visceral pain conduction, intestinal motility and secretion and immun

Release of [Met5]enkephalin from the spinal cord by intraventricularly administered endomorphin-2, but not endomorphin-1 in the anesthetized rat.

Effects of intraventricular injection of endomorphin-1, endomorphin-2 and beta-endorphin on the release of immunoreactive [Met(5)]enkephalin from the spinal cord were studied in pentobarbital anesthetized rats. Intraventricular injection of endomorphin-2, but not endomorphin-1, caused an increased release of immunoreactive [Met(5)]enkephalin in the spinal perfusates. Beta-endorphin given intraventricularly also increased the release of immunoreactive [Met(5)]enkephalin in an amount 15-fold higher than that produced by endomorphin-2. The increase of the release of immunoreactive [Met(5)]enkepha
