Laminin N-terminus α31 regulates corneal epithelial cell adhesion and migration through modifying the organization and proteolytic processing of laminin 332.

Laminin N-terminus α31 (LaNt α31) is a netrin-like protein generated by alternative splicing of the laminin α3 gene. While previously shown to regulate vascular permeability in vivo, its role in epithelial tissues remains less defined. Here, we demonstrate that LaNt α31 modulates epithelial cell behavior by altering laminin 332 (LM332) organization and hemidesmosome (HD) maturation. Adenoviral driven overexpression of LaNt α31 in corneal epithelial cells led to premature HD assembly, marked by enhanced recruitment of collagen XVII and BPAG1e to β4 integrin, and reduced cell migration. LaNt α31

Collagen XVII is destabilized by a glycine substitution mutation in the cell adhesion domain Col15.

Collagen XVII is a hemidesmosomal transmembrane molecule important for epithelial adhesion in the skin. It exists in two forms, as a full-length protein and as a soluble ectodomain that is shed from the keratinocyte surface by furin-mediated proteolysis. To obtain information on the conformation and the functions of this unusual collagen, its largest collagenous domain, Col15, was expressed in a eukaryotic episomal expression system and purified by DEAE and fast protein liquid- Mono S chromatography. The protein was triple-helical (T(m) of 26.5 degrees C) when produced in cultures containing a

MCAM is a novel metastasis marker and regulates spreading, apoptosis and invasion of ovarian cancer cells.

Melanoma cell adhesion molecule (MCAM) is a cell adhesion molecule that is abnormally expressed in a variety of tumours and is closely associated with tumour metastasis. The role of MCAM in ovarian cancer development has not been fully studied. In this study, through immunohistochemical staining of ovarian cancer tissue samples and RNA interference to silence MCAM in ovarian cancer cells, we examined the impact of MCAM on the biological functions of ovarian cancer cells and attempted to reveal the role of MCAM in ovarian cancer development. Our results showed that MCAM expression was particula

Checkerboard cellular pattern in auditory epithelia: Implications for auditory function and sensory pathology.

Sensory epithelia are composed of specialized cells arranged in specific patterns essential for function. The auditory epithelium of the organ of Corti features a highly conserved checkerboard pattern of mechanosensory hair and supporting cells, preventing direct hair cell contact and preserving epithelial integrity. In mice, disruption of this pattern results in deafness due to hair cell apoptosis caused by abnormal adherens and tight junction formation and structural fragility. This pattern is evolutionarily conserved across species, highlighting its functional significance. Direct hair cell

Immortalized intrahepatic mouse biliary epithelial cells: immunologic characterization and immunogenicity.

Nonsuppurative destructive cholangitis (NSDC), a process of T-cell-mediated destruction of biliary epithelia observed in primary biliary cirrhosis (PBC), graft-versus-host disease (GVHD), and hepatic allograft rejection (HAR), also occurs in the B10. D2-->BALB/c model of GVHD. To advance studies of immunopathogenesis in this murine model, we immortalized 4 BALB/c intrahepatic biliary epithelial cell (BEC) lines as a reliable source of target cells. Freshly isolated BEC, as well as each cell line, expressed cytokeratin-19 (CK-19), epithelial cell adhesion molecule (EPCAM) and cystic fibrosis tr

T and IgA B lymphocytes of the pharyngeal and palatine tonsils: differential expression of adhesion molecules and chemokines.

The pharyngeal (Ph) and palatine (Pa) tonsils, although located in different regions of the upper aero-digestive tract (UADT), are thought to protect the respiratory tract similarly against infections by inducing and disseminating T and surface IgA(+) (sIgA(+)) B cells. We investigated the factors controlling the migratory properties of T and sIgA(+) B lymphocytes in the UADT of pigs by comparing the expression of vascular addressins, homing receptors and chemokine transcripts in Ph/Pa tonsils, Peyer's patches (PP) and their draining lymph nodes (LN). The vascular addressin PNAd was detected o

Enteroids expressing a disease-associated mutant of EpCAM are a model for congenital tufting enteropathy.

Congenital tufting enteropathy (CTE) is an autosomal recessive disease characterized by severe intestinal failure in infancy and mutations in the epithelial cell adhesion molecule (EPCAM) gene. Previous studies of CTE in mice expressing mutant EpCAM show neonatal lethality. Hence, to study the cellular, molecular, and physiological alterations that result from EpCAM mutation, a tamoxifen-inducible mutant EpCAM enteroid model has been generated. The presence of mutant EpCAM in the model was confirmed at both mRNA and protein levels. Immunofluorescence microscopy demonstrated the reduced express

Controlling collagen I orientation on polyetheretherketone implants to improve epithelial sealing.

Transcutaneous devices such as dental implants frequently fail due to infections at their interfaces with epithelial tissues. These infections are facilitated by the lack of integration between the devices and the surrounding soft tissues. This study aims to improve epithelial integration through surface modification of a transcutaneous implant material (polyetheretherketone (PEEK)). The modification involved covalent bonding of collagen via two distinct methods: (1) nonselective binding through any primary amines present on collagen using carbodiimide-based coupling and (2) site-specific bind

Mouse models in palate development and orofacial cleft research: Understanding the crucial role and regulation of epithelial integrity in facial and palate morphogenesis.

Cleft lip and cleft palate are common birth defects resulting from genetic and/or environmental perturbations of facial development in utero. Facial morphogenesis commences during early embryogenesis, with cranial neural crest cells interacting with the surface ectoderm to form initially partly separate facial primordia consisting of the medial and lateral nasal prominences, and paired maxillary and mandibular processes. As these facial primordia grow around the primitive oral cavity and merge toward the ventral midline, the surface ectoderm undergoes a critical differentiation step to form an

Lacrimal Gland Inflammation Deregulates Extracellular Matrix Remodeling and Alters Molecular Signature of Epithelial Stem/Progenitor Cells.

PURPOSE: The adult lacrimal gland (LG) is highly regenerative and is able to repair itself even after substantial damage; however, this ability to regenerate is lost with the development of dry eye conditions in chronically inflamed LGs.This study compares changes in the cell adhesion and cell matrix molecules and stem cell transcription factors in the LGs of healthy mice and of two mouse models of Sjögren's syndrome: nonobese diabetic (NOD) and MRL-lpr/lpr (MRL/lpr) mice during the early stage of inflammation. METHODS: The LGs from 12- to 13-week-old female MRL/lpr and male NOD mice along wi

[TOPICAL ANTI-INFLAMMATORY AGENTS FOR DRY EYE DISEASE].

Dry eye disease (dry eye) is a multifactorial disorder of the ocular surface. Dry eye is one of the most frequent ocular disorders, affecting 5% to 50% of the entire population at all ages. Evidence suggests that inflammation and hyperosmolarity are considered core mechanisms in the development of dry eye. Dry eye is accompanied by changes in tear composition including enhanced hyperosmolarity and secretion of pro-inflammatory mediators such as cytokines, chemokines, matrix metalloproteinases (MMPs) and adhesion molecules. All these factors may act as mediators of tissue damage leading to lysi

Structure and function of desmosomal proteins and their role in development and disease.

Desmosomes represent major intercellular adhesive junctions at basolateral membranes of epithelial cells and in other tissues. They mediate direct cell-cell contacts and provide anchorage sites for intermediate filaments important for the maintenance of tissue architecture. There is increasing evidence now that desmosomes in addition to a simple structural function have new roles in tissue morphogenesis and differentiation. Transmembrane glycoproteins of the cadherin superfamily of Ca(2+)-dependent cell-cell adhesion molecules which mediate direct intercellular interactions in desmosomes appea

Gastric mucosa alterations in first-degree relatives of gastric cancer patients.

BACKGROUND: First-degree relatives of gastric cancer patients have an increased risk of developing such neoplasia, and several alterations in gastric mucosa of these subjects have been described. On the other hand, both gastric cell hyperproliferation and abnormalities of adhesion molecules have been involved in gastric carcinogenesis. We studied gastric mucosa alterations in first-degree relatives of gastric cancer patients. PATIENTS AND METHODS: This prospective, case-controlled study enrolled 39 first-degree relatives of gastric cancer patients and 39 matched controls. Biopsy specimens obt

Adhesion molecules and cellular infiltrate: histology of urticaria.

Urticarial reactions are characterized by dermal capillary dilatation and edema, associated with a variably intense mixed inflammatory infiltrate consisting of neutrophils, eosinophils, T-helper lymphocytes, and activated macrophages. Mast cell numbers are moderately increased by a factor of 2.4, in contrast to mastocytosis where numbers are much higher (5-48-fold increase). In urticarial vasculitis there is in addition endothelial damage, leukocytoclasia, and fibrin and complement deposition. The emigration of leukocytes is regulated by vasoactive and chemotactic mediators released firom mast

Protein O-mannosylation is crucial for E-cadherin-mediated cell adhesion.

In recent years protein O-mannosylation has become a focus of attention as a pathomechanism underlying severe congenital muscular dystrophies associated with neuronal migration defects. A key feature of these disorders is the lack of O-mannosyl glycans on α-dystroglycan, resulting in abnormal basement membrane formation. Additional functions of O-mannosylation are still largely unknown. Here, we identify the essential cell-cell adhesion glycoprotein epithelial (E)-cadherin as an O-mannosylated protein and establish a functional link between O-mannosyl glycans and cadherin-mediated cell-cell ad

Changes in CD44 isoform expression during inflammatory skin disease.

The CD44 family of cell surface glycoproteins is widely expressed in epithelial, mesothelial and haemopoietic tissues and is thought to function primarily as adhesion molecules. The molecule has an intracellular, a transmembrane and an extracellular domain. The membrane proximal region of the extracellular domain is of variable structure depending on which of 10 variable exons are involved in coding for this region. Both in vitro stimulated T cells and cytokine stimulated keratinocytes are known to express certain isoforms. In this study we have investigated whether specific isoforms of the CD

Cellular interactions in self-directed immune-mediated liver diseases.

The lymphocyte population must traverse a complex path throughout their journey to the liver. The signals which these cells must detect, including cytokines, chemokines and other soluble factors, steer their course towards further crosstalk with other hepatic immune cells, hepatocytes and biliary epithelial cells. A series of specific chemokine receptors and adhesion molecules drive not only the recruitment, migration, and retention of these cells within the liver, but also their localisation. Perturbation of these interactions and failure of self-recognition drive the development of several a

[Epithelial ovarian tumor microecology].

Epithelial ovarian cancer remains an insidious and fatal gynecological malignancy. They are associated with a poor prognosis mainly due to a late diagnosis and to acquired chemoresistance. Cancer cell environment profoundly influences tumor development. Tumor microenvironment consists of vascular component, stromal fibroblasts, inflammatory cells and extracellular matrix. The multisite development of epithelial ovarian cancer results from molecular and cellular cross-talks between cancer cells, stromal cells and their extracellular matrix environment. These interactions involve cytokines, adhe

Circulating Extracellular Vesicles Are Increased in Newly Diagnosed Celiac Disease Patients.

Extracellular vesicles (EVs) are a class of circulating entities that are involved in intercellular crosstalk mechanisms, participating in homeostasis maintenance, and diseases. Celiac disease is a gluten-triggered immune-mediated disorder, characterized by the inflammatory insult of the enteric mucosa following local lymphocytic infiltration, resulting in villous atrophy. The goal of this research was the assessment and characterization of circulating EVs in celiac disease patients, as well as in patients already on an adequate gluten-free regimen (GFD). For this purpose, a novel and validate
