Chromoblastomycosis (CBM) is a chronic subcutaneous infection caused by genera of melanized fungi: Fonsecaea, Cladophialophora, Phialophora, Exophiala, and Rhinocladiella. Melanin is a virulence factor known to influence antifungal susceptibility. A specific inhibitor of melanin biosynthesis is tricyclazole. The aim of this study was to evaluate the effect of melanin inhibition on antifungal susceptibility of chromoblastomycosis agents and describe the susceptibility profiles of some unusual CBM agents. Seventy-six clinical isolates, representing 13 species of the five main genera of CBM agent

Amphotericin B (AMB) is considered the gold standard in the treatment of serious systemic mycoses in spite of its nephrotoxicity and adverse effects. Association with lipids enables larger doses of AMB to be given with a longer t((1/2)) and C(max), without the toxic effects at lower concentrations. Liposome-encapsulated AMB shows a lower affinity for mammalian cells and improves V(d), thus decreasing toxicity. Amphotericin B lipid complex (ABLC) is an AMB formulation associated with a biodegradable phospholipid matrix (5% molar) from which the drug is released by cell phospholipases. ABLC is r

The vaginal microbiota dysbiosis is closely associated with the development of reproductive diseases. However, the contribution of mycobiome to intrauterine adhesion (IUA) disease remains unknown. Harnessing 16S and ITS2 rDNA sequencing analysis, we investigate both bacterial and fungal microbiota compositions across 174 samples taken from both cervical canal (CC) and middle vagina (MV) sites of IUA patients. Overall, there is no significant difference in microbial diversity between healthy subjects (HS) and IUA patients. However, we observe the IUA-specific bacterial alterations such as incre

BACKGROUND: Aspergillus fumigatus (Af) infection is associated with poor lung health in chronic suppurative lung diseases but often goes undetected. We hypothesised that inhibition of Af growth by Pseudomonas aeruginosa (Pa) increases the frequency of false-negative Af culture in co-infected people. Using a substantial group of cystic fibrosis (CF) airway samples, we assessed the relationship between Af and bacterial pathogens, additionally comparing fungal culture with next-generation sequencing.
METHODS: Frequency of co-culture was assessed for 44,554 sputum/BAL cultures, from 1,367 CF pati

Lung infections with multiresistant pathogens are a major problem among patients suffering from cystic fibrosis (CF). N-Chlorotaurine (NCT), a microbicidal active chlorine compound with no development of resistance, is well tolerated upon inhalation. The aim of this study was to investigate the in vitro bactericidal and fungicidal activity of NCT in artificial sputum medium (ASM), which mimics the composition of CF mucus. The medium was inoculated with bacteria (Staphylococcus aureus, including some methicillin-resistant S. aureus [MRSA] strains, Pseudomonas aeruginosa, and Escherichia coli) o

BACKGROUND: Fungi colonize the human gut and might play a key role in the pathogenesis of ulcerative colitis (UC). However, studies on the fungal composition in the gut (especially adhering to the intestinal mucosa) of UC patients is limited.
RESULTS: The number of fungi decreased significantly in inflamed mucosa compared with that in HS mucosa. Fifteen major genera were examined, among which Wickerhamomyces, unidentified genus of Saccharomycetales, Aspergillus, Sterigmatomyces, and Candida showed increasing trends, whereas Exophiala, Alternaria, Emericella, Epicoccum, Acremonium, Trametes, a

Coastal ecosystems are dynamic environments characterized by strong seasonal variability in physicochemical parameters and biological communities. In the western Mediterranean, the Region of Murcia (southeastern Spain) is characterized by notable biodiversity and ecological heterogeneity, encompassing diverse habitats, including rocky shores, sandy beaches, seagrass meadows, and the Mar Menor, one of Europe's largest hypersaline lagoons. Within these systems, marine yeasts play critical ecological roles in organic matter recycling and produce metabolites of biotechnological and medical interes

Carrion decomposition is an important component of the ecosystem and plays a significant ecological role in the food chain. During decomposition, arthropods and microorganisms help to decompose and recycle organic matter and nutrients. Although research has been conducted on microorganisms and arthropods associated with carrion across different biogeoclimatic ecoregions, no study has identified these communities from carrion placed under a tropical rainforest. Therefore, this study aims to investigate the arthropod, bacterial, and fungal communities associated with Sus scrofa Linnaeus. carrion

Background: Metagenomic sequencing has the potential to identify a wide range of pathogens in human tissue samples. Sarcoidosis is a complex disorder whose etiology remains unknown and for which a variety of infectious causes have been hypothesized. We sought to conduct metagenomic sequencing on cases of ocular and periocular sarcoidosis, none of them with previously identified infectious causes. Methods: Archival tissue specimens of 16 subjects with biopsies of ocular and periocular tissues that were positive for non-caseating granulomas were used as cases. Four archival tissue specimens that

OBJECTIVES: Despite the significant improvement in survival rates due to antiretroviral therapy, opportunistic infections continue to pose a major health risk for individuals living with HIV. As a key anatomical intersection of the upper respiratory and digestive tracts, the pharyngeal microbiome during HIV infection remains poorly characterized. This study aimed to compare the composition of bacterial and fungal communities in the pharynx between HIV-infected individuals and healthy controls, and to investigate their association with host immune status. METHODS: Throat swab samples were colle

UNLABELLED: Dysbiosis of intratumoral fungal and bacterial communities is associated with poor prognosis in various cancers. However, the mycobiome characteristics in laryngeal squamous cell carcinoma (LSCC) and its correlation with recurrence have not yet been investigated. The mycobiome in 80 LSCC samples was characterized using internal transcribed spacer sequencing, encompassing both tumor tissues and their matched para-cancerous tissues. The intratumoral bacteriome was further identified using 16S rRNA sequencing. These two microbial communities were analyzed using bioinformatics and stat

BACKGROUND: New antifungal agents are required to mitigate against azole-resistant Aspergillus and drug-resistant non-Aspergillus moulds. The novel orotomide, olorofim (F2G, Manchester, UK), has potent fungicidal activity against Aspergillus including azole-resistant Aspergillus fumigatus, Lomentospora prolificans and Scedosporium spp. Development of olorofim-specific clinical breakpoints/epidemiological cut-off values requires reliable MIC data.
OBJECTIVES: Determine the in vitro activity of olorofim compared with standard antifungals against mould pathogens at an Australian hospital.
