Mutations of the quorum sensing-dependent regulator VjbR lead to drastic surface modifications in Brucella melitensis.

Successful establishment of infection by bacterial pathogens requires fine-tuning of virulence-related genes. Quorum sensing (QS) is a global regulation process based on the synthesis of, detection of, and response to small diffusible molecules, called N-acyl-homoserine lactones (AHL), in gram-negative bacteria. In numerous species, QS has been shown to regulate genes involved in the establishment of pathogenic interactions with the host. Brucella melitensis produces N-dodecanoyl homoserine lactones (C(12)-HSL), which down regulate the expression of flagellar genes and of the virB operon (enco

Transcriptomic analysis reveals a novel LysR regulator's role in Burkholderia pseudomallei host-pathogen interaction.

Burkholderia pseudomallei is a Gram-negative pathogen responsible for melioidosis, a life-threatening disease endemic to Southeast Asia. LysR-type transcriptional regulators (LTTRs) are known key regulators of bacterial pathogenesis and metabolism, yet many remain uncharacterized. This study investigates the function of UKMD286_5923, a novel LTTR, in B. pseudomallei UKMD286. We constructed a deletion mutant and performed transcriptomic analysis via RNA sequencing. This analysis identified 67 differentially expressed genes, with 45 genes upregulated and 22 genes downregulated in the mutant comp

Global control of GacA in secondary metabolism, primary metabolism, secretion systems, and motility in the rhizobacterium Pseudomonas aeruginosa M18.

The rhizobacterium Pseudomonas aeruginosa M18 can produce a broad spectrum of secondary metabolites, including the antibiotics pyoluteorin (Plt) and phenazine-1-carboxylic acid (PCA), hydrogen cyanide, and the siderophores pyoverdine and pyochelin. The antibiotic biosynthesis of M18 is coordinately controlled by multiple distinct regulatory pathways, of which the GacS/GacA system activates Plt biosynthesis but strongly downregulates PCA biosynthesis. Here, we investigated the global influence of a gacA mutation on the M18 transcriptome and related metabolic and physiological processes. Transcr

Global transcriptomic profiling and mutant analysis suggest linked 3-chlorobenzoate metabolism and activation of an integrative and conjugative element in Pseudomonas putida.

UNLABELLED: Conjugation, the exchange of genetic material between different bacterial species, is an important process underlying adaptation. The reason for this is that conjugative elements carry additional genes potentially providing adaptive benefit to the host, such as antibiotic resistance or pathways for degradation of environmental contaminants. However, there is increasing evidence that the conjugative process itself is also responsive to cellular or environmental cues and may, thus, be under rate-dependent selection. Here, we investigate the integrative and conjugative element ICEclc

Comparative transcriptomic analysis of pathogenic Vibrio parahaemolyticus YDE17 under iron-limited conditions.

Iron plays crucial roles in the growth, virulence, and environmental adaptation of bacterial pathogens. In this study, transcriptomic analysis was performed on the important aquaculture pathogen Vibrio parahaemolyticus grown under normal and iron-limited conditions. A total of 2258 differentially expressed genes (DEGs) were identified, including 1121 upregulated and 1137 downregulated genes. Gene Ontology (GO) analysis revealed that DEGs were mainly enriched in biological processes (BPs) such as peptide metabolic process, polypeptide biosynthetic process, and translation; in contrast, BPs rela

Antimicrobial activity and possible mechanisms of juglone against Escherichia coli, Staphylococcus aureus, and Salmonella pullorum.

BACKGROUND: Escherichia coli, Staphylococcus aureus, and Salmonella pullorum are significant pathogens that threaten livestock and poultry health. Although antibiotics and synthetic antimicrobial agents can combat these pathogens, antibiotic resistance remains a major concern. Recent decades have seen growing interest in antibiotic alternatives. Juglone, a natural naphthoquinone compound from Juglandaceae plant, exhibits strong antimicrobial activity against S. aureus. However, its antimicrobial mechanism is not yet fully understood. This study investigated the antimicrobial mechanism of juglo

Growth and gene expression of Pseudomonas nitroreducens TX1 on octylphenol polyethoxylate surfactants.

Pseudomonas nitroreducens ATCC PTA-6168 (also named TX1) efficiently grows on non-ionic surfactants as the sole source of carbon and energy aerobically. The global gene expression and bacterial stress responses during degradation remain further investigated. This study compared the growth, viability, and transcriptomic profiles of strain TX1 during the log phase when grown on minimal salts basal medium supplemented with 0.5% octylphenol polyethoxylates (OPEOn, commercial name Triton® X-100) vs. in 0.5% succinate. Differentially expressed genes were identified, with 219 upregulated and 22 downr

Cyclohexane, naphthalene, and diesel fuel increase oxidative stress, CYP153, sodA, and recA gene expression in Rhodococcus erythropolis.

In this study, we compared the expression of CYP153, sodA, sodC, and recA genes and ROS generation in hydrocarbon-degrading Rhodococcus erythropolis in the presence of cyclohexane, naphthalene, and diesel fuel. The expression of cytochrome P450, sodA (encoding Fe/Mn superoxide dismutase), recA, and superoxide anion radical generation rate increased after the addition of all studied hydrocarbons. The peak of CYP153, sodA, and recA gene expression was registered in the presence of naphthalene. The same substrate upregulated the Cu/Zn superoxide dismutase gene, sodC. Cyclohexane generated the hig

Coordination of symbiosis and cell cycle functions in Sinorhizobium meliloti.

The symbiotic nitrogen fixing species Sinorhizobium meliloti represents a remarkable model system for the class Alphaproteobacteria, which includes genera such as Caulobacter, Agrobacterium and Brucella. It is capable of living free in the soil, and is also able to establish a complex symbiosis with leguminous plants, during which its cell cycle program is completely rewired presumably due, at least in part, to the action of peptides secreted by the plant. Here we will discuss how the cell cycle regulation works in S. meliloti and the kinds of molecular mechanisms that take place during the in

The transcriptional regulator, CosR, controls compatible solute biosynthesis and transport, motility and biofilm formation in Vibrio cholerae.

Vibrio cholerae inhabits aquatic environments and colonizes the human digestive tract to cause the disease cholera. In these environments, V. cholerae copes with fluctuations in salinity and osmolarity by producing and transporting small, organic, highly soluble molecules called compatible solutes, which counteract extracellular osmotic pressure. Currently, it is unclear how V. cholerae regulates the expression of genes important for the biosynthesis or transport of compatible solutes in response to changing salinity or osmolarity conditions. Through a genome-wide transcriptional analysis of t

Multi-omics profiling reveals atypical sugar utilization and a key membrane composition regulator in Streptococcus pneumoniae.

The human body comprises many different microenvironments, each with their own challenges for microorganisms to overcome in order to survive and possibly cause infection. The human pathogen Streptococcus pneumoniae is notoriously flexible in this regard, and can adapt to a wide range of host niches, including the nasopharynx, lungs, and cerebrospinal fluid. However, the molecular and genetic foundation of this ability remain largely uncharted. In this work, we demonstrate that niche adaptation imposes genome-wide changes on multiple levels, including gene essentiality, expression and membrane

The transcript profile of persistent Chlamydophila (Chlamydia) pneumoniae in vitro depends on the means by which persistence is induced.

Expression of specific bacterial genes is differentially regulated during persistent, vs. active, chlamydial infection. Transcript patterns were examined using real-time reverse transcriptase-PCR in four in vitro models of persistence for Chlamydia pneumoniae strain CWL 029, using HeLa cells and normal human monocytes as host. Differential expression of genes encoding cell division proteins was variable when persistence was induced by interferon-gamma, penicillin G, or deferoxamine mesylate treatment, and in the monocyte model of persistence. Expression of genes encoding hsp60s and those speci

Virulence gene repression promotes Listeria monocytogenes systemic infection.

The capacity of bacterial pathogens to infect their hosts depends on the tight spatiotemporal regulation of virulence genes. The Listeria monocytogenes (Lm) metal efflux pump repressor CadC is highly expressed during late infection stages, modulating lipoprotein processing and host immune response. Here we investigate the potential of CadC as broad repressor of virulence genes. We show that CadC represses the expression of the bile salt hydrolase impairing Lm resistance to bile. During late infection, in absence of CadC-dependent repression, the constitutive bile salt hydrolase expression indu

Rrp1, a cyclic-di-GMP-producing response regulator, is an important regulator of Borrelia burgdorferi core cellular functions.

Two-component systems (TCS) are universal among bacteria and play critical roles in gene regulation. Our understanding of the contributions of TCS in the biology of the Borrelia is just now beginning to develop. Borrelia burgdorferi, a causative agent of Lyme disease, harbours a TCS comprised of open reading frames (ORFs) BB0419 and BB0420. BB0419 encodes a response regulator designated Rrp1, and BB0420 encodes a hybrid histidine kinase-response regulator designated Hpk1. Rrp1, which contains a conserved GGDEF domain, undergoes phosphorylation and produces the secondary messenger, cyclic digua

Analysis of Teg41 and PSMα promoter activity using a divergent fluorescent reporter plasmid.

Staphylococcus aureus is a common commensal of human skin and anterior nares but also a notable pathogen, causing a range of diseases from minor skin infections to serious, often fatal conditions. The global health community considers S. aureus a serious threat due to the rise of multidrug-resistant strains, including community-acquired methicillin-resistant S. aureus. Effective host tissue invasion by S. aureus requires precise spatiotemporal regulation of virulence factor expression, which can be controlled by small RNAs (sRNAs). Our lab has identified over 300 potential sRNAs in S. aureus U

Transcriptomic insights into the antimicrobial effect of Pediococcus pentosaceus on multidrug-resistant Pseudomonas aeruginosa.

Antimicrobial resistance in Pseudomonas aeruginosa is a critical global health challenge. In this study, we profiled the transcriptomic response of multidrug resistant P. aeruginosa PA01 to the cell-free supernatant of Pediococcus pentosaceus ENM104. Differential expression analysis using edgeR (|log₂FC| ≥ 2, FDR ≤ 0.05) identified a high-confidence set of 952 core differentially expressed genes (492 upregulated, 460 downregulated). Functional enrichment (GO/KEGG) and STRING v12.0 mapping revealed a shift prioritizing protein synthesis and envelope defense with induction of ribosomal proteins,

Transcriptomic microarray analysis of corrinoid responsive genes in Dehalococcoides ethenogenes strain 195.

Dehalococcoides ethenogenes strain 195 utilizes corrinoid-containing reductive dehalogenases to reduce the environmental pollutants tetrachloroethene and trichloroethene to ethene. Although corrinoids are essential for dehalogenation activity, strain 195 cannot biosynthesize corrinoids de novo. To improve our understanding of corrinoid physiology in this bacterium, whole-genome microarrays were applied to characterize the transcriptome during growth with excess and limiting concentrations of the corrinoid cyanocobalamin. Additional studies examined the effects of exposure to spent medium from

Mechanisms of thermoregulation in Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a highly versatile opportunistic pathogen and a major cause of acute, chronic, and even lifelong respiratory infections in people with cystic fibrosis. It can also cause corneal infections, burn/wound infections, and bacteremia. P. aeruginosa is often found in human-associated environments such as hospitals, where it is a frequent cause of nosocomial catheter-associated urinary tract infections and ventilator-associated pneumonia. As a nosocomial pathogen, a major environmental change associated with transmission is the change from room or ambient temperature to human

A metalloprotease secreted by the type II secretion system links Vibrio cholerae with collagen.

Vibrio cholerae is autochthonous to various aquatic niches and is the etiological agent of the life-threatening diarrheal disease cholera. The persistence of V. cholerae in natural habitats is a crucial factor in the epidemiology of cholera. In contrast to the well-studied V. cholerae-chitin connection, scarce information is available about the factors employed by the bacteria for the interaction with collagens. Collagens might serve as biologically relevant substrates, because they are the most abundant protein constituents of metazoan tissues and V. cholerae has been identified in associatio

Repurposing anti-phage defenses to differentially arrest the viral lifecycle reveals the regulatory logic of a parasitic satellite.

Mobile genetic elements frequently encode defense mechanisms to protect their bacterial hosts from viral attack. In Vibrio cholerae, these defensive elements include phage-inducible chromosomal island-like elements (PLEs), which are phage satellites that act as highly specialized parasites of the lytic phage ICP1. While PLE transcriptional activation upon ICP1 infection is known to be temporally regulated, the underlying regulatory logic and dependencies on the progression of the phage's developmental program required for activation remain unclear. In this study, we took a novel approach to de
