Holistic view of understanding genetic predisposition and perceptions of genomic research in African communities: tradition, trust and transformation.

Genetic research holds immense potential to advance personalised medicine and enhance patient outcomes. However, Africa’s diverse and historically marginalised populations remain significantly underrepresented in these studies, thus limiting the relevance and reach of genomic advancements. By examining African communities through an anthropological lens, this narrative review highlights the importance of their lived realities and holistic perspectives on Genetic and Genomic Research (GGR). Beyond advocating for their inclusion, raising the importance of embedding intentional integrated medical

Mapping the evidence and research gaps on preimplantation genetic testing for aneuploidy in recurrent implantation failure: a scoping review.

BACKGROUND: Recurrent implantation failure (RIF) represents a complex and heterogeneous condition in assisted reproductive technology (ART), characterized by unclear etiology and limited evidence-based management strategies. Preimplantation genetic testing for aneuploidy (PGT-A) has been proposed to improve reproductive outcomes through selection of euploid embryos; however, its clinical utility in RIF remains controversial. METHODS: A scoping review was conducted to systematically map the available evidence regarding the role of PGT-A in RIF. A comprehensive literature search identified 18 s

Triangulating the link between depression and liver disease: An integrative analysis of genetic, observational, and research trend data.

BACKGROUND: The relationship between major depressive disorder (MDD) and liver diseases is complex, with causal links poorly understood. This study aimed to disentangle causal, mediated, and confounded links between MDD and liver diseases. METHODS: We integrated three approaches: a trend analysis of 2004 to 2024 publications; a two-sample Mendelian randomization, or MR, analysis assessing genetic causal effects of MDD on liver diseases; and an observational analysis of the NHANES 2007 to 2020 cohort using survey-weighted regression and formal causal mediation analysis to evaluate clinical pat

Disclosure of onset-predictive biomarker results to research participants at risk of genetic frontotemporal dementia: a European perspective.

BACKGROUND: As understanding of biomarkers for genetic frontotemporal dementia (FTD) advances, there is a need to develop onset-predictive biomarker tests (OPBTs) to detect changes before the onset of symptoms. OPBTs can be used to recruit carriers or individuals at 50% risk of carriership into clinical trials of investigational therapies targeting the preclinical and prodromal phases of FTD. OPBT results should be disclosed as part of the informed consent process, with positive results indicating that symptom onset is likely in the next few years. This information can be psychologically burde

Employing Informatics Strategies in Alzheimer's Disease Research: A Review from Genetics, Multiomics, and Biomarkers to Clinical Outcomes.

Alzheimer's disease (AD) is a critical national concern, affecting 5.8 million people and costing more than $250 billion annually. However, there is no available cure. Thus, effective strategies are in urgent need to discover AD biomarkers for disease early detection and drug development. In this review, we study AD from a biomedical data scientist perspective to discuss the four fundamental components in AD research: genetics (G), molecular multiomics (M), multimodal imaging biomarkers (B), and clinical outcomes (O) (collectively referred to as the GMBO framework). We provide a comprehensive

Research on genetic variant characteristics in ADME genes based on whole-exome sequencing in the Han Chinese population.

BACKGROUND: Genetic variants in absorption, distribution, metabolism, and excretion (ADME) genes affect drug efficacy and side effects. Whole-exome sequencing (WES) effectively identifies these variants. Findings from Western populations may not apply to the Han Chinese population due to ethnic differences. OBJECTIVE: This study aimed to investigate genetic variants, metabolic phenotypes, and drug impacts related to ADME genes in the Han Chinese population using WES data. METHODS: ADME genes and drug profiles were sourced from multiple databases. WES data were collected from 357 samples at T

Global research landscape on the genetics of congenital heart disease: A bibliometric and visualized analysis via VOSviewer and CiteSpace.

Genetic factors play a significant role in the development of congenital heart disease (CHD). Many studies on the genetics of CHD have been published worldwide; however, no research has assessed and mapped the global research landscape of these studies. This bibliometric and visualized study aimed to delineate research hotspots and trends in the field of CHD genetics. Scientific papers on the genetics of CHD from January 1, 1950, to December 31, 2023, were obtained by searching the Web of Science Core Collection. The bibliometric metadata of each chosen research paper were extracted, analyzed,

Centralization of care for rare genetic syndromes associated with cancer: improving outcomes and advancing research on VHL disease.

Von Hippel-Lindau (VHL) disease is a rare genetic syndrome caused by a germline pathogenic variant in one VHL allele. Any somatic event disrupting the other allele induces VHL protein (pVHL) loss of function, ultimately leading to patients developing multiple tumours in multiple organs at multiple timepoints, and reducing life expectancy. Treatment of this complex, rare disease is often fragmented, as patients visit specialist clinicians in isolation at different medical centres. Consequently, patients can receive sub-optimal treatment that results in decreased quality of life and a poor exper

The Incontinentia Pigmenti Genetic Biobank: study design and cohort profile to facilitate research into a rare disease worldwide.

Incontinentia pigmenti (IP; OMIM#308300) is a rare genetic disease resulting in neuroectodermal defects, which can lead to disability. At present, there is neither definitive cure available nor are there any sufficiently reliable insights to predict the severity of the disease. We launched the Incontinentia Pigmenti Genetic Biobank (IPGB) project ( http://www.igb.cnr.it/ipgb ) in 2015 to establish a large-scale deposit of biological samples, to provide detailed clinical information about children diagnosed with IP and to facilitate research. We have built a cohort comprising samples of 381 cli

Connecting patients, researchers and clinical genetics services: the experiences of participants in the Australian Ovarian Cancer Study (AOCS).

Population-based genetic research may produce information that has clinical implications for participants and their family. Researchers notify participants or their next of kin (NoK) about the availability of genetic information via a notification letter; however, many subsequently do not contact a family cancer centre (FCC) to clarify their genetic status. Therefore, the purpose of this study was to examine research participants' experience of receiving a notification letter and the factors that influenced contact with an FCC. Twenty-five semi-structured interviews were conducted with researc

The genetics of auricular development and malformation: new findings in model systems driving future directions for microtia research.

Microtia is a term used to describe a wide array of phenotypic presentations of the outer ear. Although the majority of the cases are isolated in nature, much of our understanding of the causes of microtia has been driven by the identification of genes underlying syndromic forms where the anomaly co-presents with various other craniofacial and extra-craniofacial structural defects. In this review we discuss recent findings in mice deficient in Hoxa2, a key regulator of branchial arch patterning, which has necessitated a revision to the canonical model of pinna morphogenesis. The revised model

Four new speech and prosody-voice measures for genetics research and other studies in developmental phonological disorders.

Research in developmental phonological disorders, particularly emerging subgroup studies using behavioral and molecular genetics, requires qualitative and continuous measurement systems that meet a variety of substantive and psychometric assumptions. This paper reviews relevant issues underlying such needs and presents four measurement proposals developed expressly for causal-correlates research. The primary qualitative system is the Speech Disorders Classification System (SDCS), a 10-category nosology for dichotomous and hierarchical polychotomous classification of speech disorders from 2 yea

[Research on genetic abnormality in the hemolytic form of hereditary elliptocytosis with homozygosity for the spectrin alpha I/74 variant].

We have undertaken to identify the spectrin gene mutation in a patient with a severe hemolytic form of Hereditary Elliptocytosis with homozygosity for the spectrin alpha I/74 variant. This variant corresponds to the presence of a 74,000 peptide which is produced during mild tryptic digestion of spectrin by cleavage at the Arginine-39 of the alpha I/80,000 domain of the spectrin alpha chain (595 amino acids). We hypothesized that the alpha I/74 mutation would be closed to the cleavage site Arg-39. A genomic library built with the patient's DNA was screened with a probe corresponding to a fragme

Research note: Molecular characteristics and genetic evolution of H1N1 avian influenza virus from wild birds in Shanghai, China.

The H1N1 influenza virus is a major pandemic and seasonal pathogen with a broad host range, posing a substantial threat to human health and underscoring the need for continuous surveillance. Wild birds, as natural reservoirs of avian influenza viruses (AIVs), carry H1N1 strains capable of reassorting with other influenza viruses, which can drive pandemic emergence. The global migration of wild birds facilitates the spread of these viruses, and their interactions with poultry increase the risk of cross-species transmission, further amplifying the public health threat. However, knowledge of H1N1

Genetic parameters of rumination time and feed efficiency traits in primiparous Holstein cows under research and commercial conditions.

Feed efficiency has the potential to be improved both through feeding, management, and breeding. Including feed efficiency in a selection index is limited by the fact that dry matter intake (DMI) recording is only feasible under research facilities, resulting in small data sets and, consequently, uncertain genetic parameter estimates. As a result, the need to record DMI indicator traits on a larger scale exists. Rumination time (RT), which is already recorded in commercial dairy herds by a sensor-based system, has been suggested as a potential DMI indicator. However, RT can only be a DMI indic
