Irreversible blockade of D2 dopamine receptors by fluphenazine-N-mustard increases glutamic acid decarboxylase mRNA in rat striatum.

The influence of dopaminergic activity on the function of GABAergic neurons in striatum was examined by administering rats the irreversible D2 dopamine receptor antagonist, fluphenazine-N-mustard (FNM), and determining the level of glutamic acid decarboxylase (GAD) mRNA in striatum. Rats were given either an acute single injection or chronic daily injections of FNM (20 mumol/kg, i.p.) for 6 days. The level of GAD mRNA in striatum was determined by in situ hybridization histochemistry. The results showed that acute treatment with FNM failed to significantly change striatal GAD mRNA. However, ch…

TRH stimulation and L-glutamic acid inhibition of proximal gastric motor activity in the rat dorsal vagal complex.

The importance of the dorsal vagal complex (DVC) in the control of gastric motor activity has been previously established by electrical and chemical stimulation of this region. We have further evaluated excitatory and inhibitory influences on motor activity of the gastric corpus by microinjection of L-glutamic acid (GLU) and thyrotropin-releasing hormone (TRH) into the DVC. GLU and TRH were ejected by pressure (20-30 psi) in 1-10 nl vol. from multibarreled micropipettes and intraluminal pressure in the gastric corpus was measured using a manometric catheter placed into the stomach through the …

Valproic acid induces the glutamate transporter excitatory amino acid transporter-3 in human oligodendroglioma cells.

Glutamate transport in early, undifferentiated oligodendrocytic precursors has not been characterized thus far. Here we show that human oligodendroglioma Hs683 cells are not endowed with EAAT-dependent anionic amino acid transport. However, in these cells, but not in U373 human glioblastoma cells, valproic acid (VPA), an inhibitor of histone deacetylases, markedly induces SLC1A1 mRNA, which encodes for the glutamate transporter EAAT3. The effect is detectable after 8h and persists up to 120h of treatment. EAAT3 protein increase becomes detectable after 24h of treatment and reaches its maximum

Reversible Isolated Opsoclonus in a Patient With Gastric Cancer and Antibodies to Glutamic Acid Decarboxylase and Myelin Oligodendrocyte Glycoprotein.

Opsoclonus and myoclonus are known paraneoplastic manifestations of malignancy. The most common antibodies found are anti-neuronal antibodies (Hu, Ri, and Ma). The most common affiliated malignancies are small-cell lung cancers, breast cancers, or gynecological malignancies. Gastric cancer is uncommon. Other antibodies have been cited, but are rare. Isolated opsoclonus without significant myoclonus is also rare. While both glutamic acid decarboxylase (GAD) and myelin oligodendrocyte glycoprotein (MOG) antibodies have individually been reported to be associated with opsoclonus, reports of a com…

Stiff tongue syndrome and multifocal immune neuropathy in a young man with anti-glutamic acid decarboxylase and unclassified antibodies.

Anti-glutamic acid decarboxylase 65 antibody (anti-GAD65Ab)-mediated neurological syndromes are constantly expanding with recent literature descriptions of peripheral nerve involvement. This report describes a man in his 30s presenting with progressive episodic tongue stiffness and upper limb weakness, with examination revealing restricted tongue movement and asymmetric proximal predominant upper limb weakness. Electrophysiology showed conduction blocks, and imaging revealed inflammatory brachial plexus changes. Anti-GAD65Ab and an unclassified neuronal antibody were identified on serological …

Transient Anti-Glutamic Acid Decarboxylase (GAD) Antibody Positivity and Insulin Secretion Reduction After IVIg in Type 2 Diabetes Mellitus.

This report describes a patient with type 2 diabetes mellitus who received intravenous immunoglobulin therapy for Guillain-Barré syndrome and subsequently experienced a sudden deterioration in glycemic control. Although anti-glutamate decarboxylase antibodies became positive and insulin secretion was transiently suppressed, both findings returned to baseline during the follow-up period. These changes initially raised concerns about new-onset or coexisting type 1 diabetes mellitus; however, the reversibility of anti-glutamate decarboxylase antibody positivity and the eventual restoration of ins

Efficient pretreatment of Phragmites australis biomass using glutamic acid for bioethanol production by a hybrid hydrolysis and fermentation strategy.

Microbial fermentation of renewable lignocellulosic biomass to produce biofuels presents significant environmental advantages. The conversion of cellulose and hemicellulose into fermentable sugars provides essential carbon sources for microbial metabolism. However, the recalcitrance of biomass limits enzymatic accessibility. In this study, mild L-glutamic acid (GA) pretreatment was applied to Phragmites australis residues (reed straw) to fractionate lignin and polysaccharides for enhancing enzymatic hydrolysis. Pretreatment with 0.20 mol/L GA at 180 °C for 50 min (logRo = 4.1) achieved glucan …

Sex differences in behavior and glutamic acid decarboxylase in Long Evans rats after prolonged social isolation beginning in adolescence.

Social isolation can have long-term effects on brain development and behavior and increases the risk of developing clinical conditions, including anxiety disorders. One modulator of the stress response is gamma-aminobutyric acid, an inhibitory neurotransmitter synthesized by glutamic acid decarboxylase (GAD). This study examined sex differences in behavior and GAD expression following prolonged social isolation beginning in adolescence in Long Evans rats. Males and females were equally divided into group-housed (GH) and socially isolated conditions on Postnatal Day 28 (n = 8 per group). Beginn…

Human Innate Immune Cells Respond Differentially to Poly-γ-Glutamic Acid Polymers from Bacillus anthracis and Nonpathogenic Bacillus Species.

The poly-γ-glutamic acid (PGA) capsule produced by Bacillus anthracis is composed entirely of d-isomer glutamic acid, whereas nonpathogenic Bacillus species produce mixed d-, l-isomer PGAs. To determine if B. anthracis PGA confers a pathogenic advantage over other PGAs, we compared the responses of human innate immune cells to B. anthracis PGA and PGAs from nonpathogenic B. subtilis subsp. chungkookjang and B. licheniformis Monocytes and immature dendritic cells (iDCs) responded differentially to the PGAs, with B. anthracis PGA being least stimulatory and B. licheniformis PGA most stimulatory.…

Lower expression of glutamic acid decarboxylase 67 in the prefrontal cortex in schizophrenia: contribution of altered regulation by Zif268.

OBJECTIVE: Cognitive deficits of schizophrenia may be due at least in part to lower expression of the 67-kDa isoform of glutamic acid decarboxylase (GAD67), a key enzyme for GABA synthesis, in the dorsolateral prefrontal cortex of individuals with schizophrenia. However, little is known about the molecular regulation of lower cortical GAD67 levels in schizophrenia. The GAD67 promoter region contains a conserved Zif268 binding site, and Zif268 activation is accompanied by increased GAD67 expression. Thus, altered expression of the immediate early gene Zif268 may contribute to lower levels of GA…

Novel thermo- and pH-responsive hydroxypropyl cellulose- and poly (L-glutamic acid)-based microgels for oral insulin controlled release.

Novel smart microgel particles made of poly (L-glutamic acid-2-hydroxylethyl methacrylate) (PGH) and hydroxypropyl cellulose-acrylic acid (HPC-AA) have been successfully prepared via emulsion polymerization. The dynamic light scattering measurement reveals that the average hydrodynamic radius 〈Rh〉 and hydrodynamic radius distributions f (Rh) of the microgel particles depend on the temperature and pH value thus the microgel particles exhibit both pH- and temperature-sensitivity. In vitro release study shows that the amount of insulin released from microgels in the gastric juice (at pH 1.2) is s…

Proline-proline-glutamic acid (PPE) protein Rv1168c of Mycobacterium tuberculosis augments transcription from HIV-1 long terminal repeat promoter.

Cells of the monocyte/macrophage lineage are shown to play a role in the pathogenesis of human immunodeficiency virus (HIV). The occurrence of HIV type 1 (HIV-1) infection is found to be accelerated in people infected with Mycobacterium tuberculosis, but the mechanism by which mycobacterial protein(s) induces HIV-1 LTR trans-activation is not clearly understood. We show here that the M. tuberculosis proline-proline-glutamic acid (PPE) protein Rv1168c (PPE17) can augment transcription from HIV-1 LTR in monocyte/macrophage cells. Rv1168c interacts specifically with Toll-like receptor-2 (TLR2) re…

Anti-glutamic Acid decarboxylase antibody-associated ataxia as an extrahepatic autoimmune manifestation of hepatitis C infection: a case report.

Extrahepatic immunological manifestations of hepatitis C virus (HCV) are well described. In addition, antiglutamic acid decarboxylase (GAD) antibody-associated cerebellar ataxia is well-established entity. However, there have been no reports in the literature of anti-GAD antibody-associated ataxia as an extrahepatic manifestation of HCV infection. We report the case of a young woman with chronic hepatitis C virus and multiple extrahepatic autoimmune diseases including Sjögren syndrome and pernicious anemia who presented with subacute midline cerebellar syndrome and was found to have positive a…

Congenital alterations of NEMO glutamic acid 223 result in hypohidrotic ectodermal dysplasia and immunodeficiency with normal serum IgG levels.

BACKGROUND: Hypomorphic mutations in the nuclear factor-κB (NF-κB) essential modulator (NEMO) gene result in a variable syndrome of somatic … atient and a substitution (667G>A) in the other. These findings predict either the deletion of NEMO glutamic acid 223 or it being replaced with lysine, respectively. Both patients had normal serum IgG levels but poor specific antibodies. Natural killer cell cytotoxicity and Toll-like and tumor necrosis factor receptor signaling were also impaired. Serious bacte …

Herpes simplex virus vector-mediated gene delivery of glutamic acid decarboxylase reduces detrusor overactivity in spinal cord-injured rats.

We examined whether replication-defective herpes simplex virus (HSV) vectors encoding the 67 kDa form of the glutamic acid decarboxylase (GAD(67)) gene product, the gamma-aminobutyric acid (GABA) synthesis enzyme, can suppress detrusor overactivity (DO) in rats with spinal cord injury (SCI). One week after spinalization, HSV vectors expressing GAD and green fluorescent protein (GFP) (HSV-GAD) were injected into the bladder wall. Rats with SCI without HSV injection (HSV-untreated) and those injected with lacZ-encoding reporter gene HSV vectors (HSV-LacZ) were used as controls. Three weeks after…
