Bilateral hypertrophic olivary nucleus degeneration on magnetic resonance imaging in children with Leigh and Leigh-like syndrome.

OBJECTIVE: Bilateral hypertrophic olivary degeneration on brain MRI has been reported in a few metabolic, genetic and neurodegenerative disorders, including mitochondrial disorders. In this report, we sought to analyse whether bilateral symmetrical inferior olivary nucleus hypertrophy is specifically associated with mitochondrial disorders in children. METHODS: This retrospective study included 125 children (mean age, 7.6 ± 5 years; male:female, 2.6:1) diagnosed with various metabolic and genetic disorders during 2005-2012. The routine MRI sequences (T1 weighted, T2 weighted and fluid-attenua

Cystic renal dysplasia as a leading sign of inherited metabolic disease.

Glutaric acidemia type II and carnitine palmitoyltransferase type II deficiency are rare, but potentially treatable, inherited metabolic diseases. Hallmarks of the early onset form of both conditions are renal abnormalities and neonatal metabolic crisis. In this article, we report on two newborns with cystic renal dysplasia as a leading sign of these metabolic diseases. We focus on the clinical presentation and discuss the diagnostic tests and the available therapeutic options. We conclude that prenatal diagnosis of cystic renal dysplasia should alert the physician to the possibility of these

Inherited metabolic disorders and cerebral infarction.

The association of genetic factors and cerebral infarction (CI) has long been established. A positive family history alone is a recognized risk factor for CI and vascular events in general. However, there are certain inherited conditions that further increase the risk of stroke. These conditions are generally metabolic and mitochondrial genetic defects that have variable modes of inheritance. This article reviews major inherited metabolic disorders that predispose an individual to CI. Ten main conditions will be discussed: Fabry's disease, cerebrotendinous xanthomatosis, tangier disease, famil

Acute bilateral striatal necrosis with rotavirus gastroenteritis and inborn metabolic predisposition.

We present a 9-month-old male with acute rotavirus gastroenteritis who developed an acute encephalopathy with focal seizures and developmental regression. Magnetic resonance imaging showed bilateral striatal necrosis and raised glutarylcarnitine levels on tandem mass spectrometry of a (crisis) blood spot, and chromatography of organic acids revealed increased urinary excretion of dicarboxylic acid. Skin biopsy demonstrated a partial decrease in glutaryl-CoA dehydrogenase activity. The case was not typical for either rotavirus encephalitis/rotavirus-associated encephalopathy or for glutaric aci

[The blocking action of snake venom polypeptides on the cholinergic mechanisms of leech dorsal muscle].

The blocking action of alpha-polypeptides from the venoms of elapid snakes (Bungarus multicinctus and Naja naja siamensis) was studied on leech dorsal muscle. It was shown that these neurotoxins failed to inhibit the responses to mononitrogen cholinomimetics (acetylcholine, nicotine, carbamylcholine, etc.). The capacity of neurotoxins to inhibit the responses to the bisquaternary cholinommimetics depended upon the length of the molecule. The neurotoxins were active against the "shorter" cholinomimetics, such as decamethonium, and bischolinic esters of malonic, succinic and glutaric acids. The

Synthesis of Novel Carborane-Containing Derivatives of RGD Peptide.

Short peptides containing the Arg-Gly-Asp (RGD) fragment can selectively bind to integrins on the surface of tumor cells and are attractive transport molecules for the targeted delivery of therapeutic and diagnostic agents to tumors (for example, glioblastoma). We have demonstrated the possibility of obtaining the N- and C-protected RGD peptide containing 3-amino-closo-carborane and a glutaric acid residue as a linker fragment. The resulting carboranyl derivatives of the protected RGD peptide are of interest as starting compounds in the synthesis of unprotected or selectively protected peptide

Impairment of neuromotor development and cognition associated with histopathological and neurochemical abnormalities in the cerebral cortex and striatum of glutaryl-CoA dehydrogenase deficient mice.

Patients with glutaric acidemia type I (GA I) manifest motor and intellectual disabilities whose pathogenesis has been so far poorly explored. Therefore, we evaluated neuromotor and cognitive abilities, as well as histopathological and immunohistochemical features in the cerebral cortex and striatum of glutaryl-CoA dehydrogenase (GCDH) deficient knockout mice (Gcdh-/-), a well-recognized model of GA I. The effects of a single intracerebroventricular glutaric acid (GA) injection in one-day-old pups on the same neurobehavioral and histopathological/immunohistochemical endpoints were also investi

Self-Healing Approach toward Catalytic Soft Robots.

Soft robotics is a rapidly evolving research field that focuses on developing robots with bioinspired actuation/sensing mechanisms and highly flexible soft materials, some of which are similar to those found in living organisms. The hydrogel has the characteristics of excellent biocompatibility, softness, and elasticity, which makes it an ideal candidate material for the preparation of soft robots. Here we utilized a self-healing approach to develop a catalytically driven soft robot, which was constructed by dynamic imine bonds between modular hydrogels. One of the modules was a hydrogel forme

An Analysis of the Effectiveness of High-level Disinfection for Surgical Instruments Used by Department of Defense Austere Surgical Teams.

INTRODUCTION: The purpose of this investigation was to evaluate the efficacy of currently employed commercial disinfectants in a simulated austere surgical environment similarly faced by ground surgical teams in forward deployed positions. Severe contamination of traumatic combat wounds along with limitations of operations in austere environments may result in available disinfectants providing inadequate surgical instrument decontamination. MATERIALS AND METHODS: The study consisted of nine experimental groups and two control groups evaluating hemostatic forceps found in kits of ground surgic

The development and application of a novel reagent for fixing red blood cells with glutaraldehyde and paraformaldehyde.

OBJECTIVE: The currently employed red blood cell reagents have a short shelf life. Some hospitals with a small number of specimens will be unable to utilize them within the validity period, resulting in a substantial increase in the purchase price. Therefore, the method of developing long-term red blood cell reagents is a problem worthy of further study. METHODS: In this experiment, the type and concentration of the red blood cell reagent treatment solution were evaluated based on the red blood cell antigen concentration 24 h after treatment. In addition, the qualified glutaraldehyde/paraform

Novel bio-inspired three-dimensional nanocomposites based on montmorillonite and chitosan.

In this study, inspired by nacre-like structural natural shells, novel three-dimensional (3D) nanocomposites based on natural nanoplatelets of montmorillonite (MMT) and polysaccharide of chitosan (CS) were prepared with solution intercalation and self-assembly process. The CS-intercalated-MMT nanoplatelets units acted as "bricks" and CS molecules acted as "mortar", arranging in fairly well-ordered layered structure. With addition of glutaraldehyde (GA) and Pd2+ cations, synergistic toughening and strengthening effects of covalent and ionic bonds could be achieved. The best mechanical propertie

Multiple acyl-COA dehydrogenase deficiency in elderly carriers.

Multiple acyl-CoA dehydrogenase deficiency, or glutaric aciduria type II, is an autosomal recessive disorder of fatty acid oxidation due to defects in electron transfer flavoprotein (ETF) encoded by ETFA and ETFB, or in electron transfer flavoprotein dehydrogenase (ETFDH) encoded by the ETFDH gene. The disease may present as a severe neonatal onset form and a mild late-onset form which is heterogeneous for the age at onset and clinical presentation. We describe two patients in their seventies, referred for a nonspecific myopathy, which resulted to manifest carriers of ETFDH gene mutation. Trea

[Indirect hemagglutination via glutaraldehyde-treated erythrocytes for determining anti-anthrax antibodies].

The so-called passive hemagglutination reaction is used in this country to determine the serum level of anthrax antibodies in immunized individuals. Use is made of an erythrocyte diagnostic means prepared from formalin- and tannin-treated sheep erythrocytes that have been sensibilized with a protective anthrax antigen. An attempt has been made to replace this means by another one obtained by treating the same erythrocytes with glutaric aldehyde. Comparative studies reveal that the new diagnostic means is equally effective in the passive hemaglutination reaction as is the classic method using f

Dried Blood Spot-Based Monitoring of Dietary Treatment in Children, Adolescents, and Young Adults with Inherited Disorders of Amino Acid Metabolism: A Four-Year Pilot Study.

Background: Regular biochemical monitoring is essential for the safe and effective dietary treatment of inherited disorders of amino acid metabolism. However, conventional venous blood sampling may limit monitoring frequency and increase the burden on families. This study evaluated the feasibility and clinical usefulness of home-collected dried blood spot (DBS) samples analyzed using tandem mass spectrometry and additional chromatographic methods for dietary monitoring in inherited disorders of amino acid metabolism other than phenylketonuria. Methods: Between 2022 and 2025, children, adolesce

Electron transfer flavoprotein: ubiquinone oxidoreductase (ETF:QO) deficiency in an adult.

A 19-year-old woman with mild myopathic symptoms from age 6 and fasting intolerance presented with a Reye-like syndrome and a myopathy. Investigations disclosed a lipid storage myopathy, type II glutaric acidemia, and carnitine deficiency in skeletal muscle. Riboflavin and carnitine treatment corrected the metabolic abnormalities and she improved clinically. She later died from pulmonary complications secondary to aspiration. Subsequent studies established electron transfer flavoprotein: ubiquinone oxidoreductase (ETF:QO) deficiency (fibroblast ETF:QO activity was 2.9 mU/mg, normal range is 14

[Use of immunospecific components labelled with beta-lactamase and horseradish peroxidase for immunoenzyme analysis].

A system for enzyme immunoassay with the use of beta-lactamase as an enzyme marker is described. Similar sensitivity of the enzyme immunoassays with the use of beta-lactamase and horse-raddish peroxidase as enzyme markers was shown. The data on decrease of the antigen binding capacity of the antibodies in the conjugates with beta-lactamase are presented. It is suggested that such a decrease may be associated with glutaric dialdehyde conjugation. The use of the schemes for enzyme immunoassay with simultaneous application of two independently recorded enzymes i. e. horse-raddish peroxidase and b
