Occurrence of cross-resistance and β-lactam seesaw effect in glycopeptide-, lipopeptide- and lipoglycopeptide-resistant MRSA correlates with membrane phosphatidylglycerol levels.

BACKGROUND: Glycopeptides (GPs), lipopeptides (LPs) and lipoglycopeptides (LGPs) are related antimicrobials important for the management of invasive MRSA infections. Cross-resistance among these antibiotics in MRSA is well documented, as is the observation that susceptibility of MRSA to β-lactams increases as susceptibility to GPs and LPs decreases (i.e. the seesaw effect). Efforts to understand the relationship between GP/LP/LGP cross-resistance and the seesaw effect have focused on the PBPs, but the role of lipid metabolism has not been investigated. OBJECTIVES: Since the cell membrane is s…

Structure-based development of an osteoprotegerin-like glycopeptide that blocks RANKL/RANK interactions and reduces ovariectomy-induced bone loss in mice.

Osteoporosis is a metabolic bone disease characterized by low bone mass and micro-architectural deterioration of bone, for which the underlying mechanism is an imbalance between bone resorption and bone remodeling. The protein-protein interactions between receptor activator of nuclear factor-κB ligand (RANKL), RANK (its receptor), and osteoprotegerin (OPG), are known to mediate the development and activation of osteoclasts in bone remodeling, and are regarded as a pivotal therapeutic target for the treatment of osteoporosis. Herein, we disclose the successful development of a novel glycopeptid

Characterization of structural variations in the peptidoglycan of vancomycin-susceptible Enterococcus faecium: understanding glycopeptide-antibiotic binding sites using mass spectrometry.

Enterococcus faecium, an opportunistic pathogen that causes a significant number of hospital-acquired infections each year, presents a serious clinical challenge because an increasing number of infections are resistant to the so-called antibiotic of last resort, vancomycin. Vancomycin and other new glycopeptide derivatives target the bacterial cell wall, thereby perturbing its biosynthesis. To help determine the modes of action of glycopeptide antibiotics, we have developed a bottom-up mass spectrometry approach complemented by solid-state nuclear magnetic resonance (NMR) to elucidate importan

Chloropolysporins A, B and C, novel glycopeptide antibiotics from Faenia interjecta sp. nov. V. Comparative studies of the biological properties.

Chloropolysporins A, B and C, as well as derivatives prepared from this group and alpha- and beta-avoparcins by enzymatic and mild acid hydrolysis, were active against Gram-positive bacteria including clinically isolated methicillin-resistant Staphylococci (MIC 0.39-6.25 micrograms/ml) and anaerobic enterobacteria (MIC 0.10-1.56 micrograms/ml). Derhamnosyl and demannosyl derivatives from both groups of antibiotics showed stronger activities than the parent compounds. The MIC and MBC values against Staphylococci were similar and were not effected by the presence of serum. Moreover, chloropolysp

Immunohistochemical localization of human pituitary glycopeptide (HPGP)-like immunoreactivity in the hypothalamus and pituitary of normal and homozygous diabetes insipidus (Brattleboro) rats.

Sections of the hypothalami and pituitary glands of normal (Sprague-Dawley) and homozygous diabetes insipidus (Brattleboro) rats were stained with antiserum to a human pituitary glycopeptide (HPGP) by using the immunohistochemical peroxidase-antiperoxidase method at the light microscopic level. Our results show in normal rats that immunoreactive HPGP was localized in the perikarya of the magnocellular neurons of the hypothalamus, in the posterior pituitary, and in the nerve fibers distributed in the median eminence (ME) and in the areas between the supraoptic nuclei (SON), paraventricular nucl

Effective Tumor Protection by a Tumor-Associated Mucin-1 Glycopeptide-Based Conjugate Vaccine in a Spontaneous Mouse Breast Cancer Model.

Human mucin-1 (MUC1) is an attractive tumor antigen due to its high expression levels on tumor cells and its important roles in tumor development. Due to the aberrant glycosylation of MUC1 on tumor cells, tumor-associated MUC1 (tMUC1) has a shortened O-glycan attached on the peptide backbone, distinguishing it from its counterpart on normal cells. Preclinical evaluation of tMUC1-based vaccine candidates typically involved the immunization of mice that are tumor-free. However, tumor development is often associated with immune suppression. To better mimic clinical conditions encountered in cance

Clinical Phenotypes and Glycopeptide Escalation Patterns in Pediatric Febrile Neutropenia: Associations with Short-Term Outcomes and Acute Kidney Injury Risk.

OBJECTIVE: Febrile neutropenia (FN) in pediatric oncology is a clinically heterogeneous syndrome in which early antibiotic escalation decisions are frequently driven by non-specific severity cues. We aimed to identify baseline clinical phenotypes of pediatric FN, to describe early glycopeptide escalation patterns across these phenotypes, and to examine their associations with short-term outcomes and acute kidney injury (AKI). MATERIALS AND METHODS: We conducted a retrospective cohort study including 106 FN episodes experienced by 82 pediatric oncology patients initially treated with piperacil

Bacterial Cell Wall Analogue Peptides Control the Oligomeric States and Activity of the Glycopeptide Antibiotic Eremomycin: Solution NMR and Antimicrobial Studies.

For some time, glycopeptide antibiotics have been considered the last line of defense against Methicillin-resistant Staphylococcus aureus (MRSA). However, vancomycin resistance of Gram-positive bacteria is an increasingly emerging worldwide health problem. The mode of action of glycopeptide antibiotics is essentially the binding of peptidoglycan cell-wall fragments terminating in the d-Ala-d-Ala sequence to the carboxylate anion binding pocket of the antibiotic. Dimerization of these antibiotics in aqueous solution was shown to persist and even to enhance the antibacterial effect in a co-opera

Chemotranscriptomic Profiling Defines Drug-Specific Signatures of the Glycopeptide Antibiotics Dalbavancin, Vancomycin and Chlorobiphenyl-Vancomycin in a VanB-Type-Resistant Streptomycete.

Dalbavancin, vancomycin and chlorobiphenyl-vancomycin share a high degree of structural similarity and the same primary mode of drug action. All inhibit bacterial cell wall biosynthesis through complexation with intermediates in peptidoglycan biosynthesis mediated via interaction with peptidyl-d-alanyl-d-alanine (d-Ala-d-Ala) residues present at the termini of the intermediates. VanB-type glycopeptide resistance in bacteria encodes an inducible reprogramming of bacterial cell wall biosynthesis that generates precursors terminating with d-alanyl-d-lactate (d-Ala-d-Lac). This system in Streptomy

Use of ceftaroline after glycopeptide failure to eradicate meticillin-resistant Staphylococcus aureus bacteraemia with elevated vancomycin minimum inhibitory concentrations.

Elevated minimum inhibitory concentrations (MICs) of vancomycin against meticillin-resistant Staphylococcus aureus (MRSA) and the emergence of heteroresistant S. aureus strains have led to increased use of anti-MRSA antibiotics other than vancomycin. Ceftaroline fosamil is a novel cephalosporin with activity against MRSA, but there are limited clinical data on its use for MRSA bacteraemia (MRSAB) and against strains exhibiting high vancomycin MICs (2-4 μg/mL). This multicentre, retrospective, case-control study compared the microbiological and clinical effectiveness of ceftaroline used after v

Design of mineral medium for growth of Actinomadura sp. ATCC 39727, producer of the glycopeptide A40926: effects of calcium ions and nitrogen sources.

Actinomadura sp. ATCC 39727 produces the glycopeptide antibiotic A40926, structurally similar to teicoplanin, with significant activity against Neisseria gonorrhoeae and precursor of the semi-synthetic antibiotic dalbavancin. In this study the production of A40926 by Actinomadura under a variety of growth conditions was investigated. The use of chemically defined mineral media allowed us to analyze the influence of carbon and nitrogen sources, phosphate, ammonium and calcium on the growth and the antibiotic productivity of Actinomadura. We confirm recent data [Gunnarsson et al. (2003) J Ind Mi

In vitro activity of fosfomycin in combination with vancomycin or teicoplanin against Staphylococcus aureus isolated from device-associated infections unresponsive to glycopeptide therapy.

Fosfomycin is a molecule that inhibits the early stage of peptidoglycan synthesis and shows a broad-spectrum bactericidal activity against G … f 1x MIC. If these results are confirmed by in vivo experiments, the combination of fosfomycin with glycopeptides might be useful for treating device-associated infections, and in preventing the phenomenon of increasing MICs for glycopeptides.

Lycium barbarum glycopeptide confers neuroprotection in chronic ocular hypertensive rats: a study investigating visual function, retinal structure, microcirculation, and pyroptosis.

BACKGROUND: Lycium barbarum glycopeptide (LbGp) has been reported to exert neuroprotective effects, yet its role in chronic ocular hypertension (COH) and the underlying mechanisms remain unclear. This study investigated whether LbGp protects against COH-induced retinal damage by inhibiting pyroptosis. METHODS: A COH rat model was established, and the animals were randomly assigned to model, LbGp, VX-765 (pyroptosis inhibitor), and VX-765+LbGp groups. After 28 days of treatment, visual function, retinal structure, microcirculation, and pyroptosis-related protein expression were assessed. In pa

Lycium barbarum glycopeptide mitigates retinal ischemia-reperfusion injury through its anti-inflammatory, anti- senescence, and anti-apoptosis properties.

This study investigates the therapeutic potential of lycium barbarum glycopeptide (LbGp) in mitigating retinal ischemia-reperfusion injury (RIRI), a key pathological mechanism in glaucoma. RIRI involves inflammation, cellular senescence, and apoptosis, leading to retinal ganglion cell (RGC) degeneration and optic nerve damage. Using in vivo and in vitro models, the study evaluated LbGp's anti-inflammatory, anti-senescence, and anti-apoptotic properties. In vivo, RIRI was induced in C57BL/6J mice by elevating intraocular pressure, followed by reperfusion, with LbGp administered orally. In vitro

An N-glycopeptide MS/MS data analysis workflow leveraging two complementary glycoproteomic software tools for more confident identification and assignments.

Complete coverage of all N-glycosylation sites on the SARS-CoV2 spike protein would require the use of multiple proteases in addition to trypsin. Subsequent identification of the resulting glycopeptides by searching against database often introduces assignment errors due to similar mass differences between different permutations of amino acids and glycosyl residues. By manually interpreting the individual MS2 spectra, we report here the common sources of errors in assignment, especially those introduced by the use of chymotrypsin. We show that by applying a stringent threshold of acceptance, e…
