Silencing long ascending propriospinal neurons after spinal cord injury improves hindlimb stepping in the adult rat.

Long ascending propriospinal neurons (LAPNs) are a subpopulation of spinal cord interneurons that directly connect the lumbar and cervical enlargements. Previously we showed, in uninjured animals, that conditionally silencing LAPNs disrupted left-right coordination of the hindlimbs and forelimbs in a context-dependent manner, demonstrating that LAPNs secure alternation of the fore- and hindlimb pairs during overground stepping. Given the ventrolateral location of LAPN axons in the spinal cord white matter, many likely remain intact following incomplete, contusive, thoracic spinal cord injury (…

Effects of load carrying on metabolic cost and hindlimb muscle dynamics in guinea fowl (Numida meleagris).

The goal of this study was to test whether the contractile patterns of two major hindlimb extensors of guinea fowl are altered by load-carrying exercise. We hypothesized that changes in contractile pattern, specifically a decrease in muscle shortening velocity or enhanced stretch activation, would result in a reduction in locomotor energy cost relative to the load carried. We also anticipated that changes in kinematics would reflect underlying changes in muscle strain. Oxygen consumption, muscle activation intensity, and fascicle strain rate were measured over a range of speeds while animals r…

Carbon monoxide wash-in method to determine gas transfer in vascular beds: application to rat hindlimb.

The vascular barrier to gas transfer is an important physiological parameter; however, no readily applicable technique exists to quantitate the process. A simple technique to measure the permeability-surface area (PS) product for gas transfer in vascular beds is proposed using wash in of carbon monoxide (CO) and Crone-Renkin analysis. Wash-in experiments were performed on the perfused hindlimbs of male Wistar rats (n = 15) by using CO as a surrogate marker for oxygen and technetium-99m-labeled albumin as the vascular marker. The use of CO and erythrocyte-free perfusate and the collection of ou…

The arterial tree in the pig hindlimb from a more comprehensive perspective: anatomical study with translational implications.

INTRODUCTION: Although various vessel-related models using pig hindlimbs have been reported, descriptions of the arterial tree of pig hindlimbs are either rare or confusing. This cadaveric study was designed to explore and describe the anatomy of the arterial tree of pig hindlimbs and to compare with the arterial tree in human legs. MATERIALS AND METHODS: The vascular anatomy of the hindlimb of lard infused pigs (Shanghai White Pig) was investigated (n = 20). The primary parameters were as follows: the course and branches of the arterial tree of pig hindlimbs; the discrepancy in the anatomica…

Normal MRI features of the manica flexoria in horses and evaluation of the anatomic variability between forelimbs and hindlimbs.

Manica flexoria tears are increasingly recognized as a cause of lameness in horses resulting in a need for improved pre-operative diagnosis. … orsolateral/dorsomedial thickness. Eighteen MRI studies fit the inclusion criteria: 6 forelimbs, 12 hindlimbs. The manica flexoria on gross dissection was thicker proximally in forelimbs, and thinner proximally in hindlimbs where it blends with overlying fascia. On proton density-weighted images, the manica flexoria was hyperintense to the superficial digi …

Numerical evaluation of internal femur osteosynthesis based on a biomechanical model of the loading in the proximal equine hindlimb.

Femoral fractures are often considered lethal for adult horses because femur osteosynthesis is still a surgical challenge. For equine femur osteosynthesis, primary stability is essential, but the detailed physiological forces occurring in the hindlimb are largely unknown. The objective of this study was to create a numerical testing environment to evaluate equine femur osteosynthesis based on physiological conditions. The study was designed as a finite element analysis (FEA) of the femur using a musculoskeletal model of the loading situation in stance. Relevant forces were determined in the mu…

Influence of Brain Stem on Axial and Hindlimb Spinal Locomotor Rhythm Generating Circuits of the Neonatal Mouse.

The trunk plays a pivotal role in limbed locomotion. Yet, little is known about how the brain stem controls trunk activity during walking. In this study, we assessed the spatiotemporal activity patterns of axial and hindlimb motoneurons (MNs) during drug-induced fictive locomotor-like activity (LLA) in an isolated brain stem-spinal cord preparation of the neonatal mouse. We also evaluated the extent to which these activity patterns are affected by removal of brain stem. Recordings were made in the segments T7, L2, and L5 using calcium imaging from individual axial MNs in the medial motor colum…

The effect of ovariectomy combined with hindlimb unloading and reloading on the long bones of mature Sprague-Dawley rats.

OBJECTIVE: To determine the effect of loss of ovarian function and mechanical loading (ie, inactivity) alone or in combination on bone mass and strength. DESIGN: Mature (aged 6 mo) rats were ovariectomized to induce loss of ovarian function and bone. Hindlimb unloading (HLU) was used to determine the effect of mechanical unloading and reloading on bone mass and strength. Bone mass of the femur and tibia was determined using dual-energy x-ray absorptiometry. Femoral and tibial bone strength was determined by a three-point bending test and by a torsion test. RESULTS: Ovariectomy (OVX) alone de…

Deviations in gait pattern in experimental models of hindlimb paresis shown by a novel pressure mapping system.

Injuries to the central and peripheral nervous system result in varying degrees of paresis and as such alter gait. We developed novel quantitative measures to assess compensatory patterns of gait in experimental models of unilateral and bilateral hindlimb paresis. We hypothesized that hindlimb paresis results in unique alterations in the gait cycle that reflect the symmetry of the initial lesion. To test this hypothesis, adult, male Sprague-Dawley rats were subjected to a unilateral sciatic nerve crush injury or a moderately severe spinal cord contusion injury at T8. Kinematic and timing param…

Bilateral receptive field neurons in the hindlimb region of the postcentral somatosensory cortex in awake macaque monkeys.

Single-neuron activities were recorded in the hindlimb region of the primary somatosensory cortex and part of area 5 in awake Japanese monkeys. A total of 1050 units were isolated from five hemispheres of four animals. Receptive fields (RFs) and submodalities were identified for 90% of isolated neurons in areas 3a and 3b. The percentage decreased as the recoding site moved to the more caudal areas. Deep or skin submodality neurons were dominant in area 3a or area 3b, respectively. Deep submodality neurons increased in more caudal areas and were the majority in areas 2 and 5. These observations…

[Changes in surface microcirculation of extensor digitorum longus tendon after ischemia-reperfusion of rabbit hindlimbs].

The circulation was stopped by tourniquet in one rabbit hindlimb. After 2 or 5 hr of ischemia, the hindlimb was reperfused. During the first 1 hr of reperfusion, dynamic changes in surface microcirculation of extensor digitorum longus tendon were observed by using intravital microscopy. Particular attention was paid to leukocyte-endothelium adhesion and microvascular perfusion condition. It was found that 1) the number of leukocytes adherent to the venular endothelium of ischemic tissue significantly increased following reperfusion and the degree of increases appeared to be directly related to…

The postural reaction to the drop of a hindlimb support in the standing cat remains following sensorimotor cortical ablation.

The postural response to an unexpected drop of either hindlimb platform was studied in the freely standing cat. The vertical forces, the forelimb electromyographic (EMG) activities and the movement of the trunk were analysed. A stereotyped diagonal pattern of support was observed. The imposed unloading of one hindlimb was followed by unloading of the diagonally opposite forelimb and loading of the two other limbs. In the ipsilateral loaded forelimb, the force increase is preceded by an activation of the biceps and is concomitant with a triceps coactivation. In the contralateral unloaded foreli…

Deep-Learning Based Multi-Joint Synchronous Tracking for Objective Quantification of Hindlimb Locomotor Kinematics in Rats.

Accurate and objective assessment of rat gait is essential to neuroscience and kinesiology research fields. Conventional systems frequently depend on footprint imaging for indirect gait inference, which cannot fully capture hindlimb multi-joint kinematics. This study establishes a markerless treadmill-based gait analysis system for rodents, integrating custom deep learning algorithms with a programmable weight-support treadmill to enable real-time tracking of multiple lower-limb joints and multidimensional kinematic quantification. The system automatically extracts gait cycle parameters, joint…

Bilateral Muscle Architecture Differences in a Rabbit Model of Unilateral Hindlimb Transtibial Amputation: A Pilot Study.

Residual muscles after amputation experience mechanical unloading, loss of distal tendon insertion, and reduced excursion, conditions known to cause muscle degeneration. However, the architecture of residual muscles following amputation remains largely unreported. In this pilot, exploratory study, five healthy, skeletally mature, male New Zealand white rabbits underwent unilateral hind-paw ankle disarticulation. The contralateral intact limb served as the within-subject control. Six hindlimb muscles that cross the intact biological ankle were harvested bilaterally at four weeks post-amputation…

Biomechanical simulations of hindlimb function in Alligator provide insights into postural shifts and body size evolution.

The shift from sprawling to erect limb postures in archosaurs during the Triassic represents a major evolutionary transformation in vertebrates. One unresolved question regarding the limb posture transition is its association with body size evolution. If adopting more erect limb postures reduces mass-specific muscle forces and bone stresses, it would enable the evolution of larger body sizes. We tested this prediction using computational modeling and simulation of hindlimb muscle activations and femoral stresses across limb postures and body sizes in juvenile to adult American alligators and a…

Rat Hindlimb amputation model shows analgesia and sexually dimorphic cold hypersensitivity with immediate targeted muscle reinnervation.

Targeted muscle reinnervation (TMR) has been used clinically to reduce pain in amputees, but its underlying analgesic mechanisms remain uncl … amputation, with cold hypersensitivity responses differing by sex. This supports the utility of the hindlimb iTMR model and highlights the need for continued mechanistic investigation and sex-specific analyses in future studies. PERSPECTIVE: Using a rat hindlimb amputation model to evaluate iTMR, we demonstrate its potential to mitigate neuropathic pain an …

Chrysin promotes angiogenesis in rat hindlimb ischemia: Impact on PI3K/Akt/mTOR signaling pathway and autophagy.

Limb ischemia occurs due to obstruction of blood perfusion to lower limbs, a manifestation that is associated with peripheral artery disease (PAD). Angiogenesis is important for adequate oxygen delivery. The present study investigated a potential role for chrysin, a naturally occurring flavonoid, in promoting angiogenesis in hindlimb ischemia (HLI) rat model. Rats were allocated into four groups: (1) sham-operated control, (2) HLI: subjected to unilateral femoral artery ligation, (3) HLI + chrysin: received 100 mg/kg, i.p. chrysin immediately after HLI, and (4) HLI + chrysin + rapamycin: recei…
